Influences of Chinese medicinal on protein expressions of CTLA-1 and PD-1 in CTL cells in SiHa-CTL-Treg co-culture system

Abstract: Objective To understand whether Xiaodu Shuan can inhibit Treg cell functin to change the local immune microenvironment for inhibiting tumor growth through detecting the protein expressions of cytotoxic T lymphocyte antigen-4 (CTLA-4) and programed death-1 (PD-1). Methods Treg cells and CTL were separated by using immunomagnetic beads. The protein expressions of CTLA-4 and PD-1 were detected by using Wstern-blot method. Results In CTL-Treg co-culture group, the protein expressions of CTLA-4 and PD-1 increased significantly, which decrased significantly after the intervention of β-elemene and Xiaodu Shuan especially in Xiaodu Shuan group. Conclusion Xiaodu Shuan and β-elemene all can inhibit the up-ragulation effect of Treg on CTLA-4 and PD-1 in CTL.
