    Allergic diseases and intestinal flora imbalance for allergic constitution research*

    Studies have shown that intestinal flora imbalance is closely related with allergic diseases, yet its specific mechanism remains unclear. Modern medicine considers that it may be associated with increased intestinal permeability and intestinal barrier disruption caused by dendritic cells and other immune cells. Traditional Chinese Medicine identifies three possible approaches, which are “Lungs invaded by evil passing to the large intestine”, “Spleen invaded by evil leading to restricted nutrition and disturbed microenvironment” and “Liver invaded by evil leading to stirred-up wind and disturbed microenvironment”. The root cause is that “Pathogenic factor invades when Zheng Qi is deficient”. This paper explores what intestinal flora research can bring to the allergic constitution study by combining the four fundamental theories “Life course theory”, “Composition of form and spirit”, “Environmental constraint theory”, and “Theory of innate endowment”.

    Behavioral changes in rats of liver-stagnation pattern, spleen-deficiency pattern and liver-stagnation with spleen-deficiency pattern and effects of Chaihu Sijunzi Decoction on different patterns*

    Objective To compare the behavioral changes of different rat models of liver-stagnation pattern, spleen-deficiency pattern, and liver-stagnation and spleen-deficiency pattern and to explore the effects of Chaihu Sijunzi Decoction (Bupleurum with Four Gentlemen Decoction) on those rat models. Methods Male Wistar rats were randomly assigned into the normal group (group 1), liver-stagnation group (group 2), spleen-deficiency group (group 3), liver-stagnation with spleen-deficiency group (group 4), liver-stagnation intervention group (group 5), spleen-deficiency intervention group (group 6) and liver-stagnation with spleen-deficiency intervention group (group 7) with thirteen rats in each group. The models were established by using chronic restraint method in group 2 and 5, by using excessive fatigue plus irregular diet method in group 3 and 6, and by using chronic restraint-excessive fatigue-irregular diet method in group 4 and 7 for 4 weeks. At the end of the 15th day, rats in the group 5, 6 and 7 were fed with Chaihu Sijunzi Decoction per 2.86 g·kg-1 for two weeks, whereas the rats in the normal group and model groups 2, 3 and 4 were fed with equivalent saline for two weeks. On the 14th and 28 th day, open-fieldtest was used to observe the crossed squares (index C), number of rearing (index N), time remaining in the central zone (index S), decorating time (index D) and the total distance of movement (index T) of all rats. Results On the 14th day, as compared with group 1, index C, index N and index T increased significantly in group 2 and 3 (P<0.05) and decreased significantly in group 4 (P<0.05). Index S increased significantly in group 2 and 4 (P<0.05) while it decreased significantly in group 3 (P<0.05). Index D decreased significantly in group 2 and 4(P<0.05) and increased significantly in group 3 (P<0.05). On the 28 th day, compared with group 1, index C, index T decreased significantly in group 2, 3 and 4 (P<0.05), while index N and index D decreased significantly and index S increased significantly in group 2 and 4 (P<0.05), which were totally opposite to group 3. Compared with group 2, index C and index T increased significantly while index S decreased significantly in group 5 (P<0.05). Compared with group 3,index C, index D, and index T increased significantly while index N decreased significantly in group 6(P<0.05). Compared with group 4, the crossed squares, number of rearing, decorating time, and index T increased significantly in group 7 (P<0.05). Conclusion The autonomic activities were at different degrees in rat models of liver-stagnation pattern, spleen-deficiency pattern, and liver-stagnation with spleen-deficiency pattern. There were decreased CNS excitement in the liver-stagnation pattern, disordered central nervous response in the spleen-deficiency pattern, and inhibited central excitatory activities in the liver-stagnation and spleen-deficiency pattern. Chaihu Sijunzi Decoction can improve the autonomic activities of those rat models, especially of the liver-stagnation with spleen-deficiency model.

    Regulatory effects of spleen-yin nourishing formula on endoplasmic reticulum stress in spleen-yin deficiency diabetic rats*

    Objective To investigate the effects of spleen-yin nourishing formula, ZiBu PiYin Recipe (ZBPYR), on endoplasmic reticulum stress (ERS) in the hypothalamus of spleen yin deficiency diabetic rats. Methods Rats were randomly divided into five groups: control (cont) group, Diabetes Mellitus (DM) group, spleen yin deficiency (pi) group, piDM group, and ZBPYR group. The expressions of p-PERK, p-eIF2α, eIF2α and GRP78, which were endoplasmic reticulum stress markers in the hypothalamus of rats, were measured using western blotting Methods. Results The protein expressions of p-PERK, p-eIF2α and GRP78 in the hypothalamus of DM group and piDM group were elevated, as compared with cont group (P<0.05). Their levles were decreased in the ZBPYR group, compared with DM group and piDM group (P<0.05). In addition, no significant difference in eIF2α expression was detected among different groups. Compared with the cont group, GRP78 mRNA expression in hypothalamus increased in the DM group, pi group and piDM group (P<0.05). The expression of GRP78 mRNA was lower in ZBPYR group than in the DM and piDM groups (P<0.05). Conclusion Endoplasmic reticulum stress seems to be present in the hypothalamus in the DM and piDM groups. Formulas that enrich and tonify the spleen yin have the effects of alleviating endoplasmic reticulum stress.

    Effects of Mongolian formula Shudage-4 on gastric ulcer*

    Objective To observe the effect of Mongolian formula Shudage-4 on alcohol-induced gastric ulcer in mice, and to discuss its protective mechanism for gastric mucosa. Methods Kunming mice were randomly divided into Shudage-4 groups of low-dose, medium-dose, and high-dose, Anweiyang (patent Chinese medicine for peptic ulcer) group, cimetidine group, model group, and normal group. Ethanol was used to induce gastric mucosal injury for all the groups except the normal group. Ulcer index and ulcer inhibition rate were measured. Concentration of nitric oxide (NO) and malonydialdehyde (MDA) were detected by using ELISA. Liver tissue sections were examined under a light microscope. Stomach tissues were stained using Hematoxylin-eosin (H-E) staining and histopathological changes were observed via light microscopy. Results NO concentration increased significantly and MDA decreased significantly in the low-, medium-, and high-dose Mongolian Medicine Shudage-4 (P<0.05) groups, as compared with model group. NO concentration in the positive group was higher than the low-dose group, but lower than the medium-dose group and high-dose group, whereas MDA level in the positive group was lower than the low-dose group, but higher than the medium-dose group and high-dose group. Since the medium-dose group was shown to be the highest in NO concentration and lowest in MDA levels among the high-, medium- and low-dose Shudage-4 groups, the medium-dose group was chosen to be compared with the positive group. However, no significant difference was present between them (P>0.05). Meanwhile, inflammatory responses were alleviated, as compared with the model group, in the high-, medium- and low-dose groups, especially in the latter two groups, with drastically reduced inflammatory cells and improved pathological conditions. Conclusion Mongolian Medicine Shudage-4 could significantly reduce the ethanol-induced gastric mucosal injury in mice, and may play a protective role.

    The mRNA expressions of ILL-22, IL-22R1, STAT3 and c-myc in skin lesions of patients with psoriasis vulgaris and their correlations*

    Objective To investigate the roles of IL-22, IL-22R1, Stat3 and c-myc mRNA expressions in abnormal proliferation of keratinocytes in psoriasis vulgaris. Methods Biopsies of skin lesions were conducted in 23 patients with psoriasis vulgaris and the skin samples of 11 normal control subjects. Levels of IL-22,IL-22R1, Stat3 and c-myc mRNA expressions were measured with reverse transcription-polymerase chain reaction (RT-PCR). Results IL-22, IL-22R1, Stat3 and c-myc mRNA were positively expressed in the psoriatic skin lesions as well as the normal controls. But the levels in the psoriatic skin lesions were significantly higher than those in the normal controls (P<0.01). There were positive correlations between the expression of IL-22 mRNA and that of IL-22R1/Stat3 mRNA (P<0.05). There were also positive correlations between the expression of Stat3 mRNA and that of c-myc mRNA (P<0.05). Conclusion The activation of Stat3 induced by IL-22 may lead to excessive proliferation of keratinocytes in psoriasis vulgaris, which could provide a new perspective in the treatment.

    Effects of Zengye Decoction on the expression of AQP3 and VIP in colon tissues of mice with slow transit constipation*

    Objective To investigate the effects of Zengye Tang (Fluid-increasing Decoction) on the expression of Aquaporin-3 (AQP3) and Vasoactine Intrestinal Peptide (VIP) in colon tissues of mice with Slow Transit Constipation (STC) and to explore the correlation between AQP3 and VIP. Methods 60 ICR mice were randomly divided into 4 groups, namely control group, model group, mosapride group and Zengye Decoction group. Each group except control group was given diphenoxylate to establish the STC model. Control group was given saline. After the establishment of the model, Zengye Decoction group was given the Zengye Decoction while mosapride group was given mosapride. Control group and model group were given saline. The expression of AQP3 and VIP in colon tissues in each group was measured by using immunohistochemistry staining. Mean density (MD) was used to analyze the correlation between AQP3 and VIP. Results Mean density of AQP3 and VIP in colon tissues in model group were markedly lower than that in control group (P<0.05). Levels of AQP3 and VIP in mosapride group and Zengye Decoction group were significantly higher than that in model group (P<0.05). AQP3 and VIP in Zengye Decoction group were not statistically different from that in mosapride group (P>0.05). The levels of AQP3 and VIP in each group were positively correlated. Conclusions Zengye Decoction can increase the expression of AQP3 and VIP in colon tissues of STC mice, which may explains the mechanism of its efficacy. The expressions of AQP3 and VIP in colon tissues were correlated, probably due to a shared signal pathway.

    Effects of the herbal pair Shanyao and Qianshi at different dosage ratios on spleen-deficiency mice’s immune function*

    Objective To compare the effects of herbal pair, Shanyao (Rhizoma Dioscoreae) and Qianshi (Semen Euryales), at different dosage ratios on spleen-deficiency mice’s weight and immunity parameters including thymus weight, proliferation of T cells as well as IL-2, IL-4 and IFN-γ, and to study the optimal dose and best ratio of this pair in terms of immunity improvement. Methods ICR mice were randomly divided into 4 experimental groups (ratios of 3∶1, 2∶1, 1∶1 and 1∶2 respectively), model group and normal group. To establish spleen-deficiency model, the experimental groups and the model group were given senna decoction by gavage, and the normal group was given equal volume of saline by gavage. Experimental groups were then given different dosages of Qianshi and Shanyao decoction by gavage, while the model group and the normal group were given equal volume of saline by gavage. Body Mass Index and thymus index were measured by weighing, T cell proliferation ability was tested by using MTT, and the serum levels of IL-2,IL-4 and IFN-γwere tested with a double-antibody sandwich ELISA method. Results After the establishment of spleen-deficiency model, mice’s weights were significantly below normal range (P<0.05), which demonstrated success of the model establishment. After the gavages of the herbal pair, associated immune indexes in all experimental groups improved at various degrees, with the most significant improvement in 3∶1 group and 2∶1 group (P<0.05), especially their serum levels of IL-2 and IL-4 as well as the thymus index. A greater degree of improvement was observed in 2∶1 group than 3∶1 group in terms of weight and T cell proliferation ability. Conclusion The herbal pair of Qianshi and Shanyao could relieve the mice’s spleen deficiency by enhancing immune function, as it could invigorate spleen and replenish Qi. The optimal dose and best ratio of Qianshi and Shanyao were expected to be 2∶1(30 g∶15 g).

    Quality representation and correlation analysis of medicinal

    Objective To determine the key factors affecting the quality of Kaixin San Heart-opening powder and the best preparation forms of Kaixin San,laying the groundwork of anti-dementia drug development from Kaixin San. Methods The content of effective index components of medicinal-preparation of Kaixin San was detected by using HPLC. The quality of medicinal preparation from Kaixin San were characterized based on the content,relative quantity,relative ratio of content, yield rate and anti-dementia effect,and the characterized Results were given correlation analysis. Results The method of the determination of effective index components from Kaixin San is easy to handle,accurate and highly reproducible. The effect of anti-dementia of three medicinal preparations from Kaixin San was listed in the following descending order of enrichment, ethanol extract, and water extract. The content of effective index components is higher with unique proportion, lower extraction rate,lower dosage and higher antibacterial effect,which is the best form in medicinal-preparation application of Kaixin San. Conclusion The key to the improvment of efficacy is to explore the fittest content and relative ratio of medicinal components based on the basic composition of Kaixin San and yield rate.

    Rapid analysis of chemical compositions in the flavonoid fraction of Mentha haplocalyx Briq. with LCMS-IT-TOF*

    Objective To rapidly analyze chemical compositions in the flavonoid fraction of Mentha haplocalyx Briq. Methods The liquid chromatography-mass spectrometry-ion trap-time-of-flight (LCMS-IT-TOF) method was applied to analyze the flavonoid fraction of Mentha haplocalyx Briq.The chromatographic separation was performed on Sunfire C18 column(4.6 mm×150 mm,5 μm)with gradient elution of methanol (A)-water (B). Multistage mass spectrometry data of different compounds was obtained by using electrospray ionization source and monitoring in positive and negative ion modes. Results Twelve flavonoids, including four flavonoid glycosides, six flavonoid aglycones, a flavonol glycoside and a flavanone glycoside, were identified based on the characteristic MS/MS fragment ions data with chemical standards and published literatures. These constituents are mainly hesperidin, rutin, apigenin-7-O-rutinoside, diosmin, luteolin-7-O-rutinoside, linarin, 5,6-dihydroxy-7,8,3′,4′-tetramethoxy-flavone, gardenin B, 5,6-dihydroxy-7,8, 4′-trimethoxy flavone, acacetin, 5-hydroxy-6,7,8,3′,4′-pentamethoxy flavone and diosmetin. Conclusion LCMS-IT-TOF is a rapid and effective technology to obtain the chemical compositions in the flavonoid fraction of Mentha haplocalyx Briq., which is of great significance for further study.

    Determination of three diterpenoids in Isodon excisoides using solid phase extraction-high performance liquid chromatography (SPE-HPLC)*

    Objective To establish a method to simultaneously determine three diterpenoids in Isodon excisoides. Methods The procedure of SPE-HPLC was performed on a Venusil C18 column(4.6 mm×250 mm,5 μm) . The mobile phases was water and acetonitrile in gradient mode (0~12 min, 32%A; 12~18 min, 32~45%A; 18~35 min, 45%A). The flow rate was 1.0 mL·min-1. The column temperature was 30℃. The UV detection wave length was 230 nm. Results All the standard compounds showed a good linearity in the range of 0.056~0.448 g/L(r=0.999 2), 0.002 6~0.020 8 g/L(r=0.999 4), 0.016~0.128 g/L(r=0.999 7) for Kamebacetal-A, Kamebacetal-B, and Reniformin C, respectively. The average recoveries of the three diterpenoids were 97.53%, 96.02%, 98.23% with RSD 2.16%, 2.01%, and 2.10%, respectively. Conclusion This method is simple, accurate and effective. It could be used for quality control of ingredients in Isodon excisoides. It could also provide reference for the study of antitumor activity spectrum.

    Comparison of two chromatographic methods for the determination of sugars in Danshen Injection (lyophilized powder)*

    Objective To compare two chromatographic Methods and then to select an accurate, convenient and rapid method for the determination of sugars in Danshen Injection (lyophilized powder). Methods Firstly, HPLC-ELSD method was applied on an Alltech PrevailTM Carbohydrate ES (4.6 mm × 250 mm, 5 μm) column, and acetonitrile-water (76∶24) was used as the mobile phase on an isocratic elution with ELSD as the detector. Apart from that, IC method was adopted on a CarboPac PA20 (150 mm × 3 mm, S/N 003654 ) anion exchange column with the CarboPac PA20 ( 50 × 3 mm, S/N 003405) column as the guard column, and sodium hydrate solution (7 mmol/L) was used as eluent with the pulsed amperometric detector. To sum up, HPLC-ELSD and IC Methods described above were applied to determine the sugars in Danshen Injection (lyophilized powder) respectively. Results Under the chromatographic conditions mentioned above, HPLC-ELSD method could simultaneously determine the contents of fructose and glucose. Linear relationship, precision, repeatability and recovery rate of the method were all qualified; IC method could simultaneously determine arabinose, galactose, glucose, sucrose, mannose as well as fructose, and the method proved to be accurate and reliable. The determination Results of fructose and glucose by both Methods were basically the same. Conclusion IC method seems to be a better choice to determine the monosaccharides and disaccharide in Danshen Injection (lyophilized powder).

    Establishing a TCM pattern element identification scale for depression*

    Objective To establish a TCM pattern element identification scale for depression. Methods The Questionnaire of TCM Pattern Element Items for Depression was designed. TCM four diagnostic data were collected from 434 cases of depression, and TCM pattern elements of≥10% occurrence frequency were retained. TCM pattern elements were extracted based on Complex System Entropy Cluster. Conditional logistic regression of forward method was used for the selection of scale items. Binary logistic regression analysis was used to empower the items. Diagnostic threshold was established by receiver operating characteristic (ROC) curve. Results The TCM pattern element identification scale for depression was established based on 39 valid scale items. There are 8 pattern elements in the scale, including Qi-deficiency, blood-deficiency, Yin-deficiency, Yang-deficiency, Qi-stagnation, fire-heat, phlegm-dampness, and blood stasis. The diagnostic threshold score is 10 points. Conclusion Establishing a TCM pattern element identification scale for depression could offer a useful reference for standardization of depression patterns.

    The effects of external use of Zibai Gel on cervical high-risk human papilloma virus infection: a clinical observation*

    Objective To observe the effects of external use of Zibai gel on cervical high-risk human papilloma virus (HR-HPV) infection. Methods 85 cases with cervical HR-HPV infection were recruited and then divided into two groups quasi-randomly. 57 cases in the treatment group were applied with Zibai gel, while 28 cases in control group did not use any medication. The outcome was measured and observed. Results In the treatment group, the load of HR-HPV was reduced in 56 cases (98.24%) compared with only 9 cases (32.14%) in the control group. There was a significant difference between the two groups (Z=-5.770,P<0.05). In the treatment group, clinical symptoms were relieved in 56 cases (98.24%),compared with only 3 cases(10.71%)in the control group. There was a significant difference between the two groups (Z=-7.485,P<0.05). Cases of 12 patients from the treatment group were followed up after six months, and the load of HR-HPV showed no significant difference compared with those at the time right after the treatment (Z=-0.405,P>0.05). Conclusion The external use of Zibai gel could reduce the load of HR-HPV, and convert some of high-risk HPV infection negative. According to the Results in the six-month follow-up observation, the clinical effect was affirmative.

    Role of static heat pattern in the occurrence of acute hemorrhagic stroke*

    Objective To investigate the distribution and dynamic changs of pattern elements, especially the role of static heat pattern in the occurrence of cerebral hemorrhage in acute stage. Methods Based on Pattern Element Scale for Hemorrhagic Stroke at Acute Stage, we collected clinical manifestation data of 1 028 patients with acute hemorrhagic stroke. Distribution and dynamic changs of eight pattern elements (Static heat, Wind, Fire, Phlegm heat, Phlegm dampness, Blood stasis, Qi deficiency and Yin deficiency) at five different timing points after admission (upon adaission Day 3, 7, 14, and 21). Results The distribution of pattern elements in hemorrhagic stroke patients at the acute stage presented with a specific rule. Proportion of Static-heat pattern was the highest at each time point followed by Wind and Fire patterns. There was statistical longitudinal difference in the number of pattern elements (P<0.05) at different time points. Conclusion Static heat pattern seems to be the fundamental mechanism and play a leading role in the occurrence of the disease while wind, fire, and phlegm seems to derive from Static heat.

    A clinical study on the effects of supplemented Danggui Buxue Decoction on cytokines in IgA glomerulonephritic patients of kidney deficiency and blood stasis pattern*

    Objective To investigate the effects of supplemented Danggui Buxue Tang (Chinese angelica blood-tonifying decoction) on inhibition of cytokine congregation and kidney cirrhosis in patients with IgA glomerulonephritis. Methods Histopathologically confirmed IgA grlomerulonephritic patients were randomly divided into two groups. Both groups received routine treatment while treatment group was given an extra supplemented Danggui Buxue Tang. Outcome measures were urine and blood cytokines and fibrosis related parameters to compare the differences between groups. Results 24h urine protein level was both reduced in treatment group (from 5.78±1.10 g to 1.25±0.67 g) and control group (from 5.25±1.21 g to 2.09±0.89 g). Cytokines and fibrosis related parameters including MCP-1, MIF, TGF-β1,HA, LN, Col-IV, and PIIINP were all significantly improved after intervention in both groups respectively (P<0.05). The difference between two groups was significant in all factors except LN. Conclusion Combined treatment of with conventional and Chinese medicine is effective for IgA glomerulonephritis in terms of inhibiting cytokine aggregation and delay kidney cirrhosis. Adding supplemented Danggui Buxue Tang according to TCM pattern identification seems to improve effectiveness significantly.
