三七总皂苷对归巢到脑梗死边缘区CD105骨髓间充质干细胞增殖分化的实验研究

Panax notoginseng saponins promote proliferation and differentiation of CD105+-BMSC homing to cerebral infarction marginal zone

摘要: 目的 探讨三七总皂苷对归巢到梗死边缘区CD105+骨髓间充质干细胞(BMSCs)增殖分化的影响。方法 参照改良Zea-Longa法建立大鼠大脑中动脉脑缺血再灌注损伤模型(MCAO)。造模成功大鼠随机分为模型组、三七总皂苷高、中、低剂量组。各组再随机按1、3、7、14、28 d时间点分为亚组。三七总皂苷各组分别以浓度20、40、60 g/L血塞通灌胃,每天10 mL/kg。正常组和模型组以等量生理盐水灌胃,1次/d,共28 d。免疫组化法检测不同时间点归巢BMSCs在梗死边沿区CD105+-BMSC、微管相关蛋白-2(MAP-2)、胶质纤维酸性蛋白(GFAP)、IIIβ-Tubulin型微管蛋白的增殖分化情况。结果 模型组、三七总皂苷各剂量组各时间点梗死周边CD105+-BMSC、MAP-2、GFAP、IIIβ-Tubulin阳性细胞数比正常组明显增多(P<0.05)。模型组、三七总皂苷各剂量组CD105+-BMSC、MAP-2、GFAP、IIIβ-Tubulin阳性细胞数从第1天开始升高,第7天到高峰,其后逐渐降低。但三七总皂苷中、高剂量组各时间点CD105+-BMSC、MAP-2、GFAP、IIIβ-Tubulin阳性细胞数比模型组增多(P<0.05)。结论 三七总皂苷可以通过促进归巢到梗死边沿区BMSCs增殖分化再生而修复损伤脑组织。

Abstract: Objective To investigate the promotion effect of panax notoginseng saponins (PNS) on CD105+-bone mesenchymal stem cells (BMSC) homing to the cerebral infarction marginal zone. Methods The rat model of MCAO was established by referencing Zea-Longa method. The modeled rats were randomly divided into model group and high-dose ,mid-dose and low-dose PNS group(high-dose group, mid-dose group and low-dose group), and then according to time points divided into 1 d,3 d,7 d,14 d and 28 d subgroups. High-dose group, mid-dose group and low-dose group were treated with Xuesaitong Capsules (60, 40 and 20 g/L once a day), and normal group and model group were orally administered saline water once a day for 28 d. The positive cells of CD105+-BMSC, MAP-2, GFAP and IIIβ-Tubulin were detected by immunohistochemical method at different time points. Results The positive cells of CD105+-BMSC, MAP-2, GFAP and IIIβ-Tubulin increased in model group and PNS groups on the first day, reached the peak on the 7th day, and then went down gradually. The positive cells of CD105+-BMSC, Map-2, GFAP and IIIβ-Tubulin in model group and all PNS groups compared with model group increased significantly (P<0.05). Conclusion PNS can repair the damaged cerebral tissue induced by infarction by promoting the proliferation and differentiation of BMSCs homing to infarction marginal zone.
