    Validation of laws of disease progression in Yellow Emperor’s Canon of Medicine based on modern clinical reports*

    Based on the modern clinical trials report on time to onset of various diseases, time to progression, and time of death, this paper used a way of deductive argument on disease course recorded in Yellow Emperor’s Canon of Medicine, so the profound practical significance of the thought of “annual variation period” and “daily variation period” in Yellow Emperor’s Canon of Medicine was proved. Yellow Emperor’s Canon of Medicine interpreted two rules of disease progression— restriction or responsibility which was validated. By comparing the two rules, this paper put forward that restriction was the variation due to exterior environment exerting on disease, and responsibility was the variation due to patients’ internal environment on disease. Only by combining with the two rules could the clinical practitioners find the keys to disease prognosis and death.

    Prolactin: A potential molecular marker of TCM pattern of liver depression in women*

    Pattern of liver depression, a very common TCM pattern in patients especially in female patients, whose pathophysiological connotation in modern society is still to be explored. This paper attempts to find out the modern molecular mechanism of pattern of liver depression in women, based on the pathogenesis and clinical characteristics of the pattern, on the pathophysiological changes of female reproductive diseases related to the pattern and on the latest research findings of prolactin, combined with retrieving the literature of clinical or animal experiment related to prolactin-pattern-female diseases. The abnormal expression of prolactin can be found in diseases related to pattern of liver depression in women and all female animal model with pattern of liver depression, furthermore, TCM formula guided by the principle of soothing the liver could inhibit the overexpression of prolactin. As the result, the author proposes that prolactin has exact correlation with pattern of liver depression in women, and prolactin may be the novel target molecule of the pattern. The author expects that the clues could help to reveal the molecular mechanism of pattern of liver depression in women.

    Development and prospect on formula-based syndrome differentiation*

    A TCM term of formula-based syndrome differentiation, which originated from Treatise on Cold Pathogenic and Miscellaneous Diseases, is a method of syndrome differentiation for TCM clinical diagnosis and treatment. The medical practitioners of later generations have been studying it in its long history. This paper tried to study the origin and development of the method, based on its thought origin, on the difference between formula-based syndrome differentiation and formula-related syndrome, and on its superiority with the other eight syndrome differentiation methods. Undoubtedly, formula-based syndrome differentiation is different from formula-related syndrome, and it is the most direct method of TCM syndrome differentiation and treatment, combined syndrome differentiation with treatment. Formula-based syndrome differentiation is the correspondence of formula-syndrome with disease-syndrome, emphasizing the importance of primary symptoms. As a new academic thought, formula-based syndrome differentiation shows very high value in use in clinical practice and deserves further studying.

    Discussion on the molecular mechanism of “liver storing blood with controlling dispersion” based on erythropoietin signaling pathway*

    Based on the connotation analysis of TCM term of “liver storing blood with controlling dispersion” combined with achievement of recent research of western medicine, this article puts forward that “liver storing blood” is not derived from the function of “controlling dispersion”, but is the material barrier. The function of “controlling dispersion” reflects in “liver storing blood” in regulation of qi, spleen and stomach, reproduction or emotion, conversely, the normal function of “liver storing blood” relies on the normal function of “controlling dispersion”. So this article attempts the first time to discuss the molecular mechanism of “liver storing blood with controlling dispersion” based on erythropoietin (EPO) signaling pathway. The effect of EPO on production or regulation of blood and against depression is the reliable basis for TCM theory of “liver storing blood with controlling dispersion”. This study should provide the new idea to explore the mechanism of depression, premenstrual syndrome and other emotional diseases.

    Interpretation of theoretical basis of therapy of activating blood, clearing heat from the nutritive level, removing toxic substances on coronary heart disease / thoracic obstruction based on TCM concept of blood stasis, heat and toxin*

    The study on etiology and pathogenesis of coronary heart disease / thoracic obstruction has been carried on for a long time, and the understanding of TCM concept of blood stasis, heat and toxin was deeper and wider. A large quantity of clinical trials have proved that one of key pathogenesis of coronary heart disease / thoracic obstruction is the toxin induced by accumulation of blood stasis and heat, which damages heart blood or nutrient. In detail, the evil of blood stasis and heat were blocked or accumulated in human bodies to generate the toxic materials called toxin, which can damage collaterals of heart, burn out nutriment-blood of heart, block vessels of heart, and in the end, cause coronary heart disease / thoracic obstruction. Undoubtedly, the therapy of activating blood, clearing heat from the nutritive level, removing toxic substances has positive effectiveness. This article explains the relationship between the evil of blood stasis, heat and toxin with the occurrence and progress of coronary heart disease / thoracic obstruction, and discuss the theoretical basis of TCM therapy mentioned above directed at the evils.

    Effect of Xiaoyin Jiedu Fang on JAK1/STAT3 signaling pathway in Jurkat T cells*

    Objective To study the effect of Xiaoyin Jiedu Fang, a Chinese medicine compound formulated by TCM principle of cooling blood with detoxification, on nonreceptor tyrosine kinase JAK1/ signal transducer and activator of transcription 3 (STAT3) signaling pathway in human acute leukemia Jurkat T cells. Methods The abnormal activation of T cells of psoriasis with TCM pattern of blood-heat was induced by stimulating Jurkat T cells with phytohaemagglutinin (PHA) and IL-6. The medicated serum contained Xiaoyin Jiedu Fang granule (XYJDF), Liangxue Fang (LXF) or Jiedu Fang granule (JDF) which was a disassembled prescription of XYJDF, acitretin (positive drug) and distilled water (negative control) were prepared. The model cells were inoculated into 24-hole cell culture plate and were cultured with the prepared medicated serum and Filgotinb(GLPG0634), an inhibitor of JAK/STAT signaling pathway for 48 hours. Jurkat T cells with no treatment was served as the blank group. Protein and mRNA expression of JAK1, STAT3, glucoprotein(GP)130 were detected by using Western blot and real-time PCR. Results The protein and mRNA expression of JAK1,STAT3 and GP130 in the model group were significantly higher than those of the blank group (P<0.01). Compared with the model group, the protein expression of pJAK1/JAK1, pSTAT3/STAT3 and GP130 in all medicated group were decreased significantly (P<0.01), and the mRNA expression of JAK1, STAT3 and GP130 declined (P<0.01 or P<0.05). Conclusion The effect of XYJDF against psoriasis could be via the way of inhibiting JAK1/STAT3 signaling pathway leading to prevent abnormal activation of T cells.

    Effect of Huoxue Jiedu Recipe on Hippo signaling pathway in diabetic rat retina *

    Objective To observe the effect of Huoxue Jiedu Recipe(HJR), a Chinese medicine compound, on Hippo signaling pathway in diabetic rat retina. Methods A total 24 healthy SD rats were fasted for 12 h and administered intraperitoneally streptozotocin (65 mg/kg) once to induce diabetic model. After 20 weeks, the diabetic rats were randomly divided into model group and HJR group. And 10 more healthy rats were assigned into normal group with administration intragastrical saline. Each group was intervened once daily for consecutive 12 weeks. The levels of phosphorylated mammalian sterile 20-like kinase(P-MST), large tumor suppressor gene 1 (Lats1), phosphorylated Yes-associated protein(P-YAP), tafazzin (TAZ), TEA domain family member(TEAD) protein in retinal tissue were detected by immunohistochemical method and Western blot. Results Compared with the normal group, the protein expression of P-MST and P-YAP were decreased in the model group. While compared with the model group, the protein expression of P-MST and P-YAP significantly increased in HJR group. And Western blot results showed that protein expression of Lats1, TAZ and TEAD in model group were higher than those of normal group, and HJR decreased the protein expression of Lats1, TAZ and TEAD. Conclusion The effect of Huoxue Jiedu Recipe on diabetic retinopathy may be via regulating Hippo signaling pathway.

    Preliminary study of lung function affected by spleen deficiency pattern combined with pulmonary fibrosis of rats*

    Objective To observe the effect of spleen deficiency syndrome induced by rhubard solution combined with lung fibrosis induced by bleomycin on lung function of rats, so as to discuss the relationship between spleen deficiency syndrome (SDS) and pulmonary fibrosis. Methods SD rats were randomly divided into normal group, SDS group, model group, SDS + lung fibrosis group. The rat SDS model was induced by intragastrical administration of rheum officinale with fasting every other day for 30 days. Then the rats were infused bleomycin(5 mg/kg)through tracheal intubation to simulate the lung fibrosis disease with SDS. Four weeks after injection of bleomycin, rats’ lung function was detected, lung index and spleen index were calculated, and the pathological changes of lung tissue and aiway were observed by using HE staining and Masson staining. Results Compared with the normal group, weight of lung, lung index, FEV0.4/FVC% were increased in the model group(P<0.05), and the lung dynamic compliance (Cdyn), forced vital capacity (FVC), FVC /lung weight decreased, peak expiratory flow rate (PEF) as well as maximal mid-expiratory flow rate (MMF) decreased in model group(P<0.05). What bleomycin caused was characterized by airway centered interstitial fibrosis. In the small airway, the shed epithelial cells and mucus plugs could been seen. The lung function of rats of SDS group had no difference with that of the healthy cats. Compared with the model group, FEV0.4/FVC% significantly increased (P<0.05) and the area ratio of interstitial fibrosis increased in the SDS+ lung fibrosis group. Conclusion Spleen deficiency syndrome is a potential adverse factor to aggravate restriction of ventilation function due to bleomycin.

    The pou1f1 target genes expression characteristic in pituitary gland of H22 tumor-bearing mice with different TCM patterns*

    Objective To study the expression of pituitary key functional genes and their pituitary-specific transcription factor 1 (Pou1fl) gene in H22 tumor-bearing mice with different TCM patterns. Methods 190 Kunming mice were injected subcutaneously into the armpit with H22 hepatoma carcinoma cells, and 60 more healthy mice were assigned into the normal group. The mice with pattern of exuberance of pathogenic toxin, asthenia of qi, asthenia of yang and qi, or asthenia of qi, yin and yang were identified by using the method of mice standardization of four diagnosis information and syndrome differentiation developed by our team. Then RNA was extracted from pituitary gland, hypothalamus and testis and was detected using GeneChip Mouse Exon 1.0 ST Array. And the genetic data of pituitary gland of different patterns were analyzed. Results Compared with normal group, the expression of pituitary key functional genes including Pomc, Gh and Prl were decreased by about 10% in mice with pattern of exuberance of pathogenic toxin or asthenia of qi, which was the pattern of early stage. At the middle or late stage, TCM patters were the pattern of asthenia of yang and qi, or asthenia of qi, yin and yang; Pomc, Gh, Prl and Tshb were upregulated by more than 30%. Pou1fl expressed at the highest level in pituitary tissue with the characteristic of tissue specific expression. Pou1f1 and Neurod1 were only upregulated in pattern of exuberance of pathogenic toxin, Tbx19 and Egr1 were upregulated in all patterns, which was just opposite to that of Nr4a1, Nr4a2, Pitx1, Gata2, Nr4a3, and Foxl2. Poulfl target genes including Crem, Tcf4, Rnf41, Lrpprc, Mrps18b, Eif3a, Lars, Arf4, Cep57, Vcpip1, Ptpn6, Myd88, F3, Lrrfip3 and Dixdc1 all expressed in the similar pattern with that of Poulfl, upregulated in pattern of exuberance of pathogenic toxin. Conclusion The abnormal expression of pituitary key functional genes was proved in the tumor-bearing mice with different TCM patterns. Pituitary-specific transcription factor 1 with its target genes expressed in the similar way in the pattern of exuberance of pathogenic toxin.

    Effect of Feixian Fang extract on mRNA expression of VEGF, VEGFR2, PAI-1 and VCAM-1 in microvascular endothelial cells of rats with pulmonary fibrosis: an in vitro study*

    Objective To investigate the effect of Feixian Fang extract (FXF) on the expression of vascular endothelial cell growth factor(VEGF), vascular endothelial cell growth factor receptor 2 (VEGFR 2), vascular cell adhesion molecule-1(PAI-1) andplasminogen activator inhibitor-1 (VCAM-1) in pulmonary microvascular endothelial cells in vitro, so as to explore the mechanism of intervention of FXF on microvascular angiogenesis of pulmonary fibrosis. Methods The lung microvascular endothelial cells which were isolated from pulmonary fibrosis model rats were equally divided into model group, losartan group, prednisone group, FXF high-, mid-, and low- dose group. The cells of model group were cultured in DMEM containing 20% fetal bovine serum, those of losartan group were cultured in DMEM containing 20% fetal bovine serum with 10 g/L losartan, those of prednisone group were cultured in DMEM containing 20% fetal bovine serum with 5 mg/L prednisone, and those of FXF groups were cultured in DMEM containing 20% fetal bovine serum with 100, 60, or 20 mg/L Feixian Fang extract. All the cells were cultured for 24 h. Then the mRNA expression of VEGF, VEGFR2, PAI-1 and VCAM-1 were detected by using method of double chain chimeric fluorescent dye SYBR Green real-time fluorescence quantitative detection. Results Campared with the model group, VEGF mRNA expression decreased significantly in all FXF group, while there was no difference of VEGFR2; PAI-1 mRNA was downregulated in FXF high-dose group; VCAM-1 mRNA expression decreased significantly in both FXF high-dose group and FXF low-dose group(P<0.05).Conclusion The effect of FXF against pulmonary fibrosis via the way of improving the blood coagulation and fibrinolysis leading to inhabitation of angiogenesis, due to downregulating the mRNA expression of VEGF, PAI-1 and VCAM-1 in the lung microvascular endothelial cells.

    Effect of Aaí on cyclic nucleotides, neurotransmitters and immune level of rats with asthenia pyrosyndrome*

    Objective To study the effect of frozen fruit pulp of Aaí (Euterpe Oleraceae Mart.) on cyclic nucleotides, neurotransmitters and immune level of rats with asthenia pyrosyndrome. Methods SD rats were randomly divided into blank group, model group, Huangbo (Cortex Phellodendri Chinensis) group, Gegen Qinlian Decoction (a Chinese herbal medicine compound) group, Aaí high-dose group and Aaí low-dose group. The rat model of asthenia pyrosyndrome was induced by intramuscular injection of dexamethasone sodium phosphate at dose of 0.35 mg/kg once daily for consecutive 21 days. The level of cyclic adenosine monophosphate (cAMP) and cyclic guanosine monophosphate (cGMP) were detected by radioimmunoassay. The levels of dopamine (DA), dopamine beta-hydroxylase (D-β-H) and norepinephrine (NE) were detected by enzyme immunoassay. The levels of IgG, IgM, C3 and C4 were detected by using colorimeter. The results of the groups were compared to analyze the effect of Huangbo, Gegen Qinlian Decoction and Aaí on rats with asthenia pyrosyndrome. Results High dose Aaí could relieve the symptoms due to asthenia pyrosyndrome, and decreased the serum level of cAMP, cGMP, cAMP/cGMP, DA, D-β-H and NE (P<0.05), while increased the serum content of IgG, IgM and C3 (P<0.05,P<0.001 or P<0.05). Conclusion The effect of Aaí on cyclic nucleotides, neurotransmitters and immune level was similar with that of Huangbo or Gegen Qinlian Decotion, which proved the cold property of Aaí.

    Dynamic changes of chemical compositions in volatile oil extracted from Menthae Haplocalycis Herba during different harvest periods using HS/SPME-GC-MS*

    Objective To determine the chemical compositions of volatile oil extracted from Menthae Haplocalycis Herba (mint) collected during different harvest periods by using headspace solid phase micro-extraction coupled with gas chromatography-mass spectrometry (HS/SPME-GC-MS). Methods The volatile oils were extracted by the method of headspace solid phase micro-extraction, then the chemical compositions were determined by using GC-MS and quantified by the area normalization method. Results A total of 47 compositions were detected in Menthae Haplocalycis Herba in mature stage, and 12 of those had been identified with the total content to be 78.23%. By comparing with the relative amount of menthone, l-limonene and l-menthol extracted from the herbal medicine samples collected during different harvest periods, which are the key components in biosynthesis pathway of the herb, the preliminary determination of the best time to harvest was September. Conclusion The effective and rapid method for the determination of volatile oils provided basis for studying the dynamic changes of the chemical compositions of volatile oil extracted from Menthae Haplocalycis Herba and the proper time to harvest.

    Study on chemical constituents in Cordyceps hawkesii by UPLC-Q-TOF-MS*

    Objective To analyze and identify the main chemical constituents of extract from Cordyceps hawkesii by using method of ultra performance liquid chromatography-qudropole time-of-flight mass spectrometry (UPLC-Q-TOF-MS). Methods UPLC condition was as the following: Agilent SB-C18 (50 mm×4.6 mm, 1.8 μm) column with gradient elution consisting of 0.1 % formic acid in water and acetonitrile, at a flow rate of 0.25 mL/min. The column temperature was set at 30℃. MS analysis was based on information associated mode (IDA) that positive and negative ions were respectively collected. Results Mass spectrometry combined with “Formula Finder” of PeakView software and database, a total 26 compounds were identified as alkaloids, glycosides, nucleosides, and amino acids. 9 of them had never been reported in Cordyceps hawkesii, and 2 of them were known. Conclusion The method is accurate, reliable and efficient for rapid identification of compounds in Cordyceps hawkesii. The results provide references for the development and utilization of Cordyceps hawkesii, as well as finding out its pharmacodynamic material basis.

    Optimization of processing technology of honey barked Hedysari Radix using orthogonal design method*

    ObjectiveTo optimize the processing technology of honey baked Hedysari Radix, so as to provide the experimental basis for the quality control of Hedysari Radix processed products and standardization of the processing. Methods L9 (34) orthogonal design method was used, and the mean content of calycosin and formononetin which were detected by HPLC with gradient elution were taken as the comprehensive evaluation indexes, the processing technology was optimized. Results The optimum processing technology of honey baked Hedysari Radix was thickness of 3 cm at the temperature of 70 ℃ for 2.5 h.Conclusion Hedysari Radix baking processed products produced by the optimum technology met with the requirement of Chinese Pharmacopoeia (2015 Edition), which was simple, feasible, and repeatable. The method would provide evidence for further research on optimum processing technology and for standardiztion of industrial production.

    Clinical effect of traditional Chinese medicine therapy on perimenopausal insomnia from the standpoint of regulating liver and kidney:a Meta-analysis*

    Objective To systematically evaluate the clinical efficacy and safety of the traditional Chinese medicine(TCM) therapy on perimenopausal insomnia from the standpoint of regulating liver and kidney. Methods Randomized controlled trials(RCT) on PubMed, EMbase, CNKI, Wanfang Database and VIP database from their inception to April 2016 were searched. Two researchers separately screened the literature, extracted data and evaluated the literature bias risk according to inclusive criteria and exclusive criteria. Then the Meta-analysis was operated using Rev Man 5.3 software. Results Altogether nine RCT with 678 patients were eligible. The result of Meta-analysis showed: the clinical effectiveness of experimental group (TCM therapy group) was superior to that of control group(RR=1.30,95%CI[1.11,1.52],P<0.05); the PSQI scores showed significant differences between the two groups (SMD=-1.16,95%CI[-2.12,-0.20],P<0.05),but SPIEGEL score did not. Although adverse reactions were appeared in five studies, the adverse reaction rate of the experimental group was lower significantly (RR=0.09,95%CI[0.04,0.22],P<0.05). Conclusion The experimental group achieved obvious clinical efficacy with less adverse effect.
