Influences of Majie Chuanke Fang on endothelin, nitric oxide and pulmonary artery in rat model of pulmonary heart disease

Abstract: Objective To observe the influences of Majie Chuanke Fang on endothelin (ET), nitric oxide (NO) and pulmonary artery in rat model of pulmonary heart disease, and discuss the therapeutic mechanism of Majie Chuanke Fang in treatment of pulmonary heart disease. Methods SD rats (n=56) were randomly divided into high-dose, mid-dose and low-dose Majie Chuanke Fang groups (high-dose group, mid-dose group and low-dose group), Feilike group, captopril group, model group and normal group. The rat model of pulmonary heart disease was established with monocrotaline. High-dose group, mid-dose group and low-dose group were given Majie Chuanke Fang in doses of 12 g/kg, 6 g/kg and 3 g/kg respectively, Feilike group, Feilike Capsules (1.08 g/kg) and captopril group, captopril (0.01 mg/kg). After 14 d, the content of NO and ET-1 was detected, and right ventricle hypertrophy index (RVHI), pulmonary artery area and pulmonary artery wall area were calculated,and morphological changes of pulmonary artery were observed in all groups. Results The content of ET-1 decreased significantly in high-dose group, mid-dose group and low-dose group, and NO content increased significantly in mid-dose group and low-dose group. The unbalance of ET-1/NO was ameliorated in high-dose group, mid-dose group and low-dose group (P<0.05 or P<0.01). The pulmonary artery wall area and lumen area were significantly reduced in low-dose group compared with model group (P<0.01). Majie Chuanke Fang had no significant effect on the indexes of right ventricular hypertrophy. The results of histopathologic examination showed that Majie Chuanke Fang reduced pulmonary artery wall thickness and increased lumen area (P<0.01) in varying degrees. Conclusion Majie Chuanke Fang can relieve the sympotoms of acute pulmonary heart disease through alleviating the unbalance of ET-1/NO, right ventricle hypertrophy and pulmonary vessel remodeling.
