Effect of needling sensation in acupuncture treatment on uterine in primary dysmenorrheal rats of cold stagnation syndrome*

Abstract: Objective To investigate the effect of needling sensation (de qi) when acupuncturing at Sanyinjiao point (SP6) on the uterine contractions, uterine microcirculation and the body torsion reaction in primary dysmenorrheal (PD )rat model with cold stagnation syndrome.Methods 96 anestrous female SD rats were divided into four groups (n=24 each) of saline control group(control group), PD model with syndrome of cold stagnation group(model group), acupuncture treatment with obtaining “de qi ”sensation group(deqi group)and acupuncture treatment without obtaining “de qi ”sensation group (no deqi group). Rat model with PD was induced by whole body freezing combined with intraperitoneal injection of estradiol benzoate. After modeling, bilateral Sanyinjiao points (SP6) of rats were stimulated by acupuncture, and the indexes of uterine contraction, uterine microcirculation and the body torsion reaction were recorded and analyzed. Results Compared with control group, the uterine activity increased significantly in model group. The number of uterine contraction wave and uterine activity decreased markedly in deqi group compared with model group. The diameter of capillaries and microvessels of uterine decreased significantly in both model group and no deqi group compared with control group, while microvessel relaxation at each time point and capillaries relaxation at 40, 50, 60 min after acupuncture in deqi group were observed. Meanwhile, compared with model group, writhing times and scores decreased markedly with writhing latency increasing in deqi group.Conclusion The effect of relieving uterine contraction and improving uterine microcirculation in deqi group maybe relies on obtaining needling sensation, which is the key of efficacy of acupuncture treatment.
