三黄泻心汤组分配伍方的图谱表征及活血化瘀优势方的成分分析*

Fingerprints for characterization of nine compatibility formula and optimization of compatibility formula engaged in activating blood and resolving stasis :a study for Sanhuang Xiexin decoction composed of Radix et Rhizoma rhei, Radix scutellariae and Rhizoma coptidis*

摘要: 目的 对9个三黄泻心汤成分配伍方进行特征图谱表征,并对活血化瘀作用优势方进行成分分析。方法 采用高效液相色谱法,Agilent C18柱,乙腈-1%甲酸梯度洗脱,检测波长280 nm,柱温30 ℃,对三黄泻心汤成分配伍方进行特征图谱分析,并对活血化瘀优势方进行配伍成分的含量测定。结果 9个三黄泻心汤组分配伍方特征图谱中均包含24个特征峰,以化学对照品为对照归属了其中的9个色谱峰。并对活血化瘀优势组分配伍方(组方7)进行了成分分析,该方中黄芩苷、蒽醌苷、蒽醌苷元及盐酸小檗碱的含量比例约为2.8∶1.7∶1.6∶1.0。结论 所建立的三黄泻心汤组分配伍方特征图谱及含量测定方法稳定可靠,可用于组分配伍方的质量控制。同时,基于三黄泻心汤的活血化瘀组分配伍优势方的成分分析也为进一步揭示组分配伍与其功效的相关性奠定了基础。

Abstract: Objective To characterize the distribution of fingerprints of nine different compatibility formula (Formula 1~9) of Radix et Rhizoma rhei, Radix scutellariae and Rhizoma coptidis, three Chinese herbal medicines that comprise Sanhuang Xiexin decoction; to analysis the components of each formula, from which best effects of activating blood and resolving stasis were obtained. Methods High performance liquid chromatography (HPLC) was performed with Agilent C18 column, acetonitrile and 1% formic acid gradient elution, detection wavelength at 280 nm, column temperature of 30℃. Results 24 characteristic peaks were included in each fingerprint of compatibility formulae, and 9 peaks were identified with chemical reference. Formula 7, whose proportion of baicalin, anthraquinone glycosides, anthraquinones aglycone and berberine hydrochloride was 2.8∶1.7∶1.6∶1.0 approximately, was determined to be optimal to activating blood and resolving stasis. Conclusion The methods established of fingerprints and component analysis are stable and reliable to be used for quality control of component compatibility formulae. Furthermore, components analysis of Formula 7, the optimal formula for activating blood and resolving stasis, is expected to explore the correlation between compatibility and effect.
