五子衍宗丸对热应激大鼠睾丸的保护作用及机制研究*

Protective effect and mechanism of Wuzi Yanzong Pill in rat sertoli cells under heat stress*

摘要: 目的 构建热应激损伤大鼠睾丸支持细胞模型并验证其结果,阐明五子衍宗丸干预生精障碍疾病的作用机制。方法 通过在线数据库筛选五子衍宗丸活性单体抗生精障碍疾病靶点,分析靶点的生物学功能和作用通路。以大鼠睾丸支持细胞为研究对象,构建43 ℃水浴热应激模型,检测活性氧(reactive oxygen species, ROS)生成量变化和热休克蛋白70(heat-shock protein70, Hsp70)的表达变化。结果 网络药理学结果显示,五子衍宗丸主要通过调控活性氧水平、细胞外刺激应答等方式干预生精障碍疾病。ROS荧光探针结果显示,与空白组相比,模型组ROS的生成量明显增多,五子衍宗丸干预后ROS生成量明显减少。Western blot结果显示,五子衍宗丸组Hsp70表达量高于空白组且低于模型组。结论 五子衍宗丸可以通过减少ROS生成、降低Hsp70表达,保护热应激的睾丸支持细胞。

Abstract: Objective To explore the pharmacological mechanism of Wuzi Yanzong Pill in the treatment of male spermatogenic dysfunction by network pharmacology in rat sertoli cells. Methods Targets of anti-spermatogenic dysfunction in Wuzi Yanzong Pill were screened and the biological function and signal pathway were analyzed by online database. The heat stress model was constructed with rat sertoli cells to detect changes in the production of reactive oxygen species (ROS) and the expression of Hsp70. Results The results of network pharmacology showed that Wuzi Yanzong Pill mainly improved male spermatogenesis dysfunction by regulating ROS level and extracellular stimulus response etc. The ROS fluorescence probe results showed that, compared with the blank group, the ROS production was significantly increased in the model group, and significantly decreased in the Wuzi Yanzong Pill group. Western blot results showed that the expression of Hsp70 in Wuzi Yanzong Pill group was higher than that in the blank group and lower than that in the model group. Conclusion Wuzi Yanzong Pill seems to protect rat sertoli cells under heat stress by reducing ROS production and Hsp70 expression.
