Effects of Yiqi Yangyin Formula on the outcome of controlled ovarian hyperstimulation in poor ovarian responders*

Abstract: Objective To observe the effects of Yiqi Yangyin(supplementing qi and nourishing yin) Formula on the embryo quality and pregnancy outcome in poor ovarian responders treated with controlled ovarian hyperstimulation (COH), and to study its effects on growth differentiation factor-9 (GDF-9)and bone morphogenetic protein-15 (BMP-15) in serum and follicular fluid. Methods Total 94 cycles of poor ovarian responders undergoing IVF-ET were grouped by the first day of COH: on odd dates into control group(n=48), on even dates into treatment group(n=46). Two groups were both stimulated by minimal stimulation protocol, in addition, the treatment group took Yiqi Yangyin Formula from the second day of the menstrual cycle to the day of HCG injection. Then the total dosage of Gn, the days of Gn stimulation, number of retrieval oocytes, fertilization rate, transplant rate, pregnancy rate and cycle cancellation rate were recorded. And the levels of GDF-9 and BMP-15 of serum and follicular fluid were detected using ELISA, and the protein expression were measured by Western blot. Results Compared with the control group, the total dosage of Gn and cycle cancellation rate decreased significantly in the treatment group, while the estradiol level, thickness of endometrium, retrieval oocytes, pregnancy rate were all increased(P<0.05). The protein expressions of GDF-9 and BMP-15 in granulose cells in treatment group were significantly higher than those in the control group(P<0.05). Conclusion Yiqi Yangyin Formula can collaborate with exogenous sexual hormones to improve the ovarian respond, so as to increase the poor ovarian responders’ retrieval oocytes and pregnancy rate. The potential mechanism is linked to the effects of Yiqi Yangyin Formula on regulating the expression of GDF-9 and BMP-15 in ovaries.
