调脂通脉中药对动脉粥样硬化兔模型炎症因子表达的影响

Influences of Chinese medicinal with actions of regulating blood lipid and unblocking vessels on expressions of inflammatory cytokines in rabbits with atherosclerosis

摘要: 目的 探讨调脂通脉中药对动脉粥样硬化兔模型炎症因子的调控机制。方法 采用高脂饲料饮食和球囊拉伤的方法制备动脉粥样硬化兔模型,随机分为正常组、模型组、假手术组、阿托伐他汀(2.2 mg/kg)组和调脂通脉(8 g/kg)组共5组,选择治疗后第45、90天为观察点,采用酶联免疫吸附测定(enzyme-linked immunosorbent assay ELISA)方法检测血清C-反应蛋白(CRP)、转化生长因子-β(TGF-β)、白细胞介素6、10、17(IL-6, 10, 17)水平。结果 不同时间点,模型组家兔血清CRP、IL-6、IL-17较假手术组明显升高(P<0.01),血清TGF-β、IL-10较假手术组明显下降(P<0.01)。调脂通脉中药组家兔血清IL-6、CRP、IL-17水平较模型组明显下降(P<0.01),血清TGF-β、IL-10水平明显升高(P<0-01),与阿托伐他汀组无明显差异。结论 动脉粥样硬化兔模型造模成功后,通过调脂通脉中药的干预,动脉粥样硬化明显改善。调脂通脉中药减少血清IL-6 、CRP、IL-17的水平,修复受损血管内皮细胞结构和功能,减少动脉壁内膜脂质沉积,可能是其作用机制。

Abstract: Objective To investigate the mechanism of Chinese medicinal with actions of regulating blood lipid and unblocking vessels in regulating inflammatory cytokines in rabbits with atherosclerosis. Methods The rabbit model of atherosclerosis was established by using high fat diet and balloon injury method. The rabbits were randomly divided into normal group, model group, sham-operation group, atorvastatin group and Chinese medicinal group. The observations were carried out after treatment for 45 d and 90 d. The levels of serum C-reactive protein (CRP), transforming growth factor-β (TGF-β), interleukin-6 (IL-6), IL-10 and IL-17 were detected by applying enzyme-linked immunosorbent assay (ELISA). Results At different time points, the levels of serum CRP, IL-6 and IL-17 increased significantly (P<0.01) and TGF-β and IL-10 decreased significantly (P<0.01) in model group compared with sham-operation group. In Chinese medicinal group, the levels of serum IL-6, CRP and IL-17 decreased significantly (P<0.01) and TGF-β and IL-10 increased significantly (P<0.01) compared with model group, but had no difference compared with atorvastatin group. Conclusion The intervention of Chinese medicinal with actions of regulating blood lipid and unblocking vessels can significantly relieve atherosclerosis. The mechanism may be related to that the Chinese medicinal can reduce the levels of IL-6, CRP and IL-17, recover the structure and function of damaged endothelial cells, and abate the intimal lipid deposition in arterial wall.
