Protective effect of Yiqi Jiedu Fang on damaged mitochondria in rats with focal cerebral ischemia

Abstract: Objective To investigate the protective effect of Yiqi Jiedu Fang and its different components on cerebral mitochondria in rats with focal cerebra ischemia. Methods Male SD rats were randomly divided into sham-operation group, model group, control group (with positive drug EGb761, 4 mg/kg), high-dose, mid-dose and low-dose Yiqi Jiedu Fang groups (25 mg/kg, 5 mg/kg and 1 mg/kg), high-dose, mid-dose and low-dose ginsenosides groups (25 mg/kg, 5 mg/kg and 1 mg/kg), high-dose, mid-dose and low-dose berberine groups (25 mg/kg, 5 mg/kg and 1 mg/kg) and high-dose, mid-dose and low-dose geniposide groups (25 mg/kg, 5 mg/kg and 1 mg/kg). The model was established with middle cerebral artery occlusion (MCAO). The neuroethology was scored 24 h after MCAO, and suspension of mitochondria from ischemic brain tissue was prepared for reviewing the morphological and functional indexes. Results Compared with sham-operation group, the scores of neuroethology increased significantly in model group (P<0.01). Compared with model group, the scores of neuroethology decreased significantly in all Yiqi Jiedu Fang groups and high-dose other component groups (P<0.05 or P<0.01). The function of mitochondria was improved at different levels in all medicated groups. Compared with other component groups, the improvements of neuroethological scores and mitochondrial function were more significant in all Yiqi Jiedu Fang groups showing some dose-dependent manner. Conclusion Yiqi Jiedu Fang can play a role in mitochondrial protection, which may be its an important mechanism in antagonizing cerebral ischemia and treating ischemic stroke. Moreover, compared with its three individual components (ginsenosides, berberine and geniposide), the combination formula (Yiqi Jiedu Fang) exerted better pharmacological effects to all indexes. It is suggested that combined medicinal administration will play better integrated therapeutic effects than its single component administration.
