丹参配方颗粒及丹参超细粉抗脑缺血再灌注损伤作用的比较研究*

Protective effects of concentrate granule and ultrafine powder of danshen on cerebral ischemia-reperfusion injury: a comparative study*

摘要: 目的 比较丹参配方颗粒及丹参超细粉对脑缺血再灌注损伤的保护作用。方法 雄性ICR小鼠随机分为假手术组、脑缺血再灌注组、丹参配方颗粒[0.45 g/(kg·d)]组及丹参超细粉[2.25 g/(kg·d)]组,采用大脑中动脉栓塞法(MCAO)制备模型。监测实验周期内小鼠体重,比较药物对小鼠健康状况的影响;神经功能评分实验、角落转向实验、前肢力量测定实验,比较药物对神经缺损、运动、感觉功能的恢复作用;脑组织含水量测定观察脑水肿程度,2,3,5-三苯基氯化四氮唑(TTC)染色计算梗死体积,ELISA法测定脑组织中肿瘤坏死因子(TNF-α)、白细胞介素-1β(IL-1β)、髓过氧化物酶(MPO)含量,比较药物对脑缺血再灌注损伤的影响。结果 与脑缺血再灌注组比较,丹参超细粉组小鼠前肢力量改善明显(P<0.01),体重恢复明显(P<0.05),TNF-α、MPO含量明显降低(P<0.01或P<0.05),差异有统计学意义;丹参配方颗粒组小鼠脑组织含水量明显下降(P<0.05),差异有统计学意义;丹参配方颗粒组和丹参超细粉组小鼠脑梗死体积明显缩小(P<0.01),差异有统计学意义。结论 丹参配方颗粒及丹参超细粉对脑缺血损伤均具有一定保护作用,等效剂量比较作用强度相近。

Abstract: Objective To compare the protective effects of concentrate granule and ultrafine powder of danshen (Salvia Miltiorrhizae Radix et Rhizome) on cerebral ischemia-reperfusion (CIR) injury. Methods Male ICR mice were randomly divided into sham operation group, model group, 0.45 g/(kg·d) danshen concentrate granule group and 2.25 g/(kg·d) danshen ultrafine powder group. CIR model was established by using middle cerebral artery occlusion (MCAO). Weight of mice was monitored throughout the experiment to evaluate their health status. The influence on neurological injury, movement and sensory function was evaluated by using neurologic score test, corner test and grip strength test. The effects of ultrafine powder and concentrate granule on CIR injury were compared using the following parameters: content of water in brain, infraction volume with TTC staining, and tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β), and myeloperoxidase (MPO) in the brain by using ELISA. Results Compared with CIR model group, the grip strength and body weight improved significantly (P<0.01 or P<0.05) and contents of TNF-α and MPO decreased markedly (P<0.01 or P<0.05) in ultrafine powder group; content of water in brain decreased significantly (P<0.05) in concentrate granule group; cerebral infarction volume decreased markedly (P<0.01) in both danshen groups (P<0.01). Conclusion The danshen concentrate granule and ultrafine powder both have a protective effect on ischemia-reperfusion injury, and their efficacy is similar at the same dosage.
