Optimization of determination method of typhaneoside and isorhamnetin-3-O-neohesperidoside in Pollen Typhae*

Abstract: Objective To establish the determination method of typhaneoside and isorhamnetin-3-O-neohesperidoside in Pollen Typhae, Puhuang. Methods Single-factor test method was used to optimize the procedure of sample preparation. HPLC was used to detect the contents of typhaneoside and isorhamnetin-3-O-neohesperidoside in Pollen Typhae, Waters SunFireTM C18 (4.6 mm×250 mm, 5 μm) column at 20℃ and 254 nm detection wavelength, with mobile phase of acetonitrile-0.05% phosphoric acid (V/V) and flow rate of 1 mL/min. Results The sample solution was prepared by extracting from Pollen Typhae with 80 times amount of methanol for refluxing 1.5 h. Typhaneoside showed a good linear relationship at the range of 0.245 2~1.471 0 μg (r = 0.999 9), with the recovery of 98.47% (RSD=2.05%, n=6). Isorhamnetin-3-O-neohesperidoside showed a good linear relationship at the range of 0.191 2~1.147 4 μg (r = 0.999 7), with the recovery of 100.56% (RSD = 2.56%, n=6). Conclusion The method established is simple, rapid, accurate and repeatable, which built up a good basis for the quality control and evaluation of Pollen Typhae.
