Evaluation and formulation of Compound Danshen Adhesive Pellet

Abstract: Objective To evaluate chitosan, hydroxypropyl methyl cellulose K100M(HPMCK100M) and carbopol as adhesion materials in this design of Compound Danshen formula. By studying the adhesion in vitro and in vivo, an appropriate formula was screened.Methods In vitro, the minimum peel strength method was performed to determine the peel force of Adhesive Pellet from swine intestine, and the tissue residue method was applied to measure the number of remaining pellets in swine intestine. In vivo, the adhesive index was obtained by direct intragastric administration by which the number of remaining pellets in intestine were detected. Results The tissue residue method showed that the remaining pellets of adhesion materials accounting for 40% were more than those of adhension materials accounting for 30% for the same amount of time.The minimum peel strength method proved that the minimum pell strength was stronger in pellets of 40% adhension materials,conformable to the Results of the tissue residue method. In vivo the adhesive index of 40% of adhesion materials was less than that of 30%, but some pellets were left in swine stomach. Conclusion The same proportion of adhesive materials showed the same adhesion. The higher the proportion was, the stronger the adhesion was and the longer retention time in the stomach which could destroy the active ingredients in Compound Danshen Adhesive Pellet.
