霍山石斛对肾阴虚小鼠抗疲劳及能量代谢的影响*

Effects of Dendrobium huoshanense on fatigue and energy metabolism in mice with kidney-yin deficiency*

摘要: 目的 研究霍山石斛对肾阴虚小鼠抗疲劳及能量代谢的影响。方法 采用灌服甲状腺素建立小鼠肾阴虚模型,观察霍山石斛对肾阴虚小鼠阴虚状态的影响。检测各组小鼠温度趋向性和负重游泳时间的变化,酶联免疫吸附法测定小鼠血清三碘甲状腺原氨酸(T3),甲状腺素(T4)。放射免疫分析法检测血清乳酸脱氢酶(LDH),肌酸激酶(CK),环磷酸腺苷(cAMP),环磷酸鸟苷(cGMP),计算cAMP/cGMP比值。比色法检测肝组织Na+-K+-ATP酶和Ca2+-Mg2+-ATP酶的含量。结果 与正常组相比,模型组小鼠热区停留比例和负重游泳时间明显下降(P<0.01),T3、T4、cAMP和cAMP/cGMP的含量明显升高(P<0.01、P<0.001),LDH显著降低(P<0.05);CK、Na+-K+-ATP酶和Ca2+ Mg2+-ATP酶显著增高(P<0.001)。与模型组相比,霍山石斛A、B、C、D 4个剂量组和铁皮石斛组、六味地黄丸组小鼠的热区停留比例和负重游泳时间明显增高(P<0.01、 P<0.05、P<0.001),cAMP/cGMP明显降低(P<0.05、P<0.01),Na+-K+-ATP酶和Ca2+-Mg2+-ATP酶明显降低(P<0.01、P<0.001),霍山石斛B、C、D剂量组和铁皮石斛组、六味地黄丸组T3、T4显著降低(P<0.05、P<001、P<0.001),霍山石斛B、C剂量组和铁皮石斛组、六味地黄丸组的LDH显著增高,CK显著降低(P<0.01、P<0.05、P<0.001)。结论 霍山石斛具有滋阴清热的作用,能降低甲状腺素诱导的肾阴虚小鼠的代谢酶活性和甲状腺素水平,调节能量代谢速率,提高阴虚小鼠的负重游泳时间,具有抗疲劳作用,其机制与调节能量代谢酶作用相关,进一步验证了霍山石斛药性偏寒。

Abstract: Objective To explore the effects of Huoshan Shihu (Dendrobium huoshanense, HSSH) on fatigue and energy metabolism regulation in mice with kidney-yin deficiency.Methods Mice model with kidney-yin deficiency was established by gavage of thyroxine. Effects of Huoshan Shihu on mice with kidney-yin deficiency were observed. The thermotropism behavior and the weight-loaded swimming time of mice were monitored. The levels of T3 and T4 were detected by using Enzyme-linked Immunosorbent Assay (ELISA); cAMP, cGMP, LDH and CK in serum were detected with radioimmunoassay, cAMP/cGMP was thns calculated. and the Na+-K+-ATP-ase and Ca2+-Mg2+-ATP-ase in liver tissue of mice were detected with chromatometry. Results Compared with the normal animals, the time duration in high-temperature zone and the loaded-swimming time of mice in model group were significantly decreased (P<0.01). The levels of T3, T4, cAMP, and cAMP/cGMP were significantly increased(P<0.01,P<0.001). The activities of LDH were decreased(P<0.05), and CK, Na+-K+-ATP-ase and Ca2+-Mg2+-ATP-ase were increased (P<0.001). Compared with the model animals, time duration in high-temperature zone and loaded-swimming time in A (0.33 g/kg), B (1 g/kg), C (2 g/kg), and D (3 g/kg) dose of HSSH group, Tiepi Shihu (Dendrobium candidum, TPSH) group, and Liuwei Dihuang Wan (Six-Ingredient Rehmannia pill, LWDH) group increased significantly; the level of cAMP/cGMP decreased significantly (P<0.05, P<0.01); and the activities of Na+-K+-ATP-ase and Ca2+-Mg2+-ATP-ase decreased significantly (P<0.01,P<0.001). B, C, and D dose of HSSH group, TPSH group and LWDH group appeared to reduce the concentration of T3 and T4 (P<0.05, P<0.01, P<0.001). the activities of LDH were significantly increased in B and C dose of HSSH, TPSH and LWDH groups, and the levels of CK decreased (P<0.05, P<0.01, P<0.001). Conclusion With the effects of nourishing yin and clearing heat, Huoshan Shihu could regulate the energy metabolism rate, improve the loaded-swimming time, and reduce the activity of energy metabolizing enzymes and the thyroxine level of the mice with kidney-yin deficiency induced by thyroxine. Its anti-fatigue mechanism may be related to the regulation of energy metabolism enzymes. This study further proves that the property of Huoshan Shihu is cold.
