Influence of therapy of reinforcing kidney and regulating qi on BDNF expression in rats with post-encephalorrhagia depression

Abstract: Objective To observe the expression of brain-derived neurotrophic factor (BDNF) in rats with post-encephalorrhagia depression, and study the mechanism of therapy of reinforcing kidney and regulating qi (Yinao Jieyu Fang) in treatment of post-encephalorrhagia depression. Methods The rats (n=32) with the similar scores of open-field test were chosen and randomly divided into normal group, model group, Chinese medicial group (treated with Yinao Jieyu Fang) and Western medicial group (treated with Baiyoujie, each n=8). The changes of BDNF expression were observed by using immunohistochemistry technique in normal group, model group, Chinese medicial group and Western medicial group. Results In model group, the areal density (AD), optical density (OD) in hippocampus CA1 and CA3 zones and OD in CA2 zone of BDNF positive cells decreased significantly compared with normal group (P<0.05), and AD of BDNF positive cells in CA1 zone, and AD and OD in CA3 zone increased significantly in Western medicinal group compared with model group (P<0.05). Compared with normal group, AD and OD in cingulum cortex and AD in hypothalamus decreased significantly in model group (P<0.05). In Chinese medicial group, AD and OD of BDNF positive cells increased in CA3 zone compared with model group (P<0.05), OD increased significantly in cingulum cortex compared with model group, and AD increased in hypothalamus compared with model group (P<0.05). Conclusion Yinao Jieyu Fang can promote the repair and regeneration of neurons and relive depression symptoms through increasing BDNF expression in rats with post-encephalorrhagia depression.
