    Analysis of three formulas against heart-yang deficiency based on syndrome differentiation in Treatise on Cold-Induced Diseases*

    Heart-yang deficiency syndrome, due to heart-yang deficiency, weak beating and internally exuberant yin cold, is characterized by palpitation or severe palpitations, stuffiness in the chest, lacking in strength, aversion to cold and cold limbs, pale complexion, or even critical signs triggered by syndrome of collapse of heart-yang. Heart yang collapse syndrome can be seen in sever conditions. Guizhi Gancao Tang, Guizhi Gancao Longgu Muli Tang and Guizhi Qu Shaoyao Jia Shuqi Muli Longgu Jiuni Tang, directed at hear-yang deficiency syndrome recorded in Treatise on Cold-Induced Diseases (Shang Han Lun) written by ZHANG Zhongjing, were referred to apply on mild, moderate and severe conditions of heart-yang deficiency. However, based on the analysis of the features of signs and syndromes of the three formulas against, this article puts forward that their indications are all emergency and intensive. And the difference among them are that Guizhi Gancao Tang focuses on tonifying heart to unblock yang, as well as warming the nutrient to nourish blood, resolving qi to move water, calming surging and directing counterflow downward, Guizhi Gancao Longgu Muli Tang emphasizes in subduing and astringing, and Guizhi Qu Shaoyao Jia Shuqi Muli Longgu Jiuni Tang can both treat unreleased of exterior pathogen and internally oppressed phlegm-heat.

    Mechanism of tetramethylpyrazine in treatment of coronary heart disease based on the coexpression-protein interaction network*

    Objective To explore the mechanism of tetramethylpyrazine in the treatment of coronary heart disease (CHD) based on the coexpression-protein interaction network analysis. Methods The targets’ information of tetramethylpyrazine was obtained from pharmacophore-based virtual screening and database searching from STITCH & Chemprot, and the protein-protein interactions of the targets were retrieved by String database. The protein interaction network was constructed by Cytoscape. Next, the coexpression-protein interaction networks were constructed by integrating gene-expression profile under physiological and disease conditions. The modules of disease associated coexpression-protein interaction network were clustered and functional annotated based on fast agglomerate algorithm based on the edge clustering coefficients (FAG-EC) and gene ontology (GO) enrichment, by which the function modules were mapped to the coexpressin-protein interaction network under the normal physiological state to get the corresponding function modules. Then the function modules under two different conditions were compared and analyzed. Results The effect of tetramethylpyrazine on CHD was achieved by means of regulating and controlling immunological regulation or inflammatory response, cell apoptosis signaling pathway, blood circulation, heme metabolism and basal metabolism; myeloperoxidase (MPO) and hemeoxygenase-1 (HMOX1) probably were two key targets in treatment of CHD. Conclusion The mechanism and the key targets of tetramethylpyrazine in the treatment of CHD were illuminated at the molecular networks level, which could provide reference for clinical practice.

    Hypolipidemic effect and mechanism of Semen Astragali Complanati on hyperlipidemia rats with kidney-yang deficiency pattern*

    Objective To explore the effect of water extracts from Semen Astragali Complanati (Flatstem Milkvetch Seed, hereinafter“SAC”) on lipid metabolism in hyperlipidemia model rats with kidney-yang deficiency pattern, so as to try to discover the mechanism. Methods Female 10-week-old SD rats were randomly divided into six groups (each n=10): normal group, model group, estrogen group, SAC low-dose group, SAC mid-dose group and SAC high-dose group. The rat model was induced by bilateral ovariectomization combined with high-fat diet feeding. And rats of normal group and model group were intragastrically (i.g.) administered with saline, those of estrogen group were i.g. with estradiol valerate (0.2 mg/kg), and three SAC groups were i.g. with SAC extract at the dose of 34, 68 and 136 mg/kg for consecutive 8 weeks. The body weight of rats were recorded, serum TC, TG, HDL-C and LDL-C were measured and serum E2, FSH and estrogen receptors (ERα, ERβ) mRNA in liver tissue were detected. Results Compared with the normal group, the body weight of rats of model group increased significantly after three weeks’high-fat feeding (P<0.001); from Week 5 the level of total cholesterol(TC) and low density lipoprotein cholesterol(LDL-C) increased significantly (P<0.001 or P<0.01), and from Week 7 triglyceride(TG) and high density lipoprotein cholesterol(HDL-C) also did (P<0.001 or P<0.05); serum E2 significantly decreased (P<0.01), and follicle stimulating hormone(FSH) increased (P<0.05), the expression of ER-α mRNA in liver significantly decreased(P<0.05). High dose of SAC significantly reduced the serum level of TG and TC from Week 7 (P<0.05 or P<0.001), middle dose of SAC reduced the level of LDL-C and increased HDL-C significantly at the same time point (P<0.05, P<0.01), and a more week later, the low dose increased the serum level of HDL-C (P<0.05). Despite the levels of E2 and FSH of model rats showed no significant difference after treatment by SAC (P>0.05), the expression of ER-α mRNA in liver tissue increased in all SAC groups (P<0.05). Conclusion The effect of regulating blood lipid metabolism of SAC had been proved. The potential mechanism could be via upregulating the expression of estrogen receptors to amplify the signals transmitted by the receptors, which may make up the low estrogen level to direct at the disorder of lipid metabolism during peri-menopausal period.

    Effect of Qingjin Huatan Decoction on Foxp3 and RORγt in lung tissue of rats with acute exacerbation of chronic obstructive pulmonary disease*

    Objective To explore the effect of Qingjin Huatan Decoction on forkhead box P3 (Foxp3) and retinoid-related orphan nuclear receptor γt (RORγt) in the lung tissue of airway mucus hypersecretion rat model with acute exacerbation of chronic obstructive pulmonary disease (AECOPD). Methods Altogether 40 male Wistar rats,SPF grade, were randomly divided into normal group, model group, Qingjin Huatan Decoction group and clarithromycin group. AECOPD rat model was induced by intratracheal instillation of lipopolysaccharide (LPS) with passive smoking. From the first day of the experiment, rats of model group were intragastrically administered saline, and those of Qingjin Huatan Decoction group and clarithromycin group took corresponding medicines at dose of 8.4 g/kg and dose of 50 mg/kg for consecutive 30 days. On day 31, all rats were sacrificed. The thin sections of the rats’ resected lungs were made, the expressions of mucin 5AC (Muc5AC), neutrophil elastase (NE), Foxp3 and RORγt in lung tissue were detected by using immunohistochemical method, and the protein expressions of Foxp3 and RORγt were measured by using Western blot. Results Compared with the normal group, Foxp3 protein expression in model group had no significant difference (P>0.05), but expression of RORγt, Muc5AC and NE increased significantly with Foxp3/RORγt decreased significantly (P<0.01). Compared with model group, Foxp3 expression in Qingjin Huatan Decoction group had no significant difference (P>0.05), but expression of RORγt, Muc5AC and NE decreased significantly with Foxp3/RORγt increased significantly (P<0.01 or P<0.05). Compared with clarithromycin group, Muc5AC decreased significantly in Qingjin Huatan Decoction group (P<0.05). Conclusion The potential mechanism of Qingjin Huatan Decoction against AECOPD mucus hypersecretion was to downregulate RORγt protein expression in lung tissue and to upregulate Foxp3/RORγt.

    Laxative-induced constipation on age-related indices: an experimental study*

    Objective To explore the correlation of constipation and aging, by detecting age-related indices of rat constipation model. Methods The rats were fed with Rhubarb powder, then the shape, color, weight and number of feces and the intestinal transport function within 24 h were measured to confirm the successful model. The serum SOD, thymus index and spleen index, and the expression of klotho protein in the hippocampus were compared between the healthy rats and the model rats. Results Compared with the healthy rats, the weight and number of feces of model rats within 24 h, percentages of the intestinal propulsion of activated carbon,the level of serum SOD, thymus index, and klotho protein expression in the hippocampus, as well as the optical density of the klotho target protein and klothoβctin, all declined significantly(P<0.01 or P<0.05). Conclusion Laxative-induced constipation has a certain correlation with aging, but its mechanism should be done.

    Effect of wind-dispelling drugs and deficiency-tonifying drugs on axonal growth inhibitory signaling pathway Nogo-A/NgR and RhoA/Rock2 after cerebral ischemia in rats:a developmental study on Hou’s Hei San*

    Objective To explore the effect of wind-dispelling drugs and deficiency-tonifying drugs of Hou’s Hei San, a TCM formula used in treatment of apoplexy, on axonal growth inhibitory factors Nogo-A,NgR, RhoA, Rock2 of rats with cerebral ischemia. Methods The model of permanent middle cerebral artery occlusion ( pMCAO) was induced by using Longa method. All rats were randomly divided into sham-operation group, model group, wind-dispelling drugs group(7.70 g/kg), deficiency-tonifying drugs group(2.59 g/kg) and combined drugs group(10.50 g/kg). After operation, each 12 h the rats were intragastrically administered for 72 h. Then the rats were sacrificed and their brain tissue were resected. The protein expression of Nogo-A, NgR, RhoA and Rock2 in hippocampus CA1 were detected by using Western blot. Results The protein expressions of Nogo-A, NgR, RhoA and Rock2 in model group increased significantly, compared with sham-operation group(P<0.01). Compared with model group, the expression of NgR in wind-dispelling drugs group, deficiency-tonifying drugs group and combined drugs group decreased remarkably(P<0.01). The expression of Nogo-A and Rock2 decreased as well, but only combined drugs group showed a statistically difference(P<0.05). Conclusion The effect of wind-dispelling drugs and deficiency-tonifying drugs of Hou’s Hei San against cerebral ischemia damage would be via regulating axonal growth inhibitory signal pathway Nogo-A/RhoA/ Rock2 to promote the recovery of neural function.

    Effect of Yijing Fang on expression of junctional adhesion molecules and zonula occludens in testicular sertoli cells of adenine-induced infertile rats*

    Objective To observe the effect of Yijing Fang, a TCM compound, on the expression of junctional adhesion molecules (JSM)and zonula occludens (ZO) in testicular sertoli cells of infertile rats with kidney-yang deficiency pattern induced by adenine. Methods Altogether 48 healthy male Wistar rats of SPF grade, two-month-old, were divided according to random number table, into blank group, model group, Fufang Xuanju group and Yijing Fang group(n=12 each). After modeling, the rats were intragastrically administered by corresponding medicines for consecutive 20 days. Then the rats were sacrificed and their testis and epididymis were resected. The sperm density and sperm activity were calcula-ted using Computer Assisted Sperm Analysis System, and the expression of JAM and ZO in testicular sertoli cells were measured using immunohistochemical method. Results Compared with the normal group, the sperm density, sperm activity rate and expression of JAM and ZO declined significantly(P<0.01). Compared with the model group, the sperm density and the sperm activity rate increased in Fufang XuanJu group and Yijing Fang group remarkably (P<0.05), while expression of JAM and ZO increased in Yijing Fang group((P<0.05). Conclusion Yijing Fang can repair the damage of testicular sertoli cells of kidney-yang deficiency pattern rats induce by adenine, via regulating the expression of JAM and ZO.

    Effect of total flavonoid glycosides from Fructus Aurantii Immaturus on visceral hypersensitivity in rats with functional dyspepsia*

    Objective To investigate the effect and mechanism of total flavonoid glycosides from Fructus Aurantii Immaturus (Immature Bitter Orange, Zhishi) on visceral hypersensitivity in rats with functional dyspepsia (FD). Methods Forty ten-day-old male SD rats were randomly divided into five groups: normal group, model group, total flavonoid glycosides of Zhishi(TFGZ) high-dose group, mid-dose group and low-dose group (each n=8). FD model rats were induced by intragatrical administration of iodoacetamide with tail-clamping stress. When the rats were 8 weeks old, they were intragstrically administered corresponding dosage of TFGZ for consecutive 14 days. Then abdominal withdrawal reflex (AWR) score, electromyographic (EMG) activity of Musculus trapezius cervicalis and gastric accommodation during gastric balloon distention were recorded. The optimal dose of TFGZ was screened for the next study. Fifteen more ten-day-old male SD rats were randomly divided into three groups—normal group, model group and TFGZ group at optimal dosage. The method of modeling and treatment were as same as before. After treatment, the expression of 5-HT in gastric and spinal cord tissue and c-fos mRNA in spinal cord tissue were measured. Results Under the intragastric balloon pressure of 40, 50 and 60 mmHg, AWR score and change rate of root-mean-square (RMS)of EMG of rats in TFGZ high-dose group and mid-dose group were significantly lower than those of model group (P<0.05), while gastric accommodation was higher than that of model group (P<0.05). While there was no significant difference between the mid-dose group and high-dose group (P>0.05), so middle dose was confirmed to be optimal. TFGZ middle dose significantly decreased 5-HT and c-fos mRNA expression of rats with FD (P<0.05), and there was no significant difference with those in normal group (P>0.05). Conclusion Total flavonoid glycosides from Zhishi could effectively reduce the visceral hypersensitivity in rats with functional dyspepsia.Its mechanism may through the way of lowering the level of 5-HT and c-fos in gastric and spinal cord tissue.

    Establishment and evaluation of psychological stress asthmatic rat model*

    Objective To induce the psychological stress-asthmatic rat model by ovalbumin (OVA)mixture sensitization with restraint, which simulated the pathogenic factors of bronchial asthma with liver-qi stagnation pattern, as well as to evaluate the model using indicators of the macro characterization, behaviors, pulmonary function, peripheral blood inflammatory cells, bronchial tissue pathomorphology. Methods A total 32 male rats were randomly divided into four groups—control group, stress group, asthmatic group, and stress-asthmatic group. Rats of stress group and stress-asthmatic group were immobilized in restrained box once daily at different time, ten minutes more than the day before with the first duration of 2 hours. Rats of stress-asthmatic group and asthmatic group were sensitized by intraperitoneal injection 1 mL mixture of OVA with aluminium hydroxide on the first day, and after 13 days, they were stimulated by inhalation of 1 % OVA for 20 min for consecutive 14 days. The rats of control group were sensitized and stimulated by saline, without immobilizing . 28 days later, the macro characterization, behaviors, peripheral blood eosinophils, pulmonary function, bronchial tissue pathomorphology were observed. Results Stress asthmatic rats showed low mood, loss of interest, dull hair,loose stools, reducing food intake,slowly weight gain, and scratched noses and abdominal skin, coughed, sneezed and wheezed in the throat when stimulated. Compared with control group, the points of vertical movement decreased significantly(P<0.05 or P<0.01). Preference for sucrose, the times of active moves in tail suspension test (TST) of stress-asthmatic rats obviously decreased(P<0.01), and the duration of immobility increased significantly(P<0.01). Pulmonary function indicators, including peak expiratory flow(PEF), forced expiratory flow (FEF 25%-75% ), forced expiratory volume in 0.3 s [FEV(0.3]/forced expiration volume capacity (FVC) significantly declined(P<0.05,P<0.01,P<0.01), while mean inspiratory airway resistance (Ri) and mean expiratory airway resistance (Re) increased remarkably (P<0.05,P<0.01).Peripheral blood eosinophil cells increased(P<0.01), and infiltration of inflammatory cell in lung tissue was obvious. Compared with the stress group or asthmatic group, the signs and indicators of stress-asthmatic group were all much worse. Conlusion The psychological stress-asthmatic rat model, induced by VOA mixture with restraint, was proved to be successful. It is in accordance with clinical characteristics of bronchial asthma with liver-qi stagnation pattern, which can be used to study asthma caused by emotion .

    Influence of electro-acupuncture of different frequency and stimulation times on skin blood perfusion around Neiguan (PC6) and sham acupoint in rats with myocardial ischemia*

    Objective To compare the skin blood perfusion around Neiguan (PC 6) and sham acupoint needled by the different frequency and stimulation times of electro-acupuncture (EA), in healthy rats and myocardial ischemic rats. Methods According to random number table, altogether 100 male Wistar rats were divided into once treatment group and three times treatment group, which were both divided into control group, sham-operation group, model group, EA low-frequency group and EA high-frequency group (each n=10). After successful modeling, rats of EA groups were acupunctured at left Neiguan (PC 6) acupoint. 60 minutes after last treatment of acupuncture, skin blood perfusion around bilateral Neiguan(PC 6) acapoints and sham acupoint were tested by laser speckle contrast imaging technology (LSCI) and analyzed, and the myocardial morphological changes were observed by HE staining. Results Compared with control group, the skin blood perfusion around PC 6 in model groups decreased significantly after three time treatment (P<0.01), and that around PC 6 or sham acupoint in model group of three times treatment group was much less than that of once treatment group (P<0.01). Compared with the model group, the skin blood perfusion around PC 6 both significantly increased after low-frequency EA or high-frequency EA after three times treatment (P<0.01). Conclusion The variation of skin blood perfusion around PC 6 should be useful for specific reflection of normal condition or ischemic condition of rats myocardium, but that around sham acupoint didn’t. The effect of three-times-treatment against declination of skin blood perfusion due to myocardial ischemia was superior to that of once-treatment regardless of high frequency or low frequency, and EA of high frequency and three-times stimulation showed the significant effect. However, the mechanism remains to be further studied.

    Relationship of time and analgesic effect of buccal acupuncture on rabbits with rheumatoid arthritis*

    Objective To explore the relationship of time and analgesic effect of buccal acupuncture on rabbits with rheumatoid arthritis (RA), so as to provide the experimental basis for the clinical practice of buccal acupuncture. Methods Altogether 40 rabbits were randomly divided into normal group, model group, body acupuncture group and buccal acupuncture group (each n=10). RA model was induced by subcutaneous injection of ovalbumin. After modeling, rabbits of body acupuncture group were needled at the acupoint of bilateral “Xiyan” and “Tsusanli” and those of buccal acupuncture group were needled at the buccal acupoint of “knee” once daily for ten times. Heat pain threshold at the time point of 0, 5, 15, 30, 60, 120, and 240 minutes were recorded at the 1st, 4th, 7th, and 10th treatment. Results The buccal acupuncture and body acupuncture both showed definite analgesia effect. The heat pain threshold of buccal acupuncture group began to increase after five-minute treatment, reached the peak value at the 30th minute, then declined at the 60th minute to the same as that before treatment at the 240th minute. The variation trend of heat pain threshold of body acupuncture group was in common with that of buccal acupuncture group, but the pain threshold at the 240th minute was higher than that before treatment. Conclusion Buccal acupuncture against pain showed obvious effect-time relationship which was as similar as that of body acupuncture, but the onset time of buccal acupuncture was shorter than that of body acupuncture, and the duration was shorter. Furthermore, the analgesic effect of buccal acupuncture showed no relationship with times.

    Application of analogiczl acupoints on dysmenorrheal: a literature study of latest 10 years*

    Objective To summarize the application of analogiczl acupoints, defined as a group of acupoints directed at the same disease, by data mining method on literature related to acupuncture and moxibustion treatment on dysmenorrheal in latest 10 years. Methods By the way of computer retrieval of Chinese Journal Full-Text database (CNKI) and Wanfang database (WF) and VIP database, a total 128 articles from 2010 to 2016 related to acupuncture and moxibustion treatment on dysmenorrheal were searched and sorted out. And the acupoints recorded were input into the computer and analyzed. Results There were 49 analogiczl points, of which 47 points at fourteen meridians and 2 extra points. The leading three points were Sanyinjiao (SP 6), Guanyuan (CV 4) and Zhongji (RN 3); the leading meridians were bladder meridian of foot-Taiyang and Ren meridian; the location of the acupoints were almost located on thoracic abdomen and low back. According to the frequency, the analogiczl points were classified into three grades: 9 of first-grade, 13 of second-grade and 27 of third-grade. Conclusion The concept of “analogiczl acupoints” make a further interpretation of the synergetic effect of combination of acupoints, which would guide the selection of acupuncture prescription to improve the clinical effectiveness.
