心肾同治法对糖尿病大鼠视网膜神经营养因子的作用

Effect of “Heart-kidney therapy” on retinal neurotrophic factor in diabetic rats*

摘要: 目的 探讨心肾同治法指导下所创密蒙花复方对糖尿病大鼠视网膜病变神经功能损伤的保护机制。方法 SD大鼠随机分为正常组、模型组、密蒙花复方组。通过链脲佐菌素(STZ)腹腔注射诱导糖尿病大鼠模型。连续灌胃给药90 d,分别于第30、60、90天检测各组大鼠视网膜电生理指标,第90天时HE染色观察视网膜形态改变、TUNEL法检测视网膜神经节细胞凋亡及PCR、Western blot检测脑源性神经营养因子(BDNF)、视网膜睫状神经营养因子(CNTF)mRNA和蛋白表达。结果 与正常组比较,模型组大鼠视网膜电图振荡电位(OPs)波幅及b波波幅降低,视神经功能下降, 差异具有统计学意义(P<0.01)。模型组视网膜神经节细胞肿胀,排列紊乱,数目减少,BDNF和CNTF mRNA及蛋白表达下降。密蒙花复方组与模型组比较,OPs、b波波幅升高(P<0.01),视网膜神经节细胞凋亡数目减少,视网膜BDNF和CNTF mRNA及蛋白表达升高。结论 密蒙花复方可通过调节视网膜神经营养因子BDNF和CNTF表达水平对糖尿病大鼠视网膜病变神经功能损伤起到保护作用。

Abstract: Objective To explore the protective mechanism of “Heart-kidney therapy” for retinal nerve dysfunction in diabetic rats.Methods SD rats were randomly divided into normal group, model group and Mimenghua (Flos Buddlejae) compound prescription (MCP) group. Diabetic rat models were created by intraperitoneal injection of streptozotocin (STZ). Intragastric administration of MCP was carried out for 90 days consecutively. Visual electrophysiology was used to detect the visual function of diabetic rats on 30, 60, 90 d respectively. The morphological changes of retina were observed with HE staining on 90 d. Apoptosis of retinal ganglion cells was detected with TUNEL. The mRNA and protein expression of brain-derived neurotrophic factor (BDNF) and ciliary neurotrophic factor (CNTF) in retina were detected with PCR and Western blot.Results Visual electrophysiology examination showed that the OPs wave amplitude and b-wave amplitude in ERG were decreased which indicated decreased optic nerve function in the model group with statistically significant difference, compared with the normal group (P<0.01). HE staining showed that retinal ganglion cells in the model group were swollen, disordered and decreased in number. PCR and Western blot showed that the mRNA and protein expression of BDNF and CNTF were decreased, compared with the normal group. After the intervention with the Chinese prescription (Mimenghua prescription), the OPs wave amplitude and b-wave amplitude in ERG were increased in the MCP group, compared with the model group (P<0.01). The number of apoptotic retinal ganglion cells decreased, and mRNA and protein expression of BDNF and CNTF in retina increased in the MCP group, compared with the model group.Conclusion Mimenghua compound prescription can protect the retinal nerve function in diabetic rats by regulating the expression levels of mRNA and protein of BDNF and CNTF.
