Effect of acupuncture on protein expression of 5-HTT, 5-HT1AR, NEα2R of post-stroke depression rats*

Abstract: Objective To observe the effect of acupuncture intervention on neurotransmitters and their receptors of post-stroke depression (PSD) rats, so as to explore the mechanism from molecular biological level. Methods Altogether 120 SD male rats randomly divide into blank group (n=10), model group(n=30), acupuncture group (n=40) and medication group (n=40). PSD rat model was induced by middle cerebral artery occlusion(MCAO) and chronic unpredictable mild stress (CUMS). After modeling, rats of medication group were intragastrically administered fluoxetine at the dose of 2 mg /kg once daily, those of acupuncture group were needled at bilateral acupoints of Baihui (GV 20), Fengfu (GV 16), Shenmen (TF 4),Taichong (LR 3) for 20 min once daily with manipulating needles once. The treatment (medication or acupuncture) were underwent three courses for 21 days . Then protein expression of norepinephrine receptor 2R(NEα2R), 5-hydroxytryptamine receptor1A (5-HT1AR) and 5-hydroxytryptamine transporter(5-HTT) in PSD rat hippocampus, raphe nucleus, locus coeruleus were detected by Western blot. Results Compared with blank group, 5-HTT protein expression in hippocampus, raphe nucleus and locus coeruleus in model group decreased significantly(P<0.01);NEα2 R protein expression in hippocampus, raphe nucleus, locus coeruleus of model group increased significantly(P<0.01); 5-HT1A R protein expression in hippocampus, raphe nucleus of model group increased significantly(P<0.01), that in locus coeruleus of model group increased(P<0.05). Compared with model group, 5-HTT protein expression in hippocampus, raphe nucleus, locus coeruleus of acupuncture group and medication group significantly increased(P<0.01);NEα 2R protein expression in hippocampus, raphe nucleus, locus coeruleus of acupuncture group and medication group significantly decreased(P<0.01). 5-HT1A R protein expression in hippocampus of acupuncture group and medication group significantly decreased(P<0.01),in raphe nucleus, locus coeruleus of acupuncture group and medication group decreased(P<0.05). Compared with medication group, there were no differences of 5-HTT protein expression, NEα2 R protein expression, 5-HT1AR protein expression in hippocampus, raphe nucleus, locus coeruleus of acupuncture group(P﹥0.05).Conclusion Acupuncture can reduce the expression of 5-HT1AR and NEα2 R in post-stroke depression model rats, increase the expression of 5-HTT. This suggests that the effect of acupuncture against depression due to stroke via increasing the content or transportation rate of 5-HT and NE.
