柴胡达原饮对大肠杆菌性腹膜炎大鼠血清及腹腔灌洗液TNF-α、IL-10、PGE2的影响*

Effects of Chaihu Dayuan decoction on the TNF-α, IL-10 and PGE2 levels in the serum and peritoneal lavage fluid of rats with Escherichia coli peritonitis*

摘要: 目的 基于膜原理论,探讨柴胡达原饮对大肠杆菌性腹膜炎大鼠血清及腹腔灌洗液促炎及抗炎因子的影响,从膜原的角度论证柴胡达原饮抗腹膜炎症的有效性。方法 54只SD大鼠随机分为正常组、模型组、柴胡达原饮组,每组18只,实验周期为10天。模型组、柴胡达原饮组按大鼠体重20 mL/kg予以大肠杆菌配置液腹腔注射建立大鼠腹膜炎模型,柴胡达原饮组给与柴胡达原饮治疗,正常组、模型组无治疗。每天测量记录大鼠体温、体重,分别于干预后第1、3、10天取材,酶联免疫法检测各组血清及腹腔灌洗液中肿瘤坏死因子-α(TNF-α)、白细胞介素-10(IL-10)、前列腺素E2(PGE2)的水平。结果 与模型组相比,柴胡达原饮组给药后1~3 h体温明显降低(P<0.05),腹腔粘连明显改善。炎症因子方面,与模型组相比,柴胡达原饮组第1、3天血清TNF-α浓度明显降低(P<0.05);柴胡达原饮组第1、3、10天血清IL-10浓度明显升高(P<0.05);柴胡达原饮组第10天血清PGE2浓度明显升高(P<0.05)。与模型组相比,柴胡达原饮组第1、3、10天腹腔灌洗液中TNF-α浓度显著升高(P<0.01);柴胡达原饮组第3、10天腹腔灌洗液中IL-10浓度明显升高(P<0.05);柴胡达原饮组第10天腹腔灌洗液中PGE2浓度明显升高(P<0.05)。结论 柴胡达原饮能够升高腹腔内促炎因子TNF-α水平,降低血清TNF-α水平,同时能够升高血清及腹腔灌洗液中抗炎因子IL-10、PGE2水平,调控促炎及抗炎因子之间的平衡。

Abstract: Objective To investigate the effect of Chaihu Dayuan decoction (Bupleurum Moyuan-Targeting Decoction, CHDYY) on the proinflammatory and anti-inflammatory factors in the serum and peritoneal lavage fluid of rats with Escherichia coli (E. coli) peritonitis from the perspective of membrane source (mo yuan, pleurodiaphragmatic interspace) theory. Methods 54 SD rats were randomly divided into normal group, model group and CHDYY group (n=18 with the duration of intervention for 10 days in each group). Peritonitis model was established in the model group and the CHDYY group with intraperitoneal injection of E.coli solution at 20 mL/kg. The norrnal group and the model group received no treatment while the CHDYY group was orally administered with concentrated CHDYY decoction. The body temperature and body weight of the rats were recorded every day, and the samples were collected on the 1st day, 3rd day and 10th day respectively after the intervention. Levels of tumor necrosis factor alpha (TNF-α), interleukin 10 (IL-10) and prostaglandin E2 (PGE2) in serum and peritoneal lavage fluid were measured with ELISA assay. Results Compared with the model group, the body temperature of the CHDYY decoction group decreased significantly at 1~3 hour after intervention (P<0.05), and abdominal adhesion improved significantly. On the 1st and 3rd days of the CHDYY group, the serum TNF-α concentration was significantly decreased (P<0.05), and the serum IL-10 concentration in the CHDYY group was significantly increased (P<0.05) on the 1st day, 3rd day and 10thday in the CHDYY group, and the serum PGE2 concentration in the CHDYY group was significantly increased (P<0.05). The concentration of TNF-α in the peritoneal lavage fluid on the 1st, 3rd and 10th day of the CHDYY group increased significantly (P<0.01), and the concentration of IL-10 was significantly increased (P<0.05) on the 3rd day and 10th day of the CHDYY group (P<0.05). The concentration of PGE2 in the peritoneal lavage fluid of the CHDYY group was significantly increased (P<0.05) on the 10th day. Conclusion Chaihu Dayuan decoction seems to increase the level of TNF-αin the abdominal cavity, reduce the level of serum TNF-α, increase the level of anti-inflammatory factors IL-10 and PGE2 in the serum and peritoneal lavage fluid, and restore the balance between proinflammatory and anti-inflammatory factors.
