    Origin of TCM pulse diagnosis——medical knowledge derivation of “pulse” and origin of textual research of pulse sphygmology*

    According to Chinese character pattern analysis of “pulse”(Chinese: 脉) in Shuo Wen Jie Zi, its original meaning referred to the road of drainage in the natural environment, and it was extended to the TCM concept of organ of blood circulation of human body. The original meaning and extended meaning implied the TCM concept of “correspondence between man and nature” and “analogy by classification”. And throughout ancient historical books as well as the pre-Qin ancient medical literature and unearthed literature related to the content of the vein, the analysis of blood, qi, meridians of arteries and veins, pulse condition, pulse-taking in the early days of TCM theory of derivative and application, comprehensive comparison and each card entries, we found “pulse” was first recorded in Guo Yu in the early Warring States period. Through observation of the natural universe and the human body, the concept of “blood pulse” (Chinese: 血脉) began to emerge, in the medical literature Zhangjiashan Bamboo Slips Mai Shu and Mawangdui Silk Texts Zu Bi Jing. The theoretical system of classification of diseases by pulse and diagnosis of diseases by pulse was established, ran through the whole meridian theory, in which “distinguish based on different pulse conditions” showed the embryonic form of pulse diagnosis in early China.

    Interpretation of Pibulun of Suwen by angiosome theory

    Dermal parts (pinyin: píbù; Chinese:皮部),a Chinese medical term of one of important human structure concepts, was referred many times in Huangdi Neijing(Yellow Emperor’s Internal Classic). Pibulun of Suwen was a specific chapter to describe the different human skin parts according to six-meridian system, but the theoretical research on dermal parts has been rarely carried out in these years. In 1987, Australian scholar Taylor put forward a similar concept of skin area — angiosome theory, based on a large number of anatomical experiments. After comparing with both the theories, this paper found that angiosome theory could be helpful for understanding deeply and systematically Pibulun of Suwen. Angiosome theory emphasized the skin partitioned by meridians and made it thorough for understanding of the characteristics of collateral vessels, and it also provided the further research basis for the complexional inspection. It is beneficial to further understanding of related theory to dermal parts and further exploring its characteristics by modern medical science and technology under the enlightenment of the angiosome theory. Moreover, an accurate understanding will make it practicable to guide clinical research and practice based on dermal theory.

    Analysis of drug indications in Treatise on Febrile and Miscellaneous Diseases based on inferential validity*

    To explore the drug indications in Treatise on Febrile and Miscellaneous Diseases, taking ephedra,cassiatwig,peony and pinellia as examples. Using inferential validity of sufficient condition hypothetical proposition and necessary condition hypothetical proposition of propositional logic,to confirm the indication of ephedra, cassia twig, peony and pinellia. It stated that ephedrais was used for edema in excess pattern, cassia twig was used for dysuria with loose stool, peony was suitable for bellyache in no-deficiency-cold pattern, and pinellia for vomiting. It is more intuitive and reliable to use inferential validity in the study on drugs. And researchers can grasp the correspondence between drugs and syndromes more easily. Meanwhile, the effectiveness of thinking structure could be guaranteed as much as possible, when the indications of drugs is explained.

    Connotation and modern scientific evidence for“heart controlling mental activities”*

    The theory of “heart controlling mental activities”, a foundational theory in TCM, has been running through the entire principles, methods, recipes and medicines of TCM and guiding traditional Chinese physician’s clinical practice. At the beginning, this article reviewed the origin and connotation of the theory. Then the modern scientific evidence for the theory of “heart controlling mental activities” was focused on. The heart pump function is the important basis of normal brain function, the endocrine function of the heart is closely associated with the brain function, and patients with cardiovascular disease often suffer from cognitive dysfunction and anxiety depression. In addition, some Results of modern physics and cases of heart transplant were showed as the supplementary evidence. At last, the clinical effectiveness of the theory was explained. From the above, the cardiac function in modern medical science is closely related to the brain function in TCM is proved. We should be effort to research it deeper in order to fully understand and expound its theoretical connotation.

    Differentation treatment of diabetic kidney disease based on body-qi-spirit*

    Wholism of body-qi-spirit is one of the important points in Huangdi Neijing, governing all vital movement. Body,qi and spirit are highly harmonious, depending on each other closely. Body is the foundation of qi and spirit, qi is the use of body and spirit,and spirit governs qi and body. The current treatment of diabetic kidney disease focuses on “qi”level, but“body” level has been paid less attention, and“spirit”level has been ignored. Treatment of diabetic kidney disease on “body” level is to nourishing genuine-yin root in kidney,while treatment on “spirit”by treating zang-organ. Concurrent treatment of body,qi and spirit is the principle of prescription.

    A report of studies on the accuracy of references cited in Compendium of Materia Medica*

    Checking the accuracy of references cited in Compendium of Materia Medica is of significance for the research of Compendium of Materia Medica. Furthermore, it can also serve as an example for exploring the general rule on citations of historical documents by ancient authors. It has been found that there is a regularity in the format of the title of cited references, including book title, abridged title, alias title, author (surname, given name, secondary name, alternative name), name of reviser (surname, given name, secondary name, alternative name) etc. These references yield important significance for the discipline of literature. For example, by verifying the true and false of various editions of WU Rui’s Materia Medica in Daily Use of the Yuan Dynasty, the author of Dietetic Materia Medica of the Ming Dynasty and the true and false of its various editions can be thus confirmed. It is found that within the 276 references cited in the Compendium of Materia Medica, 64 cited references weren’t recorded in the former bibliographies. Moreover, there were also other problems appearing in these cited historical documents, such as two different books confused as one book, overlapping of books repeatedly, wrong cited book title, mistaken author, and with reference title but not really quoted etc. All these should be paid attention by the readers.

    Protective effects of Radix Astragali on retinal ganglion cells of high intraocular pressure rats*

    Objective To explore the protective effect of Radix Astragali (Milkvetch Root, Huangqi) on the retinal ganglion cells (RGC) of high intraocular pressure(HIP) rats, and the influence of HIP on the protective effect. Methods All rats were randomly divided into sham operation group, model group, Huangqi group, eye drops group, and Huangqi + eye drops group.HIP rat model was induced by cauterization of episcleral veins of both eyes. Four weeks after modeling, rats of sham operation group and model group were administered i.g. normal saline, those of Huangqi group were i.g. Huangqi water decoction(20 g/kg), rats of eye drops group took eye drops of Bimatoprost, and Huangqi + eye drops group took i.g. Huangqi water decoction combined with Bimatoprost. After four weeks’ intervention, the RGC number and RGC apoptosis rate were analyzed quantitatively using laser confocal microscope. Results The number of RGC in sham operation group was significantly higher than that of other groups (P< 0.01), and RGC apoptosis rate was significantly lower (P<0.01). While the Results of model group was opposite. Although RGC of eye drops group were more than those of Huangqi group, and apoptosis rate was lower, there were not significant difference(P>0.05). The number of RGC of Huangqi + eye drops group was significantly higher than that of Huangqi group and eye drops group, and the apoptosis rate was significantly lower than that of the two groups (P<0.05). Conclusion Huangqi water decoction can protect visual nerves of HIP rats by inhibiting RGC apoptosis, and the protective effects can improve by combination with intraocular pressure lowering medication.

    Effects of Chinese medicine compound of warming and moistening lung with tonifying spleen on guinea pigs with allergic rhinitis*

    Objective To study the effects of Chinese medicine compound of warming and moistening lung with tonifying spleen(hereinafter“TCM”) on behavior, serum IgE and IL-6, as well as nasal mucosa pathological changes of guinea pigs with allergic rhinitis. Methods SPF grade guinea pigs, half male and half female, were randomly divided into normal group, model group, Biyankang group (0.41 g/kg), loratadine group (0.91 mg/kg), TCM high-dose group (3.04 g/kg), mid-dose group(1.52 g/kg) and low-dose group(0.76 g/kg). Except the normal group, other groups were followed by bilateralnostrils intranasal inhalation once daily with 10% TDI olive oil solution(5 μL each nostril), 7 days in a row. After the model was established successfully, the guinea pigs were i.g. corresponding medication, with dripping TDI continuously for 14 days every other day. Nasal symptoms of all groups were observed and recorded. The serum contents of IgE and IL-6 were determined by ELISA, nasal mucosa pathological morphological changes were observed under light microscope, and number of eosinophils (EOS) were counted. Results Compared with the model group, scores of behavioral symptom were significantly lower in both TCM high- and mid- dose group (P<0.01 or P<0.05), and serum content of IgE and IL-6 significantly lower in all the TCM groups (P<0.01 or P<0.05). High-dose and mid-dose of TCM relieved EOS infiltration of nasal mucosa,compared with model group (P<0.01 or P<0.05), furthermore, in TCM high dose group, no exposure-related nasal lesions or no flammatory cell infiltration was found (P<0.05). Conclusion TCM could improve the behavior of guinea pigs with allergic rhinitis and reduce serum IgE and IL-6 levels, relieve the nasal mucosa pathological changes.

    Effect and mechanism of effective components of Mongolian medicine compound Shudage-4 against experimental stomach ulcer*

    Objective To explore the protective effect and mechanism of Mongolian medicine compound Shudage-4 on gastric ulcer induced by glacial acetic acid. Methods Healthy SD rats were randomly divided into Shudage-4 high, middle and low dose (high-, mid-, and low-dose) group, positive drug groups(Anweiyang group and cimetidine group), model group and normal group. Except for the normal group, the rats in the other six groups were modeled by glacial acetic acid. After the model was established successfully,rats of each group were admistered i.g corresponding medicine for consecutive 14 days. Then the gastric tissue was resected 24 h after the last administration. Ulcer index and ulcer inhabitation rate were measured, the content of NO and MDA in gastric mucosa were detected using ELIAS kits, and pathological changes of gastric tissue was stained by HE were observed under light microscope. Results Compared with the model group, the content of NO in gastric mucosa increased significantly and MDA decreased significantly in all three Shudage-4 groups (P<0.01). The inflammation decreased in all three Shudage-4 groups, in which mid-group and high-group showed more obvious effect with much less the number of inflammatory cells and improvement of the pathological condition. Conclusion Mongolian medicine compound Shudage-4 has the therapeutic effect on gastric mucosal injury induced by glacial acetic acid in rats. The mechanism may be related to its effect of antioxidation and vasodilation.

    Effect of Scutellaria baicalensis formula granule on proliferation and apoptosis of HepG2 cells*

    Objective To investigate the effect and machanism of Scutellaria baicalensis formula granule (SBFG) on proliferation and apoptosis of HepG2 human hepatoma cells.Methods HepG2 cells were divided into normal control group and six SBFG groups at concentration of 0.05, 0.1, 0.2, 0.4, 0.8 and 1.6 g/L. After HepG2 cells were treated by SBFG for 24 hours, the proliferation was analyzed by using MTT method, apoptosis was observed by Hoechst 33342 staining method, and the changes of cell count, nuclear size, nuclear DNA content, permeability, mitochondria membrane potential(MMP) and cytochrome C were tested by high content screening assays(HCS). Results The inhibitory effect of SBFG on proliferation of HepG2 cells was significant (P<0.01) with concentration-dependence, and IC50 of (0.38±0.016) g/L. By Hoechst 33342 staining, the morphological changes were observed including hyperchromatic nuclei, concentrated karyoplasms and karyorrhexis. Furthermore, HCS indicated that 0.1 g/L SBFG decreased MMP and increased cytochrome C release significantly (P<0.01); 0.4 g/L SBFG further decreased MMP and increased cytochrome C release, and increased the membrane permeability significantly, pyknosis and nuclear fluorescence intensity (P<0.01); in addition to the effects above, 1.6 g/L SBFG also reduced the number of cells (P<0.01). Conclusion SBFG can significantly inhibit the proliferation of HepG2 cells, and induce cell apoptosis. The mechanisms may be related to decreasing the MMP, promoting the release of cytochrome C and increasing membrane permeability.

    Chemical composition from Eupolyphaga Sinensis Walker*

    Objective To isolate new chemical compositions from Eupolyphaga Sinensis Walker (woodlouse) and evaluate their inhibitory activity on PKD1/TRPP2 activity,an indicator of polycystic kidney disease. Methods Isolation and purification by using silica gel normal and reversed phase chromatography, and semi-preparative HPLC. The structure of chemical compounds were determined based on their physic-chemical property and spectral parameters. Results Altogether eight chemical compositions were isolated and identified from ethyl acetate part of 70% acetone extracts, whose structures were determined by 1D and 2D NMR spectroscopic analysis as 1H-pyrrolo[1,2-a][1,4]diazepine-1,5(2H)-dione,hexahydro-3-Para phenyl phenol (1), thymine (2), 2-methyl-6-(2’3’4’-3 OH-butyl) pyrazine (3), 3-hydroxy-pyridinol (4), (3S,8aS)-3-isobutylhexahydropyrrolo[1,2-a]pyrazine-1,4-dione(5),3-ethyl-6,8-dihydroxy-7-methyl-3,4-dihydroisochromen-1-one (6), 3-(p-hydroxyphenyl)-N-methylpropionamide (7), and 6,8-dihydroxy-3,7-dimethyl-3,4-dihydro-1H-isochromen-1-one (8). Conclusion Composition 1 was a new composition, with inhibitory effect on PKD1/TRPP2 activity, and composition 2~8 were first isolated from woodlouse.

    Release characteristics of colon-targeted pellets of the volatile oil of Radix Angelica Sinensis in rats*

    Objective To study the release characteristics of colon-targeted pellets of volatile oil of Radix Angelica Sinensis (Danggui) in rats. Methods Rats were administrated intragastrically with the colon-targeted pellets of the volatile oil of Radix Angelica Sinensis. At different time points, the rats were sacrificed and their gastrointestinal tracts were resected, in which the contents were collected. Ligustilide, as the index of volatile oli of Danggui to evaluate the release characteristics of the colon-pellets in digestive tracts, was determined using HPLC. Results The linear range of ligustilide in the digestive tract of rats was 1.268~507.2 mg/L. Four hours after administration, ligustilide was detected in cecum and colon. And 20 h later, ligustilide could still be detected. That meant the colon-targeted pellets could maintain the drug release for a long time in colorectal sites of rats. Conclusion The prepared Danggui volatile pellets can be used as the carrier for targeting on colon, which can be used in the study of the chemo-prevention of inflammation related to colorectal cancer at the late stage.

    Comparison of different extraction methods on Gegen Qinlian Decoction*

    Objective By comparing the efficacy and toxicity of three extraction Methods of Gegen Qinlian formula, so as to underline the appropriate technology of its extraction and preparation. Methods Mice were divided into blank group, pathological group, tablet group, pill group and decoction group, in which the latter three groups were divided into high-, mid- and low- dose subgroups respectively. The mice models were induced by i.g. senna decoction 20 mL/kg(4 g/kg). The tablet group were i.g. extract water solution at dose of 1.60, 0.80,0.40 g/kg, the pill group were i.g. at dose of 6,3 and 1.5 g/kg, and the decoction group at dose of 13,6.5 and 3.25 g/kg. The number of the wet dung was chosen to evaluate the antidiarrheic effect of Gegen Qinlian formula. Then the acute toxicity test of different samples was carried out through maximal dosage experiments. Mice were divided into tablet group, pill group and decoction group, with 10 mice in each group. And the maximum dosage were 0.66 g/mL(tablet), 0.63 g/mL(pill) and 0.74 g/mL(decoction), equivalent to crude drug pieces of 67.96, 89.01 and 177.77 g/kg, and equivalent to 24, 9 and 6 times daily dose of per kilogram of body weight of 70 kg. After the single dose administration, the reactions of the mice were observed for consecutive seven days. Results The sample of three dosage forms of Gegen Qinlian formula had obvious therapeutic effect (P<0.001), among which the antidiarrheic effect of tablets was best (P<0.05), pill less and decoction least. No death or obvious abnormalities was discovered under maximum dosage. Conclusion In the three preparations, Gegen Qinlian tablets extraction preparation method recorded in 2015 Chinese Pharmacopoeia had the best inhibitory effect against diarrhea caused by senna leaves.

    Distribution of patterns, pattern elements and the relationship between common patterns and corresponding symptoms/signs: a modern literature study on common hand-foot-mouth disease*

    Objective Through retrospective study of latest 20 years’ modern literature related to common hand-foot-mouth disease, the distribution of pattern elements and the relationship between common patterns and corresponding symptoms / signs were analyzed, so as to provide the basis for standardization of TCM pattern differentiation for hand-foot-mouth disease. Methods “hand-foot-mouth disease” OR“hand-foot-mouth” AND “pattern” OR“Chinese medicine” were used as subject words in CNKI,Wanfang and VIP database to retrieve the relevant literature. Search Results were input to establish the standard database by using Epidata 3.0 software, then frequency of symptoms, pattern and pattern elements logistic regression analysis were analyzed by SPSS 17.0. Results The leading symptoms and signs of common hand-foot-mouth disease were rash, fever and anorexia. The common patterns were damp-heat pattern of lung and spleen, pattern of stagnation and steaming of damp-heat and pattern of pathogen invading lung-defense phase. The disease-locations with high frequency were lung, spleen and defense and the disease-natures with high frequency were damp-heat and heat. After Logistic regression analysis, the symptoms/signs of damp-heat pattern of lung and spleen were slobbering and purple venule of index finger, those of pattern of stagnation and steaming of damp-heat were thick tongue coating, high fever and dark, thick venule of index finger, and those of pattern of pathogen invading lung-defense phase were running nose and cough. Conclusion The common patterns types of common hand-foot-mouth disease were damp-heat pattern of lung and spleen, pattern of stagnation and steaming of damp-heat and pattern of pathogen invading lung-defense phase. And the common location elements were lung, spleen and defense with the common nature elements of damp-heat and heat.

    Effect of electro-acupuncture on Aβ in cortex and serum of APP/PS1 transgenic mice*

    Objective To observe the effect of eletro-acupuncture (EA) on the expression of Aβ1-40 and Aβ1-42 in cortex and serum of APP/PS1 transgenic mice with Alzheimer’s disease (AD), so as to explore the mechanism of EA therapy on AD according to the clearance of Aβ across blood brain barrier (BBB). Methods Twenty 6-month-old APP/PS1 transgenic mice were randomly divided into model control group and EA group, and ten more C57BL/6 wild type mice were assigned into the normal control group. After one and half months’EA treatment at the acupoint“Yǒngquán”(KI 1) and“Bǎihuì”(GV 20), Aβ1-42 expression in the cortex was observed by using immunohistochemistral staining, and content of Aβ of cortex and serum were measure by ELISA method. Results In the model group, deposits of mature senile plaques were observed, which were weak in EA group . Compared with the model group, the expression of Aβ1-40 and Aβ1-42 in EA group significantly decreased (P<0.01), both in cortex and in serum. Conclusion EA therapy can decrease the content of cortex and serum Aβ, especially on Aβ1-42 which has neurotoxicity, which may be one of ways of EA treatment on AD.
