    Ancient Chinese literature has recorded theoretical reasoning and medical cases on health preservation and treatment of diseases using mental image. There are two ways of health preservation by mental image: restrictive Guan (intuitive insight) and non-restrictive Guan. The article discusses origin, methods, application, and mechanism of cultivating health and preserving life by mental image.

    Effects of paeoniflorin on histomorphology and NO pathway in hippocampus CA3 Zone in a rat model of blood deficiency and liver depression pattern*

    Objective To investigate the effects of paeoniflorin on histomorphology, Glu and NO levels in hippocampus, and on protein expressions of iNOS in the hippocampus CA3 Zone in a rat model of blood deficiency and liver depression pattern. Methods The rat model of blood deficiency and liver depression pattern was established with chronic restraint stress in conjunction with radiation. Fifty-six male SD rats were randomly divided into model group, high-dose, medium-dose, and low-dose paeoniflorin groups, as well as the blank control group of healthy SD rats(n=14 in each group). SD rats were treated with intragastric administration of paeoniflorin in the high-dose, medium-dose, and low-dose paeoniflorin groups while pure water was intragastrically administered in the model and blank groups for 21 days. Histopathological changes of hippocampus CA3 Zone were observed by using HE staining methods, and Glu and NO levels in hippocampus tissues was measured by using HPLC and the Griess methods, respectively, and protein expression of iNOS in hippocampus CA3 zone was detected with immunohistochemistry. Results Compared with the blank group, significant changes were observed in the neuron cells in the model group, including cell body shrinkage, deeply stained chromatin nuclei, triangular nuclear shrinkage and no remarkable nucleoli. There was no significant changes in paeoniflorin group. The levels of Glu and NO increased significantly in model group compared with normal group (P<0.05), and decreased significantly in paeoniflorin group compared with model group (P<0.05). The protein expression of iNOS in hippocampus CA3 zone increased significantly in the model group compared with the blank control group (P<0.05), and decreased significantly in paeoniflorin group compared with the model group (P<0.05). Conclusion Pathological changes including neuron shrinkage were present in hippocampus CA3 Zone in the model of blood deficiency and liver depression pattern. Paeoniflorin could improve hippocampus functional structural damage in the rat model of blood deficiency and liver depression pattern, which may be related to decreased levels of Glu and NO and protein expression of iNOS in hippocampus CA3 Zone.

    Effects of blood-cooling and blood-activating medicinal on IL-23/IL-17 axis in psoriasiform mice model induced by imiquimod*

    Objective To observe the effects of blood-cooling and blood-activating medicinal (Liang Xue Huo Xue Yao) on IL-23/IL-17 axis in psoriasiform mice model induced by imiquimod. Methods BALB/c male mice were randomly divided into control group, model group, and three blood-cooling and blood-activating medicinal groups of high, medium and low dose (LXHX-H, LXHX-M and LXHX-L groups) respectively. Pathological changes of skin lesions in mice were observed by using light microscope. The levels, mRNA and protein expression of the cytokines in the serum and skin lesions of mice were tested by using ELISA, RT-PCR, and western blotting assay respectively. Results Compared with model group, LXHX-H, LXHX-M and LXHX-L groups decreased the emergence, color change and infiltration of skin erythema, as well as the PASI scores, epidermal parakeratosis and epidermal over-proliferation of the mice. According to the trend graph of the PASI scores, the area under the curve of LXHX-M group was significantly lower than that in model group. HE staining showed that, compared with model group, mice in LXHX groups had more smooth epidermis cortex, less epidermal parakeratosis and decreased epidermal over-proliferation(P<0.01). Lesions protein and mRNA test showed that compared with model group, mice in LXHX groups had lower expression level of IL-17 and IL-23 protein, as well as IL-23mRNA, IL-22mRNA, and RORγt mRNA(P<0.01 or P<0.05). Conclusion LXHX medicinal could significantly decrease the intensity of the psoriasiform inflammation and the formation of psoriasiform skin lesions induced by imiquimod, the effect of which might be related to the decrease of IL-23/IL-17 axis cytokine protein and the expression of mRNA, as well as the inhibition of the expression of Th17 cell differentiation transcription factor RORγt mRNA.

    Effects of Yiqi Huayu Jiedu method on EGFR/MAPK signal pathway in rats with chronic atrophic gastritis exhibiting dysplasia*

    Objective To observe the effects of Yiqi Huayu Jiedu method on EGFR/MAPK signal pathway in rats with chronic atrophic gastritis exhibiting dysplasia. Methods 60 Wistar rats were randomly divided into model group (n=50) and blank control group(n=10). The blank control group rats were given standard SPF animal diet throughout the study. Three measures were combined to establish model of chronic atrophic gastritis with dysplasia, including intragastric administration of 120 μg/mL MNNG, 5 mL/kg, 1 time/d, assisted by 0.1% ammonia drinking water and 0.03% ranitidine in feed. After the model was successfully established at the 28th week, 30 modeled rats were randomly divided into model group, vitacoenzyme group and Yiqi Huayu Jiedu group, with 10 rats in each group. Each group was given the corresponding medicine once daily: 3 mL/kg saline for the model group, 2 mL/kg vitacoenzyme suspension liquid (0.3 g/kg vitacoenzyme) for the vitacoenzyme group and 3 mL/kg herbal solution (containing 9 g/kg crude herb). All the rats were sacrificed after 12 weeks. The method of Western blot was used for measuring the protein expression of EGFR,ERK1/2, and p-ERK1/2 in rats of all groups. Results The EGFR protein expression of the model group was higher than that in the vitacoenzyme group, the Yiqi Huayu Jiedu group, and the blank group (P=0.000); it was significantly lower in Yiqi Huayu Jiedu group than the model group (P=0.000) and also lower than vitacoenzyme group (P<0.05). Compared with the blank control group, there was no difference (P>0.05). The ERK1/2 protein expression of the model group was higher than that in the vitacoenzyme group, the Yiqi Huayu Jiedu group, and the blank group (P=0.000) in a descending order; it was significantly lower in Yiqi Huayu Jiedu group than the model group and vitacoenzyme group (both P=0.000) and significantly higher than the blank group (P<0.01). The p-ERK1/2 protein expression of the model group was significantly higher than that in the blank group (P=0.000); it was significantly lower in Yiqi Huayu Jiedu group than the model group and vitacoenzyme group (both P=0.000). Compared with the blank group, there was no significant difference (P>0.05). Conclusion Yiqi Huayu Jiedu method could reverse gastric mucosal dysplasia. Its mechanism of action could be reducing high expression of EGFR protein and blocking abnormal activation of the EGFR/MAPK cellular signal transduction pathway.

    Effects of Danzhi Xiaoyao San on cell apoptosis of Papez’s circuit in rat model of Generalized Anxiety Disease (GAD)*

    Objective To study the effects of Danzhi Xiaoyao San on cell apoptosis of Papez’s circuit in rat model of Generalized Anxiety Disease (GAD). Methods Male Wistar rats were divided into the normal group, model group, Chinese Medicine group, and control group of western medicine; each group contained 10 rats. The GAD model was established by using randomly assigned empty drinking bottle to upset the rats. Then the Chinese medicine group was intragastrically given Danzhi Xiaoyan San, while the western medicine group was given buspirone hydrochloride, and the model group was given distilled water once a day for a week. Then morphological changes of hippocampus and amygdaloid nucleus were observed by using light and electron microscopes, and the early apoptosis rate of hippocampus and mamillary body were evaluated by applying flow cytometry assays after removing rat brain. Results Cells in hippocampus and amygdaloid nucleus of model group showed typical morphological changes of apoptosis such as margination and karyopyknosis. Danzhi Xiaoyao San and buspirone hydrochloride could revert the changes markedly. Flow cytometry assay showed that the early apoptosis rates in hippocampus and mamillary body of model group increased significantly compared with normal group (P<0.01). Compared with model group, the apoptosis rates of Chinese medicine group and Western medicine group decreased (P<0.05 or P<0.01). Conclusion There is extensive and obvious apoptosis in Papez’s circuit of GAD rat’s brain. Danzhi Xiaoyao San can prevent and inhibit apoptosis to some extent.

    Effects of Blood Mansion Stasis-Expelling Decoction combined with Two Matured Substances Decoction on adipocytokines in obese rats*

    Objective To investigate the effects of Blood Mansion Stasis-Expelling Decoction (Xuefu Zhuyu Tang) in combination with Two Matured Substances Decoction (Er Chen Tang) on adipocytokines in subcutaneous adipose tissues, visceral adipose tissues and serum of obese rats. Methods Nutritional obesity rat models were established with high-fat feeding, and they were randomly divided into four groups according to body mass, namely, Blood Mansion Stasis-Expelling Decoction group, Yang-Supplementing and Five-Returning Decoction group (Buyang Huanwu Tang), Blood Mansion Stasis-Expelling Decoction in combination with Two Matured Substances Decoction group, model group plus blank control group (n=10). The first three groups were administered with Blood Mansion Stasis-Expelling Decoction, Yang-Supplementing and Five-Returning Decoction and Blood Mansion Stasis-Expelling Decoction in combination with Two Matured Substances Decoction, respectively at 10 times the adult dosage. The latter two groups were fed with saline. All the groups were intervened for 4 consecutive weeks. The levels of subcutaneous adipose tissue, visceral adipose tissue, serum of visceral fat element, adiponectin, leptin, fibrinolytic enzyme activators inhibitor-1, tumor necrosis factor alpha, and endothelin-1 (plasma) were measured by using Elisa method. Results ①The visfatin levels in subcutaneous fat, visceral fat and serum were increased in the model group, which were lowered in the Blood Mansion Stasis-Expelling Decoction group and the Blood Mansion Stasis-Expelling Decoction in combination with Two Matured Substances Decoction group compared with the model group (P<0.05), more significant in the latter (P<0.05). ②The contents of subcutaneous fat, visceral fat and serum adiponectin were reduced in the model group,while the contents of subcutaneous adipose tissue and serum adiponectin in every blood-activating group were increased compared with that of model group (P<0.05), more significant in Yang-Supplementing and Five-Returning Decoction group (P<0.05). Moreover, the level of adiponectin in visceral adipose tissue in Yang-Supplementing and Five-Returning Decoction group was also higher than that in model group(P<0.05). ③The level of leptin was decreased in subcutaneous fat and increased in visceral fat and in serum in model group(P<0.05). Serum leptin levels were lowered in three blood-activating groups. Moreover, leptin in visceral adipose tissue was also lowered in Blood Mansion Stasis-Expelling Decoction in combination with Two Matured Substances Decoction group (P<0.05).④The level of TNF alpha in visceral fat was reduced (P<0.05), No significant difference was observed in serum concentration and content of TNF alpha in subcutaneous fat between groups(P>0.05). Compared with model group, levels of TNF alpha in visceral fat were elevated in Yang-Supplementing and Five-Returning Decoction group and Blood Mansion Stasis-Expelling Decoction in combination with Two Matured Substances Decoction group (P<0.05). ⑤No statistically significant differences were shown in endothelin-1 in subcutaneous fat, visceral fat and plasma (P>0.05). ⑥The levels of PAI-1 in visceral fat and subcutaneous fat were increased in model group (P<0.05). However, PAI-1 level in visceral fat was reduced in Blood Mansion Stasis-Expelling Decoction group, and Blood Mansion Stasis-Expelling Decoction in combination with Two Matured Substances Decoction group (P<0.05). In addition, the level of PAI-1 in subcutaneous fat tissues was also lowered (P<0.05). Conclusion There was a significant endocrinological difference between visceral and subcutaneous adipose tissue. For model rats, formulas that can activate the blood and resolve blood stasis have therapeutical effects on disorder of adipocytokines.

    Effects of heat-clearing and blood-cooling herbs on psoriatic mice and their mechanism of action*

    Objective To investigate the effects of three heat-clearing and blood-cooling traditional Chinese herbal medicine (Shengdi, Zicao, Danpi) on mouse models of psoriasis, including estrogen-induced vaginal epidermis and the mouse-tails’ granular flakes, and to explore their mechanism of action. Methods The induced vaginal epidermis mouse models were established by feeding the mouse with estrogen to facilitate the formation of their vaginal epithelia. Whereas the mouse-tails’ models were built by using mouse tail that were born without stratum granulosum. The researcher gave three different heat-clearing and blood-cooling traditional Chinese medicinals to mouse models for 2 weeks at different dosages(1.93,3.86,7.72 g/kg), and then counted the mitotic index of the mouse’s vaginal epithelia and the number of mouse-tails’ granular flakes. Results Compared with the control groups, each dose of three medicinals inhibited mouse vaginal epithelial mitosis (P<0.05); they also facilitated the formation of the granular layers in the flakes of the mouse-tails (P<0.05), but the there were no marked differences in the effects among these three medicinals and there was no direct dose-effect relation in each medicinal. Conclusion Three heat-clearing and blood-cooling traditional Chinese medicine (Shengdi, Zicao, Danpi) could help alleviate psoriasis through inhibiting mouse vaginal epithelial mitosis and facilitating the formation of the granular layers in the flakes of the mouse-tails.

    Effects of saikosaponina on the behaviors and expressions of Caspase-3 and Caspase-9 in hippocampus of rats with depression*

    Objective To investigate the effects of saikosaponina on the behaviors and expressions of Caspase-3 and Caspase-9 in hippocampus of rats with depression. Methods SD rats (2-3 months old, n=60) were randomly divided into control group, model group, saikosaponina group, and fluoxetine group (n=15 in each group). The model of depression was established by using chronic unpredictable mild stress (CUMS) combined with isolated rearing. The fluoxetine group and saikosaponina group were given intragastric administration of fluoxetine at 1.2 mg/Kg or saikosaponina at 25 mg/Kg once daily from the second day of the establishment of the model. The sucrose consumption experiment and open field test were carried out to assess rat ethological changes on the 21st day. The protein and mRNA expressions of Caspase-3 and Caspase-9 in rat hippocampus were detected by using immunohistochemical technique and RT-PCR. Results Ethological parameters decreased significantly in model group (P<0.05) at Day 21 after the establishment of the model, compared with control group. Compared with model group, they were significantly improved in fluoxetine and saikosaponina group (P<0.05). The results of immunohistochemistry and PCR test showed that the expressions of Caspase-3, Caspase-9 protein, and mRNA increased in model group compared with control group (P<0.05), and decreased in fluoxetine group and saikosaponina group (P<0.05). Conclusion Saikosaponina can effectively relieve the symptoms of depression in rats, and its mechanism may be related to the decrease of the expressions of Caspase-3 and Caspase-9 in hippocampus.

    Qingdu Huoxue Huatan Compound inhibits the expression of nf-kappa B and STAT3 in umbilical vein endothelial cell line ECV304 OGD/R model*

    Objective To study the protective effects of Qingdu Huoxue Huatan (Toxin-removing blood-activating and phlegm-resolving) compound against OGD/R damage to umbilical vein endothelial cell and its underlying mechanism. Methods SD rats were intragastrically given Qingdu Huoxue Huatan compound (12 g/kg) while the control group received normal saline of the same dosage. Sera of rats in two groups were extracted afterwards. Drug serum of Qindu Huoxue Huatan compound was added to the OGD/R-damaged umbilical vein endothelial cell line ECV304 in the process of regainning oxygen and sugar, and cell survival rate was evaluated by using MTT and mRNA; protein levels of NF-κB and STAT3 were measured by using rt-PCR and western blotting. Results After 24h, Qindu Huoxue Huatan compound drug serum promoted ECV304 proliferation, and the promotion reached the peak at 48 h. Inhibition of mRNA and protein of NF-κB and STAT3 was observed at each time period. Conclusion Qindu Huoxue Huatan compound drug serum could effectively improve OGD/R damage to umbilical vein endothelial cell line ECV304, and promote cell’s proliferation. Its mechanism may be related to the inhibited expressions of nf-kappa B and STAT3.

    Synthesis and immunogenicity identification of artificial antigen of Saikosaponin a*

    Objective To establish an enzyme linked immunosorbent assay (ELISA) for Saikosaponin a on the basis of the synthesis of immunizing antigen and coating antigen of Saikosaponin. Method The method of Sodium Permanganate Oxidation was used to synthesize immunizing antigen (SSa-BSA) and coating antigen (SSa-PLL) of Saikosaponin a separately; the success of the synthesis of artificial antigens was examined by using UV spectrometry and TLC (thin layer chromatography) method. BALB/c mice were immunized with the prepared SSa-BSA immunogenic antigen. The titer and specificity of the anti-serum were detected with ELISA and indirect competitive ELISA(ic-ELISA) respectively. Results The results of TLC showed that Saikosaponin a was successfully conjugated with BSA and PLL. After immunized with SSa-BSA, the mice can produce specific anti-saikosaponina antibodies, whose titer was up to 1:80000. Conclusion The research results showed that TLC had a unique advantage over other artificial antigen identification methods. Artificial antigen of Saikosaponin a was successfully synthesized, which could be applied to the preparation of monoclonal antibodies, as well as the establishment of corresponding immune method.

    Research on dynamic changes of five chemical components of Ziziphora clinopodioides Lam. at different growth stages using HPLC*

    Objective To measure the dynamic changes of five chemical components of Ziziphora clinopodioides Lam. at different growth stages, and to determine the optimal harvesting time. Methods The Ziziphora clinopodioides Lam. was extracted from 20 % dimethyl sulfoxide-methanol solution. The method of High Performance Liquid Chromatography (HPLC) was used, with the 0.3% mobile phase of formic acid-methanol -0.3% formic acid on a gradient elution, to detect the Caffeic acid, Rosemary acid, Diosmin, Linarin and Pulegone in different growth periods of Ziziphora clinopodioides Lam. at 290 nm wave length at the same time. Results With these chromatographic parameters, the linear relationship, degree of separation, precision, stability, repeatability and recovery rate of the five components were all qualified. The contents of Linarin and Diosmin were the highest, with Pulegone second to it while the Caffeic acid and Rosemary acid’s contents were lower at different growth periods. And these five chemical components in Ziziphora clinopodioides Lam. varied at different growth periods. But all of them were higher in pre-flowering period and flowering period. Conclusion According to the data, we can determine the flowering period was the optimal harvesting time for Ziziphora clinopodioides Lam. initially. The pattern of dynamic changes of the chemical components of Ziziphora clinopodioides Lam. could provide the basic data for the Good Agriculture Practice (GAP) of Ziziphora clinopodioides Lam.

    Protective effects of kidney-tonifying liver-soothing medicinals on visual function of early glaucoma: a clinical trial*

    Objective To investigate the protective effects of kidney-tonifying liver-soothing medicinals on visual function of early glaucoma patients with liver depression and qi stagnation. Methods 80 patients (136 eyes) with early glaucoma of liver depression and qi stagnation pattern were recruited from out-patient clinic, whose intraocular pressure had been controlled down to target intraocular pressure (IOP) for 3 months from 2010 to 2013.The participants were randomly divided into either the treatment group or the control group with 40 cases in each. Treatment group was given kidney-tonifying liver-soothing medicinals for 3 months, with glaucoma-related knowledge education, while the control group was treated with cobamamide tablets. The traditional Chinese medicine (TCM) patterns, the best corrected visual acuity, intraocular pressure, visual field, and visual quality scores were evaluated before and after treatment. Curative rates were compared between the two groups after treatment at 3 months and 6 months respectively. Results Visual field, TCM pattern, and the mean visual quality scores improved significantly after treatment in kidney-tonifying liver-soothing medicinals group, compared with cobamamide group. Curative effect of liver-soothing depression-relieving medicinals was superior to cobamamide tablets at 3 months (P<0.05), yet this difference failed to be significant at 6 months (P>0.05). Conclusion Kidney-tonifying liver-soothing medicinals seems to be superior to cobamamide in protecting visual function and visual quality of life in patients with early glaucoma. There was no significant difference between kidney-tonifying liver-soothing medicinals and cobamamide tablets in the treatment of early glaucoma patients with liver depression and qi stagnation after 6 months.

    Objective To investigate the efficacy of fitness qigong Ba Duan Jin in pulmonary rehabilitation of patients with chronic obstructive pulmonary disease (COPD) at stable stage. Methods The patients who met the diagnosis criteria were randomly divided into Ba Duan Jin group (n=31) and control group (n=28). Patients in the control group received conventional pharmacotherapy, while patients in the Ba Duan Jin group practiced fitness qigong Ba Duan Jin on the basis of using the conventional pharmacotherapy. The duration of the study lasted for 180 days. On the 1st day and the 180th day, COPD assessment test (CAT),lung function test, and acute exacerbation frequency test were performed. Results Fitness qigong Ba Duan Jin significantly improved the sum of CAT scores and scores of single symptoms(P<0.05); Fitness qigong Ba Duan Jin seemed to increase FVC of grade A patients and slow down the descending rate of FEV1 and FEV1/FVC of grade B patients(P<0.05). Conclusion On the basis of using the conventional pharmacotherapy, fitness qigong Ba Duan Jin can relieve the symptoms of patients, increase their exercise tolerance, and improve their quality of life at the stable stage of COPD.
