葛根素注射液对2型糖尿病动物模型 KKAy小鼠肾损伤病理改变的影响

Influences of Puerarin Injection on pathological changes of renal injury in KKAy mice with type 2 diabetes

摘要: 目的 研究葛根素对糖尿病肾病的治疗作用机制。方法 将12只雄性 KKAy小鼠饲养至 14 周龄时随机分成模型组和葛根素治疗组 (腹腔注射1.3 mL/kg)。8只同龄雄性 C57BL/6J 小鼠为正常对照组。分别于 20、24周龄时检测各组小鼠血糖、血清肌酐、血尿素,于24周龄处死小鼠,取肾脏,光镜、电镜下观察各组小鼠不同周龄时的病理变化情况。结果 血清生化指标检测可见治疗组血糖、血肌酐、血尿素水平较模型组显著降低(P<0.05或P<0.01),治疗组血糖水平仍明显高于正常组(P<0.01),但治疗组血肌酐和血尿素水平与正常组无统计学差异。形态学观察可见模型组肾小球系膜细胞及系膜基质增多,肾小管上皮细胞胞浆出现明显空泡,管腔可见蛋白管型,而经葛根素注射液治疗后,治疗组病变较轻。结论 葛根素注射液能改善 2 型糖尿病KKAy小鼠肾功能,减轻肾脏病理损害,具有肾脏保护作用。

Abstract: Objective To study the effective mechanism of puerarin to diabetic nephropathy (DN). Methods Male KKAy mice (n=12, 14 weeks old) were randomly divided into model group and puerarin treatment group (treatment group, 1.3 mL/kg/d), and other 8 male C57BL/6J mice with the same age were included into normal group. On the 20th w and 24th w, the levels of plasma glucose, serum creatinine (Cr) and blood urea (BU) were detected in mice of all groups. On the 24th w, the mice were executed to collect kidney sample for observing renal pathological changes with light microscope and electron microscope. Results The levels of plasma glucose, serum Cr and BU decreased significantly in treatment group than those in model group (P<0.05 or P<0.01), the level of plasma glucose was significantly higher in treatment group than that in normal group (P<0.01), but levels of Cr and BU had no statistical difference. The morphological observation showed increased renal tubular mesangial cells and mesangial matrix, obvious vacuoles in cytoplasm of tubular epithelial cells and protein casts in renal tubules in model group. After the treatment with Puerarin Injection, all renal pathological changes were alleviated in treatment group. Conclusion Puerarin Injection can improve kidney function, abate renal pathological lesion and protect kidney in KKAy mice with type 2 diabetes.
