雷公藤内酯醇对胶原诱导性关节炎大鼠γ-干扰素和白细胞介素-17A表达的影响*

Effects of triptolide on IFN-γ and IL-17A expression in collagen-induced arthritic rats*

摘要: 目的 研究雷公藤内酯醇(TPL)对胶原诱导性关节炎(CIA)大鼠γ-干扰素(IFN-γ)和白细胞介素-17A(IL-17A)表达的影响。方法 50只雌性Wistar大鼠被随机分为5组:正常组、模型组、甲氨蝶呤组、雷公藤多苷组和雷公藤内酯醇组,每组10只。正常组常规饲养,其余各组在复制CIA大鼠模型后分别以蒸馏水、甲氨蝶呤(1.78 mg/kg)、雷公藤多苷(20 mg/kg)及雷公藤内酯醇(4 mg/kg)灌胃干预4周,期间每周评价各组大鼠体质量、关节炎指数(AI)、足肿胀排水体积等。4周后ELISA法检测大鼠外周血IFN-γ和IL-17A水平;RT-q PCR检测大鼠膝关节滑膜IFN-γ mRNA和IL-17A mRNA的相对表达量。结果 给药基线期各给药组较正常组体质量减轻(P<0.05),AI值和双足排水体积均明显升高(P<0.05),给药后各组足趾肿胀持续减轻,AI值降低;同时期各给药组间AI和双足排水体积比较差异无统计学意义(P>0.05)。模型组及各给药组外周血IFN-γ、IL-17A较正常组明显升高(P<0.05),各给药组较模型组明显降低(P<0.05),各治疗组间差异无统计学意义(P>0.05)。模型组IFN-γ mRNA及IL-17A mRNA相对表达量较正常组明显升高(P<0.05),治疗后各组mRNA相对表达量均有下降,除甲氨蝶呤组外其余各组与模型组差异无统计学意义(P>0.05)。结论 雷公藤内酯醇可改善CIA大鼠关节肿胀程度、降低AI值及外周血IFN-γ和IL-17A的表达。

Abstract: Objective To study the effect of triptolide on IFN-γ and IL-17A expression in collagen-induced arthritis (CIA) rats. Methods 50 female Wistar rats were randomly divided into 5 groups (n=10 in each group): the normal group, model group, methotrexate (MTX) group and Tripterygium wilfordii glycosides tablet (TGT) group and triptolide (TPL) group. CIA rat model was established in all groups except the normal group. The normal group was fed routinely, while other groups were given by gavage with distilled water, MTX (1.78mg/kg), TGT (20mg/kg) and TPL (4mg/kg) respectively once a day for 4 weeks after CIA model was established. The weight, arthritis index (AI), foot swelling drainage volume of each groups were evaluated once a week. At week 4, ELISA assay was used to detect the IFN-γ and IL-17A levels in the peripheral blood of rats. RT-qPCR assay was used to detect the relative expression levels of IFN-γ mRNA and IL-17A mRNA in the rat knee joint. Results Compared with normal group the weight loss in each interventional group (MTX, TGT and TPL) at baseline(P< 0.05), and the AI value and the drainage volume of both feet were significantly increased (P< 0.05). After intervention, the swelling of the foot of the group reduced and AI value decreased continously. There was no significant difference in the AI and the double feet swelling drainage volume at the same time among drug-treated groups (P> 0.05). The IFN-γ and IL-17A levels in peripheral blood of model group and administration group were significantly higher compared to normal group (P< 0.05), and the level of each administration group was significantly lower than that in model group (P< 0.05).There was no significant difference between MTX, TGT and TPL group of it (P> 0.05). The IFN-γ mRNA and IL-17A mRNA relative expression of model group was significantly higher than that in normal group (P< 0.05), and the relative expression of mRNA were decreased after treatment. Except for MTX group, there was no significant difference between the other administration groups and the model group of the relative mRNA expression (P> 0.05). Conclusion Triptolide can relieve the joint swelling of CIA rats, reduce AI value and the expression of IFN-γ and IL-17A in the peripheral blood of CIA rats.
