密蒙花颗粒对去势诱导的干眼症兔泪腺细胞IL-12及IL-6的影响*

Effects of Mimenghua Granule on IL-12 and IL-6 in lacrimal gland cells of castrated male rabbits with induced dry eyes*

摘要: 目的 观察密蒙花颗粒对去势雄兔泪腺细胞白细胞介素6(IL-6)及白细胞介素12(IL-12)表达的影响。方法 30只新西兰长耳白兔随机平均分为5组:正常组、模型组、密蒙花颗粒组、生理盐水组、睾酮组。通过切除双侧睾丸及附睾造模。给药30 d后,进行基础泪液分泌试验(SIT)和泪膜破裂时间(BUT)检测,然后将所有兔处死,摘取泪腺组织行HE染色,观察泪腺组织结构,免疫组化法检测各组兔泪腺组织中IL-6、IL-12的表达。结果 正常组兔泪腺组织排列整齐,结构清晰,未见IL-6、IL-12表达;模型组及生理盐水组泪腺组织排列不整齐,结构大片变性,IL-6、IL-12大量表达于细胞膜和细胞浆中;密蒙花颗粒组及睾酮组泪腺组织排列整齐,结构较清晰,仅见少量IL-6、IL-12表达于细胞膜和细胞浆中。与正常组比较,模型组、生理盐水组SIT值及BUT值减少(P<0.05),密蒙花颗粒组、睾酮组SIT及BUT值差异无统计学意义(P>0.05)。结论 密蒙花颗粒可以通过抑制泪腺组织中IL-6、IL-12的表达减轻泪腺组织的炎症,达到治疗干眼症的目的。

Abstract: Objective To observe the effects of Mimenghua (Flos Buddlejae) Granule (MG) on the expressions of IL-6 and IL-12 in lacrimal gland cells of castrated male rabbits.Methods Thirty New Zealand long-eared white rabbits were randomly and evenly divided into 5 groups: normal group, model group, MG group, saline group and testosterone group. Except rabbits in the normal group, all the others were castrated by bilateral orchiectomy and epididymectomy. SIT and BUT test were performed 1 day before and 30 days after castration. After 30 days of intervention, all rabbits were executed. Lacrimal gland tissues were collected for HE staining to observe the structure of lacrimal gland. Immunohistochemistry was used to detect the expressions of IL-6 and IL-12 in lacrimal gland tissues from rabbits of all groups.Results In the normal group, the lacrimal gland tissue was neatly arranged and the structure was clear. No expression of IL-6 and IL-12 was observed. In the model group and the saline group, the lacrimal gland tissue was not aligned, and the structure was degenerated. IL-6 and IL-12 were expressed in the cell membrane and in the cytoplasm. The lacrimal gland tissue of the MG group and the testosterone group were neatly arranged, and the structure was clear. Only small amounts of IL-6 and IL-12 were expressed in the cell membrane and cytoplasm. Compared with the normal group, the SIT and BUT values in the model group and saline group were decreased (P<0.05), and the differences in SIT and BUT values in the MG group and testosterone group were not statistically significant (P>0.05).Conclusion MG can reduce inflammation of lacrimal gland tissues by inhibiting the expression of IL-6 and IL-12 in lacrimal gland tissues to treat dry eyes.
