血府逐瘀合二陈汤对肥胖大鼠内脏脂肪素等脂肪因子含量的影响*

Effects of Blood Mansion Stasis-Expelling Decoction combined with Two Matured Substances Decoction on adipocytokines in obese rats*

摘要: 目的 观察血府逐瘀合二陈汤对营养性肥胖大鼠皮下脂肪组织、内脏脂肪组织、血清中内脏脂肪素等脂肪因子含量的影响。方法 利用高脂饲养方法复制营养性肥胖大鼠模型,依体质量随机分为4组,即血府逐瘀汤组、补阳还五汤组、血府逐瘀合二陈汤组、模型组,另设空白对照组,每组10只。前3组分别给予血府逐瘀汤(13.00 g/kg)、补阳还五汤(23.75 g/kg)、血府逐瘀合二陈汤(19.67 g/kg)灌胃,后2组灌胃等量的生理盐水,连续干预4周。酶联免疫法检测皮下脂肪组织、内脏脂肪组织、血清中的内脏脂肪素(PBEF)、脂联素、瘦素(Leptin)、纤溶酶原激活物抑制剂-1(PAI-1)、肿瘤坏死因子α(TNFα)和内皮素-1(血浆ET-1)的含量。结果 ①模型大鼠皮下脂肪、内脏脂肪、血清中PBEF含量均升高,血府逐瘀汤组、血府逐瘀合二陈汤组较模型组降低(P<0.05),血府逐瘀合二陈汤组较血府逐瘀汤组下降更显著(P<0.05)。②模型大鼠皮下脂肪、内脏脂肪、血清中脂联素含量均降低,各活血方组皮下脂肪组织、血清中脂联素含量较模型组升高(P<0.05),补阳还五汤组较血府逐瘀汤组、血府逐瘀汤合二陈汤组均显著升高(P<0.05),而且补阳还五汤组较模型组内脏脂肪组织脂联素含量也升高(P<0.05)。③模型大鼠皮下脂肪中Leptin含量降低,内脏脂肪、血清中含量升高(P<0.05)。三活血方均可降低血清中Leptin含量,血府逐瘀合二陈汤组内脏脂肪组织Leptin含量也有降低(P<0.05)。④模型大鼠内脏脂肪中TNFα含量降低(P<0.05),血清浓度和皮下脂肪中TNFα含量在各组之间差异均无统计学意义(P>0.05)。补阳还五汤组、血府逐瘀合二陈汤组内脏脂肪中TNFα含量较模型组升高(P<0.05)。⑤各组大鼠皮下脂肪、内脏脂肪、血浆中ET-1含量差异无统计学意义(P>0.05)。⑥模型组内脏脂肪、皮下脂肪中PAI-1含量均升高(P<0.05)。血府逐瘀汤组、血府逐瘀合二陈汤组内脏脂肪中PAI-1含量下降(P<0.05),血府逐瘀汤组皮下脂肪组织中PAI-1含量也下降(P<0.05)。结论 内脏脂肪、皮下脂肪组织分泌功能存在显著差异,模型大鼠脂肪因子分泌紊乱,活血化瘀药物有一定调节作用。

Abstract: Objective To investigate the effects of Blood Mansion Stasis-Expelling Decoction (Xuefu Zhuyu Tang) in combination with Two Matured Substances Decoction (Er Chen Tang) on adipocytokines in subcutaneous adipose tissues, visceral adipose tissues and serum of obese rats. Methods Nutritional obesity rat models were established with high-fat feeding, and they were randomly divided into four groups according to body mass, namely, Blood Mansion Stasis-Expelling Decoction group, Yang-Supplementing and Five-Returning Decoction group (Buyang Huanwu Tang), Blood Mansion Stasis-Expelling Decoction in combination with Two Matured Substances Decoction group, model group plus blank control group (n=10). The first three groups were administered with Blood Mansion Stasis-Expelling Decoction, Yang-Supplementing and Five-Returning Decoction and Blood Mansion Stasis-Expelling Decoction in combination with Two Matured Substances Decoction, respectively at 10 times the adult dosage. The latter two groups were fed with saline. All the groups were intervened for 4 consecutive weeks. The levels of subcutaneous adipose tissue, visceral adipose tissue, serum of visceral fat element, adiponectin, leptin, fibrinolytic enzyme activators inhibitor-1, tumor necrosis factor alpha, and endothelin-1 (plasma) were measured by using Elisa method. Results ①The visfatin levels in subcutaneous fat, visceral fat and serum were increased in the model group, which were lowered in the Blood Mansion Stasis-Expelling Decoction group and the Blood Mansion Stasis-Expelling Decoction in combination with Two Matured Substances Decoction group compared with the model group (P<0.05), more significant in the latter (P<0.05). ②The contents of subcutaneous fat, visceral fat and serum adiponectin were reduced in the model group,while the contents of subcutaneous adipose tissue and serum adiponectin in every blood-activating group were increased compared with that of model group (P<0.05), more significant in Yang-Supplementing and Five-Returning Decoction group (P<0.05). Moreover, the level of adiponectin in visceral adipose tissue in Yang-Supplementing and Five-Returning Decoction group was also higher than that in model group(P<0.05). ③The level of leptin was decreased in subcutaneous fat and increased in visceral fat and in serum in model group(P<0.05). Serum leptin levels were lowered in three blood-activating groups. Moreover, leptin in visceral adipose tissue was also lowered in Blood Mansion Stasis-Expelling Decoction in combination with Two Matured Substances Decoction group (P<0.05).④The level of TNF alpha in visceral fat was reduced (P<0.05), No significant difference was observed in serum concentration and content of TNF alpha in subcutaneous fat between groups(P>0.05). Compared with model group, levels of TNF alpha in visceral fat were elevated in Yang-Supplementing and Five-Returning Decoction group and Blood Mansion Stasis-Expelling Decoction in combination with Two Matured Substances Decoction group (P<0.05). ⑤No statistically significant differences were shown in endothelin-1 in subcutaneous fat, visceral fat and plasma (P>0.05). ⑥The levels of PAI-1 in visceral fat and subcutaneous fat were increased in model group (P<0.05). However, PAI-1 level in visceral fat was reduced in Blood Mansion Stasis-Expelling Decoction group, and Blood Mansion Stasis-Expelling Decoction in combination with Two Matured Substances Decoction group (P<0.05). In addition, the level of PAI-1 in subcutaneous fat tissues was also lowered (P<0.05). Conclusion There was a significant endocrinological difference between visceral and subcutaneous adipose tissue. For model rats, formulas that can activate the blood and resolve blood stasis have therapeutical effects on disorder of adipocytokines.
