    Application of theory of life gate in hypertension treatment*

    In order to illustrate the application principles and methods of the theory of life gate in hypertension treatment, following opinions were presented through analyzing ancient literature and summarizing clinical experience. Hypertension, in early stage, is related to excited state in the aspects of physiological function, pathological changes and pathogenesis, while the excited state can be induced by life gate deficiency failing to control fire in the body. The life gate deficiency failing to control fire in the body is divided into deficiency of genuine yin leading to frenetic stirring of the ministerial fire and floating upward of life gate brontosaurus fire. The former can be treated with Liuwei Dihuang Wan for nourishing life gate, inhibiting fire and guiding water, and the latter can be treated with Bawei Wan for warm-tonifying life gate and conducting fire back to its origin. And purifying one’s heart and limiting one’s desires as well as cherishing and nourishing one’s genuine yin and yang really make sense in prevention.

    On theoretical thought of assistant medications*

    The theory of assistant medications, proposed by Professor WANG Xinlu, is a new idea for the innovation of TCM theory under the environment of modern science and technology. The evolution, connotation, specific content and future direction and significance of assistant medications and theory of assistant medications have been systematically expounded. The direction of further research on theory of assistant medications has been pointed out in order to develop TCM basic theory, and obtain legal support in Chinese clinical medicine.

    Mechanism of Ganmai Dazao Tang in treatment hysteria based on principle of correspondence between formula-syndrome factors*

    Ganmai Dazao Tang is a famous formula for hysteria in Jinguiyaolue(Synopsis of the Golden Chamber). The connotation of hysteria has been explored in the paper through summing up relative knowledge of medical practitioners in the successive dynasties. The pathogenesis of hysteria lies in function disorder of five viscera resulting in that five viscera fail to restrain their spirits, viscera spirits float and heart spirit is uneasy. Hysteria reflects the clinical pathological features of estrangement among qi, blood, yin and yang, and viscera spirits failing to be restrained and floating in the exterior. The combining and medicating laws of Ganmai Dazao Tang, and pathological features hysteria have been further analyzed according to principle of correspondence between formula-syndrome factors. The pharmacodynamic material basis of Ganmai Dazao Tang has been analyzed and the opinion of placebo efficacy has been negated according to the progresses in modern pharmacological research and clinical research. The combination of Ganmai Dazao Tang is rigorous, and similar to Guizhi Tang, the first one of all TCM formulas. These two formulas both are based on regulating yin and yang and widely applied in the clinic with significant curative effects.

    On statement of “if keeping health, Zusanli (ST36) frequently being given festering moxibustion”*

    This article has analyzed the source and clinical significance of statement of “if keeping health, Zusanli (ST36) frequently being given moxibustion” based on philology and medical principles. The statement was recorded in Suosuilu (Trivial Details of Traditional Chinese Medicine) written in early Southern Song Dynasty, which was originally designed for preventing and curing wind diseases. The moxibustion on ST36 is to diffuse and unblock qi and blood of zang-fu organs, but is not to give festering moxibustion. If the statement is explained as repeated festering moxibustion, Yubaizhuang moxibution recorded in Beijiqianjinyaofang (Essential Prescriptions Worth a Thousand Gold for Emergencies) and Bao moxibution recorded in Zhenjiuzishengjing (Classic of Nourishing Life with Acupuncture and Moxibustion) used only in the condition of excessive yang deficiency will be misunderstood. Acupressure, frottage and foot bath can be selected in ST36 healthcare. Considering safety, some mild moxibustion therapies are recommended, such as suspended moxibustion and sandwiched moxibustion, in patients with obvious cold symptoms.

    Influence of Yiqi Huoxue Fang on Akt/mTOR signaling pathway in rats with myocardial ischemia of myocardial infarction*

    Objective To investigate the myocardial expressions of protein kinase B (Akt)/ mammalian target of rapamycin (mTOR) signaling pathway and effective mechanism of Yiqi Huoxue Fang depending on model of myocardial ischemia. Methods The model of acute myocardial infarction (AMI) was established through ligation of left coronary anterior descending branch in SD rats, and randomly divided into model group, perindopril group, Qishen Yiqi Gutta Pills group, Yiqi Huoxue Fang group and sham-operation group, and all groups were given orally corresponding medications respectively 2 d after operation. The observation time points were set on the 7th d and 28th d. The changes of heart function and structure were detected by using ultrasonic testing, changes of myocardial pathomorphology were observed after HE staining, and expressions of Akt, mTOR, p-Akt and p-mTOR in infarction marginal zone were detected by using Western blot test, real-time fluorescence quantitative polymerase chain reaction (RT-PCR) and immunohistochemistry technique in all groups. Results The level of left ventricular ejection fraction (LVEF) and left ventricular fraction shortening (LVFS) decreased significantly in model group compared with sham-operation group on the 7th d and 28th d (P<0.01). The level of left ventricular end-systolic volume (LVESV) increased significantly on the 7th d (P<0.01), and levels of LVESV and left ventricular end-diastolic volume (LVEDV) increased significantly (P<0.01) on the 28th d in model group. The levels of LVEF and LVFS increased and levels of LVEDV and LVESV decreased significantly in all medicated groups compared with model group (P<0.01). The thinner ventricular wall, less cardiomyocytes, disorder myocardial cells arrangement and inflammatory cell infiltration were observed in infarction marginal zone in model group, and disorder myocardial cells arrangement and less cardiomyocytes were observed in all medicated groups. The ratio of p-Akt/Akt and p-mTOR/mTOR increased (P<0.05), expressions of Akt m-RNA and mTOR m-RNA increased significantly (P<0.01), and levels of p-Akt and p-mTOR positive cell rates increased significantly (P<0.01) in model group. The ratio of p-Akt/Akt and p-mTOR/mTOR increased (P<0.05), expressions of Akt m-RNA and mTOR m-RNA increased (P<0.05 or P<0.01), and levels of p-Akt and p-mTOR positive cell rates increased significantly (P<0.05) in all medicated groups. Conclusion Yiqi Huoxue Fang can improve heart function and protect myocardial cells through activating cardiac self-protective mechanism-Akt/mTOR signaling pathway in rats with myocardial infarction.

    Influence of medicated cerebrospinal fluid of modified Sanjia San on relative factors of BV2 microglia cell inflammatory model and protein expressions induced by lipopolysaccharide*

    Objective To investigate the influence of medicated cerebrospinal fluid of modified Sanjia San on inflammatory reactions of BV2 microglia cells induced by lipopolysaccharide (LPS). Methods BV2 microglia cells were divided into blank control group, model group, three modified Sanjia San groups and huperzine group. After the cells cultured and adhered to wall, modified Sanjia San groups and huperzine group were, respectively, given medicated cerebrospinal fluid (10.0%, 5.0%, 2.5%, 10.0%) and incubated at 37 ℃ for 2 h. Except of blank control group, all groups were given LPS (1 mg/L as final concentration) and incubated continuously for 24 h, and then supernatant was collected for extracting protein samples. The content of nitric oxide (NO) in supernatant was detected by using nitrate reductase method in all groups. The content of inducible nitric oxide synthase (NOS), interleukin-1β (IL-1β) and tumor necrosis factor-α (TNF-α) in supernatant were detected by using enzyme-linked immunosorbent assay (ELISA) in all groups. The expressions of c-Jun N-terminal kinase (JNK) and phospho-c-Jun N-terminal kinase (p-JNK) were detected by using Western blot test in all groups. Results The content of supernatant NO, iNOS, IL-1β and TNF-α increased significantly, and expression of p-JNK was up-regulated in model group. The content of supernatant NO, iNOS, IL-1β and TNF-α decreased significantly, and expression of p-JNK was down-regulated in 10.0% and 5.0% modified Sanjia San groups. Conclusion The medicated cerebrospinal fluid of modified Sanjia San has the regulative effect on inflammatory reactions of BV2 microglia cells induced by LPS, and the mechanism may be related to the controlling of p-JNK expression and reducing of generation of inflammatory media.

    Regulating effect of Zuogui Jiangtang Jieyu Fang on hippocampus neuron synaptic plasticity related proteins-CaMKⅡ and SynGap in simulated condition of diabetes complicating depression*

    Objective To investigate the influence of Zuogui Jiangtang Jieyu Fang (ZJJF) on hippocampus neuron synaptic plasticity related proteins-CaMKⅡ and SynGap in rats simulated diabetes complicating depression DD. Methods The hippocampus neurons were primitively isolated, purified and cultured in SD rats. The DD model was established by applying intervention of high glucose and cortisone. The cultured hippocampus neurons were randomly divided into normal group, model group, medicated serum of ZJJF (ZJJF group), medicated serum of positive drug (fluoxetine and metform) group (positive drug group) and blank group. The damages of hippocampus neurons were detected by using Nissl’s staining in all groups, and expressions of CaMKII and SynGap were detected by using immunofluorescence staining and high-content analysis (HCA) after drug intervervention for 18 d. Results Compared with normal group and blank group, the quantity of Nissl bodies decreased significantly, neural network and dentritic spines ruptured or disappeared, and expressions of CaMKII and SynGap were significantly down-regulated in model group (P<0.01). Compared with model group, the quantity of Nissl bodies, neural network and dentritic spines, and synaptic connection were significantly recovered, and expressions of CaMKII and SynGap were un-regulated in positive drug group and ZJJF group (P<0.01 or P<0.05). Conclusion ZJJF had significant regulative effect on CaMKII and SynGap in simulated condition of diabetes complicating depression, which may be one of important mechanisms of ZJJF antagonizing cellular damage and neural degeneration.

    Influence of Mongolia medicine-Garidi San on Th1 and Th17 in rats with ulcerative colitis*

    Objective To discuss the influence of Mongolia medicine-Garidi San on helper T cell 1 (Th1) and helper T cell 17 (Th17) in rats with ulcerative colitis (UC). Methods All rats were randomly divided into normal group, model group, low-dose Garidi San group (low-dose group), mid-dose Garidi San group (mid-dose group), high-dose Garidi San group (high-dose group), sulfasalazine group (SSZ group) and Bupi Yichang Wan group. The UC model was induced and established by using immune method and TNBS. Garidi San was orally given to, respectively, low-dose group (1.76 g/kg), mid-dose group (3.51 g/kg) and high-dose group (7.02 g/kg), SSZ was given to SSZ group (3.60 g/kg), and Bupi Yichang Wan was given to Bupi Yichang Wan group (1.62 g/kg). After 4 weeks the samples of peripheral blood were collected from all groups. The ratio of Th1 to Th17 (Th1/Th17) in peripheral blood was observed by using flow cytometry, and levels of PB transforming growth factor β (TGF-β), interleukin-6 (IL-6), interleukin-17 (IL-17) and interferon-γ (IFN-γ) were detected by using enzyme-linked immunosorbent assay (ELISA). Results The level of TGF-β was decreased significantly in mid-dose group, low-dose group and SSZ group. The levels of serum IL-6 and IL-17 were decreased significantly(P<0.05), level of IFN-γ was increased (P<0.05), and Th1/Th17 was increased (P<0.05) in all medication groups. Conclusion Garidi San can regulate the differentiation direction of helper T cell 0, and recover the balance of Th1/Th17 in UC rats, which may be one of mechanisms of Garidi San curing UC.

    Inhibitory effect of astragaloside IV on PC12 apoptosis induced by oxygen-glucose deprivation/oxygen-glucose rehabilitation through activating autophagy*

    Objective To study the inhibitory effect of astragaloside IV (AST) on the apoptosis of PC12 cells induced by oxygen-glucose deprivation/oxygen-glucose rehabilitation (OGD/OGR) through activating autophagy. Methods PC12 cells in logarithmic phase were collected and randomly divided into normal group, OGD/OGR group (model group), AST solvent control group (DMSO group), AST group, AST+autophagy inhibitor group (AST+3-MA group), 3-MA group and autophagy activator group (RAPA group). Except of normal group, all other groups were given treating of OGD for 3 h and then OGR for 12 h and simultaneous medication treating. The morphological changes of autophagosomes were observed by using transmission electron microscope (TEM), expressions of LC3II, Beclin1 and Caspase-3 were detected by using Western blotting, and apoptosis of PC12 cells were detected by using flow cytometry after TUNEL staining. Results Compared with normal group, the typical autophagosomes were observed, expressions of LC3II, Beclin1 and Caspase3 increased, and apoptosis rate and apoptosis increased (P<0.05). Compared with model group, the quantity of autophagosomes and expressions of LC3II and Beclin1 increased (P<0.05), and expression of Caspase3 and apoptosis rate and apoptosis decreased in AST group and RAPA group (P<0.05). Compared with model group, the quantity of autophagosomes and expressions of LC3II and Beclin1 decreased (P<0.05), and expression of Caspase3 and apoptosis rate and apoptosis increased in 3-MA group (P<0.05), while DMSO group and model group had no significant difference (P>0.05). Compared with AST group, the quantity of autophagosomes and expressions of LC3II and Beclin1 decreased (P<0.05), and expression of Caspase3 and apoptosis rate and apoptosis increased in AST+3-MA group and 3-MA group (P<0.05). Conclusion AST play a neuroprotective role through activating autophagy and inhibiting apoptosis of PC12 cells induced by OGD/OGR.

    Mechanism of Ningxin Anshen Fang in regulation of unbalanced Glu/GABA-Gln metabolic loop in insomnia rats*

    Objective To study the therapeutic effect of Ningxin Anshen Fang on insomnia, and regulative mechanism to the metabolic loop of glutamic acid (Glu)/γ-aminobutyric acid (GABA)-glutamine (Gln). Methods The model of insomnia was established by using modified multiple water platform sleep deprivation method in rats, and then all rats were randomly divided into normal group, model group, diazepam group, Ningxin Anshen Fang low-dose group (low-dose group), Ningxin Anshen Fang mid-dose group (mid-dose group) and Ningxin Anshen Fang high-dose group (high-dose group). The general conditions were observed, sleep latency and sleep duration were observed by applying pentobarbital sodium (PS) cooperative sleep test, and model and curative effect of Ningxin Anshen Fang on insomnia were reviewed. The content of Glu and GABA in brainstem and hypothalamus was detected by using high performance liquid chromatography (HPLC). The expressions of glutamic acid decarboxylase (GAD) and astrocyte glutamine synthetase (GS) in brainstem and hypothalamus were detected by using Western blot experiment. Results The sleep latency was significantly shortened, sleep duration was prolonged, GABA level in brainstem and hypothalamus was increased and ratio of Glu to GABA (Glu/GABA) was decreased by Ningxin Anshen Fang in insomnia rats. The expression of GAD in brainstem and hypothalamus was significantly promoted in all Ningxin Anshen Fang groups. Conclusion Ningxin Anshen Fang can affect the synthesis and decomposition Glu and GABA and expressions of Glu and GABA, and normalize the excitotary/inhibitory metabolic imbalance of Glu/GABA through regulating Glu/GABA-Gln metabolic loop, which may be related to that Ningxin Anshen Fang has the curative effects of calming heart and tranquillizing mind, and sedation and hypnosis.

    Relevant analysis on quality representation based on nucleosides in Aurea helianthus*

    Objective To establish a method of high performance liquid chromatography (HPLC) for simultaneously determining of 7 nucleoside (uracil, cytidine, uridine, adenine, guanosine, adenosine and 2'-deoxyadenosine) in Aurea helianthus, and conduct the relevant analysis and review on quality of Aurea helianthus. Methods HPLC-PDA was applied for determining 7 nucleosides in Aurea helianthus. The quality representation of 11 batches of Aurea helianthus was carried out and representation results were given relevant analysis based on the content of index components and index component adducts from 7 nucleosides. Results The linear relations of 7 nucleosides were good, and precision, stability, repeatability and sample recoveries were all accorded with the requirements of methodology determination. The content of 7 nucleosides in 11 batches of Aurea helianthus was determined by using the established method. Taking batch lot 1 of Aurea helianthus as criterion material, the content of index components was higher in batch lot 2, 6, 9, 10 and 4, and relevancy was the highest among criterion Aurea helianthus and batch lot 6, 9, 8, 4, 3 and 11. The comprehensive review showed that excellent degrees of batch lot 1, 2, 6, 9 and 4 were in the lead. Conclusion The established method for determining content of nucleosides in Aurea helianthus and correlation analysis mode of quality representation can analyze overall quality relevancy and application validity, which provides the reliable evidence for quality control and review of Aurea helianthus.

    Systematic review and network Meta-analysis on effectiveness of acupuncture-moxibustion in treatment of optic atrophy*

    Objective To analyze the curative effect of acupuncture-moxibustion on optic atrophy by applying network Meta-analysis model based on frequency-statistic data filling strategy. Methods The databases of Cochrane Library, PubMed, EMbase, AMED, Nature, Science Online, WorldSciNet, CBM, CNKI and WanFang Database were retrieved with computer for collecting RCT/CCT literature from database establishing time to Jun. 2016. The review on quality and bias risk of every included study were conducted by 2 researchers according to 5.1.0 standard in Cochrane Handbook. Results There were totally 16 studies involving 1 369 samples included. The results of network Meta-analysis showed that the possibility of the best curative effect of each therapy was respectively as follows: Western drug (10.5%), acupuncture (5.9%), acupuncture+moxibustion (4.4%), ear acupoints+ Chinese medications (4.7%), electro-acupuncture (8.0%), acupuncture+Chinese and Western medications (4.9%), moxibustion+Western medications (3.2%), electro-acupuncture+ Chinese and Western medications (4.3%), Chinese and Western medications (10.1%), Chinese medications (6.7%) and acupuncture+Western medications (3.4%). The results indicated that the curative effect of Western medications was the best. Acupuncture+Western medications had advantages compared with only Western medications, and difference in odds ratio had statistical significance. In terms of safety, the safety evaluation could not be confirmed because none of the included literature described the occurrence of adverse events. The modern medical studies on the mechanism of acupuncture in treatment of optic atrophy were relatively less, so it was unable to judge the modern medical mechanism. Conclusion Western medications, Chinese and Western medications and electro-acupuncture may be the best therapies for optic atrophy, but the exact conclusion still needs to be verified further by high-quality clinical researches.

    Clinical effect of Pingchan Yisui Fang on Parkinson’s disease complicated with mild cognitive impairment of type of marrow sea deficiency*

    Objective To observe the clinical efficacy of Pingchan Yisui Fang on Parkinson’s disease complicated with mild cognitive impairment (PD-MCI) of type of marrow sea deficiency. Methods The patients with Parkinson’s disease complicated with mild cognitive impairment (PD-MCI) of type of marrow sea deficiency (n=103) were randomly divided into control group and Chinese medicine group (CM group), and control group was given basic treatment of Western medicine and CM group was additionally given Pingchan Yisui Fang for 36 weeks. The curative effect was reviewed by using Unified Parkinson’s Disease Rating Scale (UPDRS), Montreal Cognitive Assessment Scale (MoCA) and Minimum Mental State Examination (MMSE) before and after treatment. Results The scores of UPDRS were significantly lower (P<0.05), and scores of MoCA and MMSE were significantly higher (P<0.01) in CM group than those in control group after treatment. The factor scores of attention, abstract ability, delayed recall and orientation in MoCA, and orientation, memory ability, attention and recall ability in MMSE were significantly higher in CM group than those in control group (P<0.05 or P<0.01) after treatment. Conclusion Pingchan Yisui Fang can effectively relieve motor symptoms and cognitive disorder on the base of standardized treatment of Western medicine in patients with PD-MCI.
