ERα基因TA重复序列多态性与月经过少中医证候相关性研究

Correlation of TA repeat polymorphisms of estrogen receptor-α gene and TCM patterns of hypomenorrhea*

摘要: 目的 探讨雌激素受体α(ERa)基因TA(胸腺嘧啶thymine,腺嘌吟adenine)重复序列多态性在不明原因月经过少人群中的分布情况及其与中医证候的相关性。方法 采用测序方法检测TA重复序列的重复次数,比较月经过少组及正常人群的TA重复序列多态性的分布差异及其在不同中医证候间的分布差异。结果 ①TA重复序列与原因不明月经过少存在相关性,实验组TA 19等位基因频率(22.73%)高于对照组(4.55%),差异有显著性(P=0.007);②TA 19等位基因型针对月经过少不同证候的构成比:肾虚型<血瘀型<血虚型,经卡方检验无统计学差异(χ2=5.137, P=0.077),肾虚型与血虚型经卡方检验有统计学意义(χ2=4.574,P=0.032,OR值=0.283)。结论 ERa基因多态性TA重复序列与不明原因月经过少存在相关性,TA 19等位基因可能是其易感因素,但其与中医证候的相关性无统计学意义。

Abstract: Objective To find out the distribution of TA (thymine-adenine) repeat polymorphisms of estrogen receptor-α (ERα) gene in patients with unexplained hypomenorrhea as well as its correlation with TCM patterns of hypomenorrhea. Methods The present case-control study was carried out by using the sequencing methods to find out the TA repeat polymorphisms distribution differences in patients and controls, and in TCM patterns of patients. Results The frequency of TA19 allele in experimental group was 22.73%, significantly higher than 4.55% in control group (P=0.007) . The rank of distribution of TA19 allele in patterns listed as kidney deficiency, blood stasis and blood deficiency, showed no significant difference (X2=5.137, P=0.077). But the distribution of TA19 in pattern of kidney deficiency and blood deficiency showed significant difference(X2=4.574,P=0.032,OR=0.283). Conclusion The correlation of TA repeat polymorphisms of estrogen receptor-α gene and TCM patterns of unexplained hypomenorrhea was proved. The TA19 allele was more susceptibility to hypomenorrhea, whose association with the different TCM patterns showed no statistical significance.
