补阳还五汤对舒张性心衰大鼠心肌α-MHC、 β-MHC、Drp1 mRNA及线粒体超微结构的影响*

Effects of Buyang Huanwu decoction on myocardial α-MHC, β-MHC, Drp1 mRNA and mitochondrial ultra structure in rats with diastolic heart failure*

摘要: 目的 研究补阳还五汤对舒张性心衰(DHF)大鼠心肌肌球蛋白重链α、β(α-MHC、β-MHC) mRNA,GTP酶动力相关蛋白1(Drp1)mRNA 及线粒体超微结构的影响。方法 将48只SD大鼠随机分为假手术组与模型组,模型组大鼠运用腹主动脉缩窄法建立DHF大鼠模型。将造模成功的大鼠随机分为模型组、美托洛尔组和补阳还五汤组,灌胃给予相应药物,假手术组、模型组给予等量的去离子水,各组均每日灌胃1次。药物连续干预8周后,采用实时荧光定量 RT-PCR 法测定心肌 α-MHC、β-MHC和Drp1mRNA;采用透射电镜观察心肌线粒体超微结构。结果 与假手术组比较,模型组α-MHC mRNA表达水平升高(P<0.05),与模型组比较,美托洛尔组α-MHC mRNA表达水平降低(P<0.05)。与假手术组比较,模型组β-MHC mRNA表达水平降低(P<0.05),与模型组比较,补阳还五汤组、美托洛尔组β-MHC mRNA表达水平均升高(P<0.05)。与假手术组比较,模型组Drp1mRNA表达水平降低(P<0.05),与模型组比较,补阳还五汤组和美托洛尔组Drp1 mRNA表达水平升高(P<0.05)。透射电镜下观察:与假手术组比较,模型组大鼠线粒体超微结明显肿胀、空泡变形,数量减少;与模型组比较,补阳还五汤组、美托洛尔组大鼠线粒体形态明显改善,数量较前增加。结论 补阳还五汤可通过抑制α-MHC转变为β-MHC,从而延缓心肌重构、保护心肌组织。同时,通过抑制Drp1 mRNA对线粒体的分离作用,保护线粒体结构和功能,维持舒张性心衰时正常心肌能量代谢、减缓心衰进程。

Abstract: Objective To study the effect of Buyang Huanwu decoction (Yang-supplementing and Five-returning Decoction, BYHWT) on cardiac myosin heavy chain α, β (myosin heavy chain alpha and beta,α-MHC、β-MHC) mRNA and GTP enzyme dynami-lelated protrin 1(Drp 1) mRNA expressions in rats with diaslic heart foilure(DHF). Methods 48 SD Rats were randomly divided into sham operation group and model group,and the rat model was established by using abdominal aorta constriction method in model group of DHF rats. These rats were then randomly divided into model group, metoprolol group and BYHWT group; and intragastric intervention was administered accordingly, while the sham operation group and model group were given equal amount of deionized water, all once daily. After 8 weeks of continuous drug intervention, real-time fluorescence quantitative RT-PCR was used to determine myocardial α-MHC、β-MHC and Drp1 mRNA. Myocardial ultra structure was observed under Transmission Electron Microscope(TEM) . Result Compared with sham operation group, the expression level of α-MHCmRNA in model group increased (P<0.05); compared with model group, the α-MHCmRNA expression level of metoprolo group increased (P<0.05); compared with sham operation Group, the β-MHCmRNA expression level of model group decreased (P<0.05); compared with model group, the expression level of β-MHCmRNA in BYHWT group and metoprolol group increased (P<0.05); compared with sham operation Group, the Drp1 mRNAexpression level of model group decreased (P<0.05). Compared to the sham operation group, rat mitochondria in the model group seemed to be enlarged with hollow deformity and reduced numbers. Compared to the model group, rat mitochondria in the metoprolol and BYHWT group were improved in architecture and increased in quantity. Conclusion Buyang Huanwu decoction can inhibit α-MHC’s changing into β-MHC, thus delaying myocardial remodeling and protecting myocardium tissue. At the same time, by inhibiting the isolation of Drp1to mitochondria, protecting the structure and function of mitochondria, it can maintain the normal myocardial energy metabolism in diastolic heart failure and slow down the progression of heart failure.
