    Functional localization of five-zang system of Chinese medicine

    Western medicine classifies the functions of human body into 10 anatomical systems, while Chinese medicine classifies the functions into 5-zang systems. Because of Western medicine and Chinese medicine have the same study object, a living human body, there must be matching relationships between the functions that coming from the two kinds of classification. This paper founded the components, specific functions and anatomical locations of five-zang system. The specific functions of Pi system are corresponding to the functions of digestive system, hemostasia function of liver, and motive power of skeletal muscle. The special functions of Fei system include inhaling oxygen and eliminating carbon dioxide of the respiratory system, pronunciation, and defense function of the immune system. The specific functions of Shen system include the functions of the urinary system, reproductive system, endocrine system and hematopoietic system, discriminating and clearing away senile or variable cells of the immune system, and support function of skeleton of the motor system. The special functions of Gan system are corresponding to producing and transferring sensory signals, and transferring motor signals to regulate somatic and visceral movements of the nervous system. The special functions of Xin system include brain's distinguishing different sensory signals, generating mental activities, such as consciousness, thoughts, emotions, and etc., and substance transporting functions of the circulatory system.

    Theory of time-concerned treatment in Huangdineijing: an analysis based on xiang (phenomena) thinking mode

    The paper discussed, based on xiang (phenomena) thinking mode, the theory of time-concerned treatment in Huangdineijing from 3 aspects: the meaning of time, changes of time and appropriate time of action. The author pointed out that time, manifesting as dynamic xiang, is essentially the waxing and waning of yin qi and yang qi. The essence of time-concerned treatment is seasonal qi-concerned treatment, so the integration of xiang of time, position of time and qi of time will be helpful in holding the phenomena and changes of qi of time and grasping therapeutic measures in health preservation, disease prevention and formula design.

    Identification of sovereign medicinal in decocting formulas of Shanghanlun by using formula metrology method

    Objective To identify the sovereign medicinal in decocting formulas in Shanghanlun, and discuss their commonness and characteristics in order to study deeply the theory of formula design in Shanghanlun and apply flexibly the formulas. Methods A Song Dynasty block-printed edition of Shanghanlun (inscribed by ZHAO Kai-mei in the Ming Dynasty) was taken as a sample, and decocting formulas were selected from it. The sovereign medicinal were identified by using formula metrology method. The indexes of sovereign medicinal were calculated, and their characteristics of nature, flavors and meridian entry were analyzed. The degree of coincidence between identified sovereign medicinal in the paper and those affirmed by ancient physicians was compared. Results The identification of sovereign medicinal in 88 decocting formulas was carried out in Shanghanlun, and among them there were 79 formulas with 1 sovereign medicinal and 9 with 2, therefore there were 97 sovereign medicinal totally. After removing repeated ones, there were 31 sovereign medicinal. The statistics of indexes of sovereign medicinal showed that the indexe of Douchi (Semen Sojae Praeparatum, 0.80) was the highest, and indexes of Chishizhi (Halloysitum Rubrum), Mahuang (Herba Ephedrae), Chaihu (Radix Bupleuri) and Banxia (Rhizoma Pinelliae) were secondly high. The coincidence rate was 76.9% between identified sovereign medicinal in the paper and those affirmed by ancient physicians. The comparison in the features between sovereign medicinal and non-sovereign medicinal showed that the medicinal with warm nature were more and those with cold nature were less in sovereign medicinal than those in non-sovereign medicinal, the medicinal with pungent flavor were more and those with sweet flavor were less in sovereign medicinal than those in non-sovereign medicinal, and the medicinal entering meridians of spleen, stomach, bladder and kidney were more in sovereign medicinal than those in non-sovereign medicinal. Conclusion The identification of sovereign medicinal by using formula metrology method can provide scientific basis for studying and applying formulas in Shanghanlun.

    Knowledge discovery of formula classification according to therapies in Shanghanlun based on representation method of multi-layer complex concept network

    With the concepts of formulas, medicinal and formula-medicinal relationship in formula database of Shanghanlun, a formal context was created based on mathematical formal concept analysis. The formal context was then given object sub-block division and optimization according to the classification method (harmonizing formulas, warming formulas and sweating formulas) in Shanghangutongfang written by WANG Zi-jie. The multi-layer complex concept network diagrams of corresponding group structure of formula compatibility classified according to therapies in Shanghanlun were established by using generation method of complex concept network. These multi-layer complex concept network diagrams expressed and explained visually the relational structure between corresponding formulas (objects) and medicinal (properties). The research is a new attempt to integrate information science and traditional TCM study of cold damage, which will bring great benefit in relearning traditional classification methods and improve the inheritance and innovation of academic thought in Shanghanlun.

    Influence of Fufang Zhebei Granules combining different chemotherapy regimens on clinical effect of refractory acute leukemia

    Objective To observe the influence of Fufang Zhebei Granules combining different chemotherapy regimens on clinical effect of refractory acute leukemia (RAL). Methods The patients (n=238) diagnosed as RAL patients were randomly divided into control group and treatment group by applying randomized double-blind, placebo-controlled and multicenter clinical trial and central randomization system (online). After the trial finished, 3 patients who were not eligible to the inclusion criteria were excluded, and then treatment group (n=118, treated with Fufang Zhebei Granules combining different chemotherapy regimens) and control group (n=117, treated with placebo combining different chemotherapy regimens) were determined after double unblinding. The curative effect was reviewed according to efficacy criterion after one chemotherapy cycle. Results The rate of complete remission (CR) was 32.35% in treatment group and 23.08% in control group. The test of Z/Cmh showed P=0.0444. The comparison in rate of CR in one chemotherapy cycle between Fufang Zhebei Granules combining different chemotherapy regimens and standard chemotherapies reported by Chinese and foreign literature including CAG, MAE, FLAG and CsA showed that Fufang Zhebei Granules combining different chemotherapy regimens promoted the rate of CR, and the curative effects of Fufang Zhebei Granules combining CAG or MAE were the best. Conclusion Fufang Zhebei Granules combining CAG or MAE can significantly promote the rate of CR of RAL in one chemotherapy cycle.

    Influences of Xuezhikang Capsules on blood fat level and vascular endothelial function in CHD patients with different TCM constitutional types

    Objective To definite the lipid-lowering effect of Xuezhikang Capsules, and discuss the influence of TCM constitutional factors on the curative effect of Xuezhikang Capsules. Methods The patients with coronary heart disease (CHD) and 4 constitutional types (n=50) were randomly selected as treatment group, 50 patients with CHD were selected as control group, and other 50 health volunteers were selected as health group. The control group was treated with routine therapy, and treatment group was additionally given Xuezhikang Capsules for 12 w. The changes of levels of endothelin-1 (ET-1), nitric oxide (NO) and blood fat were observed before and after treatment. Results All lipid indexes had significant difference in blood stasis constitution group and phlegm dampness constitution group after treatment compared with those before treatment and those in control group after treatment (P<0.05). The level of LDL-C had significant difference in qi deficient constitution group after treatment compared with those before treatment and those in control group after treatment (P<0.05). The indexes of vascular endothelial function had significant difference in blood stasis constitution group and phlegm dampness constitution group after treatment compared with those before treatment and those in control group after treatment (P<0.05). The curative effects on angina pectoral and electrocardiogram were better in blood stasis constitution group and phlegm dampness constitution group than those in qi deficient constitution group and yang deficient constitution group (P<0.05). Conclusion The overall curative effect and improving effects of Xuezhikang Capsules on blood fat and vascular endothelial function are better in CHD patients with blood stasis constitution or phlegm dampness constitution than those in CHD patients with qi deficient constitution or yang deficient constitution.

    TCM syndromes and syndrome factors of nodular goiter

    Objective To study the laws of TCM syndromes of nodular goiter through epidemiological survey. Methods The cross-sectional method of epidemiological survey was applied for analyzing the distribution laws of TCM syndromes and syndrome factors in 236 patients with nodular goiter who were eligible to internalized standard. The correlation between syndrome types and age, course of disease, outcomes of thyroid ultrasound and thyroid function was studied. Results The syndrome of liver depression and qi stagnation had the highest percentage (40.3%) and second ones were syndrome of binding of phlegm and stasis (27.5%) and syndrome of liver depression and spleen deficiency (17.4%). The syndrome factors related to disease locations were mainly liver, spleen and kidney, and those related to pathogenesis were qi stagnation, phlegm coagulation and blood stasis. The difference in the diameter of the largest thyroid nodule had no significance among different groups (P<0.05). The index of thyroid function-TSH level was lower in group of dual deficiency of qi and yin compared with other syndrome types (P<0.05). The difference in age had statistical significance among different groups (P<0.05), and age of liver depression and kidney deficiency group was the eldest and that of liver depression and qi stagnation group was younger. The course of disease was longer in liver depression and kidney deficiency group than that in liver depression and qi stagnation group and binding of phlegm and stasis group (P<0.05). Conclusion The most common syndrome types of nodular goiter are syndromes of liver depression and qi stagnation, binding of phlegm and stasis and liver depression and spleen deficiency, and its important pathogeneses are qi stagnation, phlegm coagulation and blood stasis.

    The authors, after proofreading word for word the Part of Pulse of Song-edition Qianjinyaofang and Xindiaosunzhenrenqianjinfang, found out that the scholars in the Song Dynasty adjusted and modified in a large scale the literature of pulse theory in original edition of Qianjinyaofang. There were 3 characteristics summed up as follows: modification and supplement of titles and readjustment of arrangement order made the academic frame of pulse theory more rigorous, recognition to pulse theory was constantly deepened in the Song Dynasty, and guiding function and practicability of pulse theory in clinic were improved. The conclusion of this paper will provide some basis for investigating the changes of pulse theory at the junction between the Tang and Song Dynasties, exploring the principle and methods of adjusting ancient TCM classics by Song scholars, and reevaluating the status of Song scholars in Chinese medical history.

    Discrimination of original plants of Jungui and Mugui

    Objective The original plants of Jungui and Mugui recorded in Shennongbencaojing had been confused in ancient times,and in moder times, they were regarded through textual research as Rougui (Cinnamomum cassia), a widely cultivated plant belonging to Lauraceae family, and Qinmu was regarded as a highly poisonous plant-Japanese andromeda (Pieris japonica) belonging to Ericaceae family. These conclusions are erroneous, especially the latter. To conduct a textual research on their original plants, Jungui and Mugui were discriminated from morphological features, naming basis, producing regions, habitats, applications and efficacies. It is deemed that in ancient times, Jungui and Gui were the same plant, that is today’s Chuangui (C. wilsonii), Shaohuagui (C.pauciflorum) and Yehuanggui (C.jensenianum) in Laurace family. Mugui and Qin, Qinmu, Yangui and Guihua refer to today's Muxi (Osmanthus fragrans) in Oleaceae family.

    Quality reviewing of Gaoben (Rhizoma Ligustici) based on drug system

    Objective To establish the quality representation of Gaoben (Rhizoma Ligustici) and correlation analysis method based on drug system, and provide a new mode for quality evaluation of Chinese medicinal. Methods HPLC method was used for simultaneous determination of 5 effective index components in 7 batches of Gaoben decocting pieces, including ferulic acid, senkyunolide A, senkyunolide H, senkyunolide I, and ligustilide, and then a correlation analysis was carried out;The quality representation based on the content relative ratio of key effective index components showed as follows: taking the content of senkyunolide I as reference, the quality representations of decocting pieces were determined based on the content relative ratio of effective index components, and using batch 4 (decocting pieces with identified therapeutic effect) as baseline for getting the difference between these relative ratio of effective index components, and its sum of absolute value represented as non-correlation coefficient. Results The quality representation based on the content of effective index components showed as follows: by the content of ferulic acid, the sorting was batch 7>batch 6>batch 4>batch 3>batch 1>batch 5>batch 2 and the maximum content difference was about 2.2 times; by the content of senkyunolide I, the sorting was batch 7>batch 4>batch 3>batch 6>batch 1>batch 5>batch 2 and the maximum content difference was about 4.9 times; by the total content of phthalides, the sorting was batch 7>batch 3>batch 6>batch 4>batch 1>batch 2>batch 5 and the maximum content difference was about 3.6 times. The total content of effective index components was higher in batch 7, batch 3, batch 4 and batch 6. The sorting of the correlation degree ( δ/correlation degree) was analyzed as batch 4 (0/100%)>batch 7 (1.21/79.8%)>batch 3 (1.36/77.3%)>batch 6 (1.83/69.5%)>batch 5 (2.18/63.7%)>batch 1 (2.28/62.0%)>batch 2 (9.14). The results indicated that the correlation degree was higher between the quality representations of batch 7 or batch 3 and that of batch 4. Conclusion The analysis on the correlation degree of quality representation is conducted based on the content of effective index components and content relative ratio and taking decocting pieces with identified therapeutic effects as baseline for evaluating the overall correlation of quality and the effectiveness of application.

    Objective To investigate the effective mechanism of Danzhi Xiaoyao San to generalized anxiety disorder. Methods The influence of Danzhi Xiaoyao San on the ethology in rat model of stress anxiety in Vogel's drinking conflict test combining predator exposing test. All 30 rats were randomly divided into normal group, model group and Chinese medicinal group. The rat model of stress anxiety was established by using Vogel's drinking conflict test combining predator exposing test in model group and Chinese medicinal group for 20 d. After the last time of stress finished and 24-h medicinal intervention, the test results of open field test (OFT) and elevated plus maze (EPM) were compared among all groups. Results After the last time of combined stress for 24 h, the times of horizontal movement and vertical movement, and percentage of open arm entry (OE%) and percentage of open arm time (OT%) decreased in model group compared with normal group. The times of horizontal movement and vertical movement and OE% and OT% increased in Chinese medicinal group compared with model group. All the above results had significant difference except of OT%. Conclusion Danzhi Xiaoyao San has a significant anti-anxiety effect in rat model of stress anxiety in Vogel's drinking conflict test combining predator exposing test.

    Influences of antagonistic medicianl combination of Haizao (Sargassum) and Gancao (Radix Glycyrrhizae) in Haizao Yuhu Tang on functions of liver and kidney in rats

    Objective To investigate the influences of antagonistic medicianl combination of Haizao (Sargassum) and Gancao (Radix Glycyrrhizae) in Haizao Yuhu Tang on functions of liver and kidney in rats. Method All Wistar rats were randomly divided into blank group, model group ,positive drug group, whole formula group, non-Haizao group, non-Gancao group and non-Haizao non-Gancao group, and except of normal group, other groups were orally given propylthiouracil for establishing goiter model. Taking Euthyrox 0.02 mg/kg as a positive control drug, and all groups were given corresponding medicinal 14 g/kg once a day for 28 d. The coefficients of liver and kidney and serum indexes related to liver and kidney functions were detected in rats of all groups. Results The coefficients of liver was significantly lower in model group and all Chinese medicinal groups than that in normal group and positive drug group (P<0.05). The levels of ALT, AST and ALP were significantly higher in model group than that in normal group, and trended to normal in positive drug group, non-Haizao group, non-Gancao group and non-Haizao non-Gancao group. Compared with normal group and model group, the level of albumin (ALB) increased in non-Haizao group and decreased in non-Haizao non-Gancao group (P<0.05). The value of globulin (GLB) was significantly lower in non-Gancao group than that in normal group and model group (P<0.05), and value of total protein (TP) was lower in model group, non-Gancao group and non-Haizao non-Gancao group than that in normal group (P<0.05). The level of blood urea nitrogen (BUN) was higher in model group, whole formula group, non-Haizao group and non-Haizao non-Gancao group than that in normal group (P<0.05). The level of uric acid (UA) was lower in non-Gancao group and non-Haizao non-Gancao group than that in model group (P<0.05). The level of serum creatinine (Scr) had no significant difference among all groups. Conclusion The antagonistic medicianl combination of Haizao and Gancao in Haizao Yuhu Tang has no significant effects on liver and kidney functions in rats.

    Influence of Huiyang Shengji Formula on wound healing in rats with diabetic skin ulcer

    Objective To investigate the influences of Huiyang Shengji Formula and its disassembled formulas on wound healing in rats with diabetic skin ulcer. Methods SD rats (n=56) were randomly divided into control group, model group, whole formula group (treated with whole Huiyang Shengji Formula), wenyang group (treated with disassembled formula of warming yang and tonifying qi) and huoxue group (treated with disassembled formula of activating blood and promoting tissue regeneration). The method of intraperitoneal injection of STZ-hormone intervention-skin defect-embedding of plastic ring was used for establishing model of diabetic skin ulcer. All groups were given petrolatum gauzes or corresponding medicated gauzes, and after 3 d, 7 d, 10 d and 14 d, wound healing rate was calculated and wound morphologic changes were observed. After 14 d, the wound granulation tissue was collected from all groups and given HE staining and Masson staining for observing the structure changes of granulation tissue and collagen distribution. Results The wound healing rate was lower in model group than that in control group (P<0.01) after 3 d, 7 d, 10 d and 14 d. After 3 d, wound healing rate was higher in wenyang group than that in model group, whole formula group and huoxue group (P<0.05). After 7 d, 10 d and 14 d wound healing rate was higher in all medicinal groups than that in model group (P<0.05). After 7 d and 14 d, wound healing rate was higher in whole formula group than that in model group, wenyang group and huoxue group (P<0.05). The granulation tissue grew vigorously, fibrocytes and capillaries were abundant, and inflammatory cells were less in all medicinal groups. Conclusion Huiyang Shengji Formula can effectively improve wound healing in rats with diabetic skin ulcer. The effect of disassembled formula of warming yang and tonifying qi is significant at early stage of ulcer healing and effect of disassembled formula of activating blood and promoting tissue regeneration is better at late stage.

    Influences of Yangjing Capsules on proliferation, apoptosis and caspase-3 expression of GC-1 cells

    Objective To discuss the possible mechanism of Yangjing Capsules in improving spermatogenic function and then treating male infertility. Methods The extracting solution of Yangjing Capsules in different doses were added into GC-1 cells and the cells were divided into blank group, low-dose group, mid-dose group and high-dose group. The absorbance values in all groups were detected by using methyl thiazolyl tetrazolium test (MTT) respectively after 24 h, 48 h and 72 h. The cell cycle and apoptosis were examined by using flow cytometer after 48 h, and the expression of caspase-3 (apoptosis-related gene) was detected by using real-time fluorescence quantitative polymerase chain reaction (RT-PCR) and Western blot method. Results After 24 h, 48 h and 72 h, the proliferation of GC-1 cells was speeded up significantly in low-dose group, mid-dose group and high-dose group, and the comparison in absorbance had a statistical significance between blank group and medicated groups (P<0.01). After 48 h, the proportion of S phase cells increased gradually (P<0.05) and apoptosis rate decreased significantly (P<0.01) in low-dose group, mid-dose group and high-dose group and had statistical significance compared with blank group. The expression of caspase-3 mRNA showed a decreasing tendency with a statistical significance compared between mid-dose group or high-dose group and blank group (P<0.01), and expression of caspase-3 protein showed a decreasing tendency with a statistical significance compared between high-dose group and blank group (P<0.01). Conclusion Yangjing Capsules has the effects of improving the proliferation of GC-1 cells, inhibiting apoptosis and reducing caspase-3 expression in a dose-dependent manner.
