Metabonomic study of compound gouteng antihypertensive tablet's effects on left ventricular hypertrophy in hypertensive rats*

Abstract: Objective To study the metabolite changes and mechanism of compound gouteng (uncaria) anti-hypertensive tablet in the treatment of left ventricular hypertrophy. Methods 24 rats with spontaneously hypertension were randomly divided into the model group, compound gouteng antihypertensive tablet(CGTA) group and telmisartan(TMST) group. WKY (Wistar-Kyoto) rats were chosen as the control group. Metabolites were detected and analyzed by using gas chromatography-mass spectrometry (GC-MS), and the differential metabolite profiles of left ventricular myocardium were analyzed with partial least squares (PLS). Based on the importance of independent variables (VIP) and the coefficient of independent variable, the left ventricular metabolites of the four groups were investigated. Results The variable mapping of SHR group and CGTA group showed positive correlations with CGTA group were shown in cytosine nucleotide, putrescine, succinic acid, D-acid, glycerol fructose, inosine, dihydrolanosterol, ascorbic acid, glutamic acid, and 1,5-ag; and that negative correlations were found in lactic acid, 3-phosphoglycerate, ribose, xylitol, and proline. The variable maps of CGTA group and TMST group showed positive correlation with CGTA group were identified in putrescine, cytosine nucleotide acid, glycerol, lyxonic acid, twenty-four acid, inositol, and inosine; negative correlation were present in xylitol, sorbitol, ribose, and lactic acid. Conclusion The compound gouteng antihypertensive tablet may have specific effects on cell structure, energy optimization and oxidative stress reaction.
