Effects on P21WAF1 and CyclinD1 in chronic atrophic gastritis with dysplasia in rats: TCM therapy of replenishing qi, resolving stasis and detoxification

Abstract: Objective To observe the effects of Xiaopi granule on gene P21WAF1 and gene CyclinD1, which was formulated by replenishing qi, resolving stasis and detoxification, in chronic atrophic gastritis with dysplasia in rats. Methods 50 of 60 Wistar rats were randomly selected to be modeled, and the rest of rats in blank group (n=10). After establishment of model, the alive rats (n=33) were randomly divided into model group, vitacoenzyme tablet group (western medicinal group) and Xiaopi granule group(Chinese medicinal group) (each n=11). Model group, western medicinal group and Chinese medicinal group were intragastrically administered once a day saline water, vitacoenzyme and Xiaopi granule respectively for 12 consecutive weeks. Real-time fluorescence quantitative polymerase chain reaction (RT-PCR) and Western blot were used to determine the expressions of P21WAF1 and CyclinD1 genes in rats of all groups. Results The expression of P21WAF1mRNA and protein significantly increased in Chinese medicinal group compared with model group and western medicinal group (P<0.05). The expression of CyclinD1 mRNA and protein dramatically decreased in Chinese medicinal group compared with model group and western medicinal group (P<0.05). Conclusion Chinese medicine Xiaopi granule can reverse the gastric mucosal dysplasia, due to increasing the expression of P21WAF1 and inhibition the expression of CyclinD1, leading to redressing the balance of P21-CDK-CyclinD1 pathway.
