五子衍宗丸对肾精亏虚证大鼠睾丸支持细胞骨架蛋白表达及生精功能的影响

Effects of Wuzi Yanzong Pill on spermatogenesis by modulating expression of cytoskeleton protein in Sertoli cells in rats with kidney essence deficiency pattern*

摘要: 目的 观察五子衍宗丸对肾精亏虚证大鼠睾丸支持细胞骨架蛋白表达和生精功能的影响。方法 正常SD雄性大鼠25只,随机分为5组,每组5只。分别为正常组,模型组,五子衍宗丸低、中、高剂量组。正常组早晚均予0.5%羧甲基纤维素钠(50 mg/kg)灌胃;模型组上午给予雷公藤多苷(20 mg/kg)灌胃,下午给予0.5% 羧甲基纤维素钠(50 mg/kg)灌胃;低、中、高五子衍宗丸组大鼠上午予雷公藤多苷(20 mg/kg)灌胃,下午分别予0.5,1.0,2.0 g/kg五子衍宗丸灌胃。30 d后,取睾丸及附睾组织。检测大鼠精子总数、前向运动精子百分率,免疫组化法检测睾丸支持细胞中波形蛋白、α-微管蛋白的表达,TUNEL法检测生精细胞的凋亡。结果 和正常组相比,模型组精子总数、前向运动精子百分率下降,睾丸支持细胞中波形蛋白、α-微管蛋白表达下降,生精细胞凋亡增加;与模型组相比,五子衍宗丸中、高剂量组精子总数、前向运动精子百分率均提高;五子衍宗丸低、中、高剂量组睾丸支持细胞波形蛋白、α-微管蛋白的表达均增加,生精细胞的凋亡率下降。结论 五子衍宗丸可通过调控睾丸支持细胞骨架蛋白的表达,抑制生精细胞的凋亡,改善生精功能。

Abstract: Objective To observe the effects of Wuzi Yanzong (Five-ingredient Family Development) Pill (WYP) on expression of cytoskeletal proteins in Sertoli cells and on spermatogenesis in rats with kidney essence deficiency pattern.Methods Normal adult male Sprague-Dawley rats (n=25) were randomly divided into five groups: normal group, model group, low-dose WYP group, mid-dose WYP group and high-dose WYP group (n=5 in each group). The normal group was given 0.5% sodium carboxymethyl cellulose (CMC-Na) (50 mg/kg) by gavage in the morning and afternoon daily; the model group received multiglycosides of Tripterygium wilfordii Hook f (GTW) (20 mg/kg) in the morning and 0.5% CMC-Na (50 mg/kg) in the afternoon; and the WYP groups were given GTW (20 mg/kg) in the morning followed by WYP (0.5, 1.0, or 2.0 g/kg daily) in the afternoon respectively for 30 days. The testis and epididymal tissues were then collected to obtain the total sperm count and the percentage of progressively motile sperm. Immunohistochemistry was employed to detect the expressions of vimentin and α-tubulin in the Sertoli cells. TUNEL assay was used to measure the apoptosis of spermatogenic cells.Results Compared with the normal group, the total sperm count and the percentage of progressive motility (PM) in the model group were decreased. Expressions of vimentin and α-tubulin in the Sertoli cells were decreased. Apoptosis of spermatogenic cells was increased. Compared with the model group, the total sperm count and the PM percentage in the mid-dose WYP group and the high-dose WYP group were increased. Meanwhile, the expressions of vimentin and α-tubulin in Sertoli cells in all three WYP groups were increased with decreased apoptosis of spermatogenic cells.Conclusion WYP seem to improve spermatogenesis, possibly through modulating the expressions of cytoskeletal proteins in Sertoli cells and inhibiting the apoptosis of spermatogenic cells.
