丹栀逍遥散对广泛性焦虑大鼠Papez环路内   细胞凋亡的影响*

Effects of Danzhi Xiaoyao San on cell apoptosis of Papez’s circuit in rat model of Generalized Anxiety Disease (GAD)*

摘要: 目的 研究丹栀逍遥散对广泛性焦虑(GAD)大鼠脑Papez环路区域细胞凋亡的影响。方法 将雄性Wistar大鼠随机分为正常组、模型组、中药治疗组(中药组)和西药对照组(西药组)4组,每组10只大鼠。模型组、中药组、西药组采用不确定空瓶饮水刺激方法造成广泛性焦虑大鼠模型。中药组和西药组分别以丹栀逍遥散(10.5 g/kg)和盐酸丁螺环酮(2 mg/kg)水溶液灌胃,每天1次,连续7 d。模型组蒸馏水灌胃7 d。取脑组织,光镜、电镜观察海马、杏仁核的形态学变化,流式细胞术法检测海马和乳头体组织的早期细胞凋亡率。结果 光电镜观察结果显示,模型组的海马、杏仁核区域出现明显的染色质边集,核固缩等细胞凋亡的形态学变化,中药组和西药组能够有效的改善这些变化。流式细胞术结果显示,模型组的海马及乳头体组织的早期细胞凋亡率较正常组有明显的升高(P<0.01),中药组和西药组与模型组相比有较明显的降低(P<0.05或P<0.01)。结论 GAD大鼠脑内Papez环路区域存在着广泛且明显的细胞凋亡,丹栀逍遥散能在一定程度上干预和防止细胞凋亡,保护神经组织。

Abstract: Objective To study the effects of Danzhi Xiaoyao San on cell apoptosis of Papez’s circuit in rat model of Generalized Anxiety Disease (GAD). Methods Male Wistar rats were divided into the normal group, model group, Chinese Medicine group, and control group of western medicine; each group contained 10 rats. The GAD model was established by using randomly assigned empty drinking bottle to upset the rats. Then the Chinese medicine group was intragastrically given Danzhi Xiaoyan San, while the western medicine group was given buspirone hydrochloride, and the model group was given distilled water once a day for a week. Then morphological changes of hippocampus and amygdaloid nucleus were observed by using light and electron microscopes, and the early apoptosis rate of hippocampus and mamillary body were evaluated by applying flow cytometry assays after removing rat brain. Results Cells in hippocampus and amygdaloid nucleus of model group showed typical morphological changes of apoptosis such as margination and karyopyknosis. Danzhi Xiaoyao San and buspirone hydrochloride could revert the changes markedly. Flow cytometry assay showed that the early apoptosis rates in hippocampus and mamillary body of model group increased significantly compared with normal group (P<0.01). Compared with model group, the apoptosis rates of Chinese medicine group and Western medicine group decreased (P<0.05 or P<0.01). Conclusion There is extensive and obvious apoptosis in Papez’s circuit of GAD rat’s brain. Danzhi Xiaoyao San can prevent and inhibit apoptosis to some extent.
