Effects of electroacupuncture on astrocytes at subventricular zone in rats with hyperlipidemia and cerebral ischemia*

Abstract: Objective To observe effects of electroacupuncture on astrocyte at subventricular zone (SVZ) of the ipsilateral forebrain in rats with hyperlipidemia and cerebral ischemia. Methods 49 adult male SD rats were randomly divided into normal group, model group, sham operation group, electroacupuncture 1 (EA1) group and electroacupuncture 2 (EA2) group. Rats were fed with high-fat diet for 42 days except those in the normal group. Rats in EA1 group were needled at ST40, once a day for 7 days. Rats in the model and EA groups were induced with hyperlipidemia and cerebral ischemia by 50% FeCl3 at the 50th day. Rats in EA group were needled at ST40 and GV20, once a day for 14 days. Behavioral were assessed with neurological function score. Levels of total cholesterol (TC), triglyceride (TG), high-density lipoprotein (HDL) and low-density lipoprotein (LDL) in serum were tested by biochemical methods. Hematoxylin-eosin (HE) staining assay was used to evaluate histopathology while immunohistochemically assay was used to evaluate GFAP at SVZ. Results TC, TG, LDL rose significantly and HDL reduced significantly in model group after hyperlipidemia rats with cerebral ischemia was established (P<0.05). The level of TC and LDL decreased significantly in EA group (P<0.05). Rats in model group had higher neurological function score; shrinking neurons were present in HE staining, indicating cerebral ischemia. Percentage of GFAP-positive cells area at SVZ increased (P<0.01). Electroacupuncture seemed to improve neurobehavioral score and recover cerebral ischemia injure. Percentage of GFAP area in rats of EA group had increased from day 1 to day 14, and at day 7 percentage of GFAP area in EA1 group was higher than that in EA2 group (P<0.05). Conclusion Stimulating ST40 in hyperlipidemia period, and stimulating ST40 and GV20 after hMCAT occurred can activate astrocytes and repair damaged nervous system by regulating blood lipid metabolism.
