济阴颗粒对围绝经期综合征大鼠卵巢Caspase依赖性细胞凋亡通路的影响*

Influence of Jiyin Capsule on apoptosis pathway of ovarian caspase dependent cells in rats with peri-menopausal syndrome*

摘要: 目的 探讨济阴颗粒干预围绝经期综合征的作用机制。方法 雌性SPF级SD大鼠105只,随机选择10只正常大鼠为空白对照组,其余95只采用每日1次腹腔注射去氧乙烯基环己烯(VCD)溶液的方法造模,连续60 d。动情周期紊乱,血清雌二醇(E2)含量低于空白对照组最低值,卵泡雌激素(FSH)高于空白对照组最高值为造模成功。将造模成功大鼠随机分为6组,每组10只,分别为模型组、补佳乐组、坤宝丸组和济阴颗粒低、中、高剂量组。分组后,空白对照组与模型组给予蒸馏水,其他各组给予相应剂量药物,每日1次,连续30 d。采用荧光RT-PCR法检测卵巢组织Caspase-3,8,9 mRNA,Apaf-1 mRNA的表达。结果 与空白对照组比较,模型组大鼠卵巢细胞中Caspase-3,8,9 mRNA,Apaf-1 mRNA的表达明显升高(P<0.01);与模型组比较,济阴颗粒可显著降低卵巢细胞中Caspase-3,8,9 mRNA,Apaf-1 mRNA的表达。结论 济阴颗粒可通过下调Caspase依赖性细胞凋亡通路的传导;抑制卵巢细胞凋亡,可能是其干预围绝经期综合征的作用机制之一。

Abstract: Objective To discuss the effective mechanism of Jiyin (Nourishing Yin) Capsule in intervening peri-menopausal syndrome. Methods Normal rats (n=10) were randomly chosen into blank group. The model of peri-menopausal syndrome was established with intraperitoneal injection of VCD once a day for 60 d. When estrous cycle was disorder, level of serum estradiol (E2) was lower than the lowest level of that in blank group and level of follicle-stimulating hormone (FSH) was higher than the highest level of that in blank group, the modeling was success. The modeled rats were randomly divided into 6 groups: model group, progynova group (PGV group), Kunbao (Female Treasure) Pill group (KBP group), and low-dose, mid-dose and high-dose Jiyin Capsule groups (each n=10). After grouping, blank group and model group were given distilled water, and other groups were given corresponding medicinals once a day for 30 d. The mRNA expressions of caspase-3, caspase-8, caspase-9 and Apaf-1 were detected by using real-time fluorescence quantitative polymerase chain reaction (RT-PCR). Results Compared with blank group, mRNA expressions of caspase-3, caspase-8, caspase-9 and Apaf-1 increased significantly in model group (P<0.01). Compared with model group, mRNA expressions of caspase-3, caspase-8, caspase-9 and Apaf-1 decreased significantly in Jiyin Capsule groups. Conclusion Jiyin Capsule can inhibit cell apoptosis of ovarian through down-regulating conduction of apoptosis pathway of caspase dependent cells, which maybe one of its effective mechanisms of intervening peri-menopausal syndrome.
