响应面法优化蒙药肋柱花总黄酮超声提取工艺*

Optimization of ultrasonic-assisted extraction of total flavonoids from Lomatogonium rotatum by response surface analysis*

摘要: 目的 采用单因素试验和响应面法优化肋柱花总黄酮的超声波辅助提取工艺,为肋柱花中黄酮类化合物的开发利用提供理论依据。方法 以乙醇溶液为提取溶剂,在单因素试验的基础上,以乙醇体积分数(X1)、液料比(X2)、超声时间(X3)为自变量,以总黄酮含量(Y)为响应值进行Box-Behnken中心组合试验设计,建立肋柱花总黄酮含量的二次回归方程,得到优化组合条件。结果 响应面法分析结果表明,肋柱花总黄酮最佳提取工艺条件为65%乙醇,液料比20 mL/g,提取时间为20 min,在此条件下总黄酮含量为21.26 mg/g,试验结果与模型预测值相符。二项式拟合复相关系数r=0.956 8。结论 应用响应面方法优选的肋柱花总黄酮提取工艺稳定、合理、可行。

Abstract: Objective To optimize ultrasonic-assisted extraction process of total flavonoids from Lomatogonium rotatum by using single factor experiment and response surface methodology, so as to provide the theoretical evidence for the exploitable and utilization of flavonoids in Lomatogonium rotatum.Methods Ethanol solution was selected as the extraction solvent. Based on the single factor experiment,three independent variables of ethanol volume fraction(X1),material to liquid ratio(X2) and sonication time(X3)with the response value of the content of total flavonoids(Y) were designed for Box-Behnken central composite experiment, then a quadric regression equation was established to predict the yield of total flavonoids.Results The optimum extraction process was 65% ethanol solution(V/V)with liquid to material ratio of 20 mL/g for 20 minutes.The actual yield of total flavonoids were 21.26 mg/g,very close to the predictive values.The binomial fitting multiple correlation coefficient (r) is 0.956 8.Conclusion Optimizing the extraction process of total flavonoids of Lomatogonium rotatum by response surface methodology was proved to be stable,reasonable and feasible.
