Time-variant effects of needling Zhigou on the rest-state fractional amplitude of low-frequency fluctuations inside healthy human brains*

Abstract: Objective To observe time-variant effects of needling Zhigou (SJ6) on fractional Amplitude of Low Frequency Fluctuations(fALFF) in thebrainsof healthy subjects atresting state by using functional magnetic imaging (fMRI). Methods Eighteen healthy participants were scanned with fMRI. The scanning involved pre-treatment pure resting state(R1), then needle-retention resting states(R2-R5) and post-treatment resting states (R6~R7). All gathered data was analyzed with fALFF method. Results ①The fALFF of bilateral precuneus transformed from inactivation before treatment topositive activation after treatment and maintained tillR6 state(except for R5), and the amplitude as well as clusters of activation both remained stable. ②fALFF of superior frontal gyrus and medial frontal gyrus were strengthened on the basis of pre- treatment activation within R1; alternating positive activation on both sides of the two gyri were observed within R2, R4, R5 and R6 states except for R3 state, during within which both sides had positive activations. ③Compared with R1, the fALFF of inferior parietal lobule transformed from deactivations to positive activations only within R2 and R3 state, as there were no significant changes within R4-R7.④The fALFF of cerebellum posterior lobe presented positive activation within R2 state and both cerebellum anterior and posterior lobe presented positive activation within R6 while made deactivations within R3 and R5 states. Conclusion s The findings of this study could provide certain theoretical foundation for acupuncture on SJ6 in the treatment of diseases with weakened function of precuneus, superior frontal gyrus, medial frontal gyrus, inferior parietal lobule and strengthened function of cerebellum anterior and posterior lobefrom the perspective of brain energy metabolism. It could also provide some scientific evidence for the timing of manipulating needle during needle retention and studies of post-acupuncture effects.
