左归降糖解郁方对糖尿病并发抑郁症大鼠海马区星形胶质细胞谷氨酸转运体的影响*

Effect of Zuogui Jiangtang Jieyu Formula on amino acid transporter expression in hippocampal astrocytes of diabetes mellitus rats with depression*

摘要: 目的 研究左归降糖解郁方对糖尿病并发抑郁症大鼠星形胶质细胞兴奋性氨基转运体3(EAAT 3)、囊泡谷氨酸转运体2(VGLUT 2)表达的影响。方法 采用高脂乳剂灌胃、尾静脉注射链脲佐菌素、28 天慢性应激联合的方法复制糖尿病并发抑郁症大鼠模型,SD大鼠随机分为5组:糖尿病并发抑郁症组,二甲双胍+氟西汀组,左归降糖解郁方高、中、低剂量组,以正常大鼠为空白对照组,灌胃给药 4 周。药物干预后,采用 Morris水迷宫检测大鼠空间学习能力,免疫组化双标法检测星形胶质细胞中EAAT 3、VGLUT 2的表达情况。结果 与空白对照组比较,糖尿病并发抑郁症组大鼠逃避潜伏期延长(P<0.01),EAAT 3表达下降,VGLUT 2表达升高,差异有统计学意义(P<0.05);与糖尿病并发抑郁症组比较,二甲双胍+氟西汀组和左归降糖解郁方高剂量组大鼠逃避潜伏期缩短,差异有显著统计学意义(P<0.01),且二甲双胍+氟西汀组和左归降糖解郁方高、中、低剂量组EAAT 3表达升高,二甲双胍+氟西汀组和左归降糖解郁方高剂量组VGLUT 2表达下降,差异有统计学意义(P<0.05)。 结论 左归降糖解郁方可以改善糖尿病并发抑郁症大鼠海马区星形胶质细胞EAAT 3、VGLUT 2表达的紊乱状态。

Abstract: Objective To investigate the effects of Zuogui Jiangtang Jieyu Formulation (ZGJTJYF) on the expression of excitatory amino acid transporter 3(EAAT 3) and vesicular glutamate transporter 2(VGLUT 2) in hippocampal astrocytes of diabetes mellitus rats with depression (DD). Methods DD rats were induced by intragastrical administration of high-fat diet, tail intravenous injection of streptozotocin (STZ) with 28 days’ chronic stress and were randomly divided into five groups—model group, positive drug group (metformin with fluoxetine), high-, middle-, and low- dose of ZGJTJYF (high-dose, mid-dose, and low-dose) group. Healthy SD rats were assigned into blank control group. After four consecutive weeks the cognition ability was measured using Morris water maze, and the expression of EAAT3 and VGLUT2 were detected by using immunohistochemical double labeling method. Results Compared with blank control group, escape latency of model group was significantly longer (P<0.01), EAAT 3 expression decreased, VGLUT 2 expression increased remarkably (both P<0.05). Compared with model group, escape latency of positive drug group and in high-dose group decreased significantly(P<0.01),expression of EAAT3 in positive drug group and all ZGJTJYF groups increased(P<0.05), and VGLUT2 expression in positive drug group and high-dose group decreased significantly (P<0.05). Conclusion Zuogui Jiangtang Jieyu Formula can improve the disorder of EAAT3 expression and VGLUT2 expression in hippocampal astrocytes of diabetes mellitus rats with depression.
