Influences of Guipi Tang on phenotype Sca-1 and CD34 of hematopoietic stem cell and mitotic cycle in benzolism mice

Abstract: Objective To observe the influences of Guipi Tang on phenotype Sca-1 and CD34+ of hematopoietic stem cell (HSC) and cell cycle in benzolism mice, and discuss its possible mechanism in hematopoietic regulation. Methods Kunming mice (n=72, 8-12 weeks old, half male and half female) were randomly divided into normal group, model group, Western drug group, and high-dose, mid-dose and low-dose groups (each n=12). The benzolism model was established by putting mice into a manual simulated condition with high concentration benzene (2000 mg/m3) for 8 h a day for 14 d continuously, until the level of WBC dropped below 4.5×109/L. The high-dose, mid-dose and low-dose groups were orally given solution of Guipi Tang (0.072g/mL, 0.036g/mL and 0.018g/mL) respectively and normal group and model group were orally given normal saline water (20 mL/kg) once a day for 3 weeks. The influences of Guipi Tang on mitotic cycle in peripheral blood and bone marrow and phenotype Sca-1 and CD34+ of HSC were detected by using automotive blood cell analyzer and flow cytometry. Results HSC was activated, expressions of Sca-1 and CD34+ increased and primordial HSC increased in mid-dose group and Western drug group (P<0.05). In high-dose group, the cell transition from G0/G1 phase to S phase and from S phase to G2/M phase were promoted, proliferation index increased significantly (P<0.05 or P<0.01), and levels of WBC and HBG increased. The change of platelet was not significant. Conclusion Guipi Tang may improve the quantity and function of early HSC and hemopoietic ability through promoting the cell transition from G0/G1 phase to S phase and from S phase to G2/M phase.
