Chemical composition from Eupolyphaga Sinensis Walker*

Abstract: Objective To isolate new chemical compositions from Eupolyphaga Sinensis Walker (woodlouse) and evaluate their inhibitory activity on PKD1/TRPP2 activity,an indicator of polycystic kidney disease. Methods Isolation and purification by using silica gel normal and reversed phase chromatography, and semi-preparative HPLC. The structure of chemical compounds were determined based on their physic-chemical property and spectral parameters. Results Altogether eight chemical compositions were isolated and identified from ethyl acetate part of 70% acetone extracts, whose structures were determined by 1D and 2D NMR spectroscopic analysis as 1H-pyrrolo[1,2-a][1,4]diazepine-1,5(2H)-dione,hexahydro-3-Para phenyl phenol (1), thymine (2), 2-methyl-6-(2’3’4’-3 OH-butyl) pyrazine (3), 3-hydroxy-pyridinol (4), (3S,8aS)-3-isobutylhexahydropyrrolo[1,2-a]pyrazine-1,4-dione(5),3-ethyl-6,8-dihydroxy-7-methyl-3,4-dihydroisochromen-1-one (6), 3-(p-hydroxyphenyl)-N-methylpropionamide (7), and 6,8-dihydroxy-3,7-dimethyl-3,4-dihydro-1H-isochromen-1-one (8). Conclusion Composition 1 was a new composition, with inhibitory effect on PKD1/TRPP2 activity, and composition 2~8 were first isolated from woodlouse.
