Effects of Feixian Formula on vascular endothelial growth factor (VEGF) and VEGF receptor 1 and 2 mRNA expression in rat model of interstitial pulmonary fibrosis

Abstract: Objective To investigate the inhabitation effects of Feixian Formula on angiogenesis in rat with interstitial pulmonary fibrosis. Methods Male Wistar rats were randomly divided into 7 groups: sham-operation group, model group, 3 groups treated with high-,mid-,and low-dose of Feixian Formula (high-dose group, mid-dose group and low-dose group), and 2 groups treated with western medicine losartan and prednisone respectively(losartan group and prednisone group). All the rats, except for those in sham-operation group, were established as the model of interstitial pulmonary fibrosis by perfusion of bleomycin (3 mg/kg) through tracheal intubation. From the second day after modeling, all rats were intragastrically administrated with drugs or saline, according to different groups designed. The rats were sacrificed at the 14th day and at the 28th day. The pathological changes of interstitial pulmonary by HE staining and MASSON staining were observed, to evaluate the degree of the alveolitis and pulmonary fibrosis. Expressions of VEGF, VEGF 1 and 2 mRNA were detected by RT-PCR. Results Both high-dose and mid-dose Feixian Formula had obvious effects of inhabiting alveolitis and pulmonary fibrosis , as well as downregualting the expressions of VEGF, VEGFR1 and 2 mRNA, which were remarkable in mide-dose group compared with model group (P<0.05). Conclusion The effects of Feixian Formula on inhabition of angiogenesis and resistance to interstitial pulmonary fibrosis may be involved in the down-regulation of VEGF, VEGFR1 and VEGFR2 gene expression.
