    On pathogenic theory of “speculating nosopoietic factors according to symptoms and signs”*

    Qian Huang highly summarized pathogenic laws of disease, disease invasion due to external pathogen, in traditional Chinese medicine (TCM) with sixteen Chinese words, including “speculating nosopoietic factors according to symptoms and signs”, and later doctors have summed up the thinking process of speculating nosopoietic factors, acquiring pathogenesis and giving treatment according to symptoms and signs. The essence of “speculating nosopoietic factors according to symptoms and signs” actually has 3 procedures: speculating disease causes according to patterns, speculating pathogenesis according to disease causes, and giving treatment after investigating pathogenesis. Disease invasion is predicted according to disease causes and pathogenesis, disease causes and pathogenesis are understood from disease invasion, and treatment is given after investigating pathogenesis. In clinical practice, modern science and technology and modern differentiation pattern system are combined to predict and give interventional treatment to the diseases without showing clinical symptoms. “Speculating nosopoietic factors according to symptoms and signs” can be also applied to understand disease transmission and transmutation, and guide to know disease prognosis and outcome.

    Exploration on formulas in Shanghan Lun (Treatise on Cold Damage Diseases)-theory of seven formula types*

    The laws of constructing formulas of “Da, Xiao, Huan, Ji, Qi, Ou, Fu” were explained early in Huangdi Neijing (the Yellow Emperor’s Internal Classic). The laws of medicinal combinations in Shanghan Lun (Treatise on Cold Damage Diseases) are discussed based on theory of seven formula types, and there were totally 30 typical formula-related patterns from Shanghan Lun expounded in the paper. After analyzing and summarizing seven formula types, it is found that the formulas in Shanghan Lun reflected the core content of theory of seven formula types, and the medicinal combinations is in accordance with those in Huangdi Neijing. The theory of seven formula types is proved by formulas in Shanghan Lun.

    Objectization of conceptual and state of affairs of spatiotemporal sequence by qi’s reference in traditional Chinese medicine*

    As the initial word of traditional Chinese medicine (TCM) and traditional philosophy, qi is a core term in TCM practice. To provide a perspective and point of view for the establishment of modernized discourse system of TCM theory, the conceptual connotation of qi cognition is explored. Through phenomenological reduction and analytic philosophical wordings, the intentionality analysis on the cognitive content and experience process of qi, and the situational analysis on the meaning, form and objects of qi’s word reference are conducted, respectively, and other methods under the guidance of dialectical materialism are applied. The cognitive mode of qi-image-fact-truth corresponding to word-meaning-referent is expounded in traditional thinking after systematic analysis. The grasp of state of affairs of spatiotemporal sequence (SASTS) by qi manifests as organizational forms of phenomenon flow and spatial experiential flow, and thus forms object targets, such as basic state of affairs (BSA) and complex state of affairs (CSA). The form of spatiotemporal sequence of CSA is considered to be the entity that exists as the essence of qi understanding, and comes into being the referent of words. Thus, the conceptual reference of qi, with SASTS as connotation, makes that the terms related to qi present a hierarchical epitaxial system under the same perspective, and provide a new way for the theoretical research of TCM.

    Historical evolution of formula-related patterns of Banxia Shumi Tang (Pinellia T uber and Husked Sorghum Decoction)*

    The correspondence between formulas and patterns is a practical therapeutic method in traditional Chinese medicine (TCM) in clinic practice. It has a guidance value to study on the evolution of Banxia Shumi Tang (Pinellia Tuber and Husked Sorghum Decoction) in later generations in making a clear view on development history of TCM formula-related patterns, grasping the rules of this evolution, and improving inheritance and innovation of TCM theory. There is a clear rout of the changes in origin and development of Banxia Shumi Tang, which is a progressive development on the foundation of previous evolution. The changes in the composition of Banxia Shumi Tang have showed that herbal medicinals increased from the Jin Dynasty to the Ming Dynasty, and returned to the original formula recreation during the Qing Dynasty. The indications of this formula have also changed and expanded, from simple insomnia to a variety of sleep disorders and to mental discomfort, which reveals that the formula corresponds to pattern development. The knowledge on pathogenesis has also been continuously enriched, from yang failing to enter Yin to the pathogenesis changes of gallbladder and the middle energizer. That reveals the development law of TCM academy based on the characteristics of formula-related patterns utilization. The perspective on the pathogenesis of Banxia Shumi Tang has also been enriched and gradually refined.

    A textual research on formula name of Didang Tang (Resistance Decoction) in Shanghan Lun (Treatise on Cold Damage Diseases)*

    A textual research on formula name of Didang Tang (Resistance Decoction) in Shanghan Lun (Treatise on Cold Damage Diseases) indicates that the current versions of Suwen (Plain Questions), Shanghan Lun and Qianjin Yaofang (Essential Prescriptions Worth a Thousand Gold) have been revised by scholars in the Song Dynasty. The textual materials of these works deviate original versions in varying degrees, and have obvious Song’s medical characteristics. There are differences in content among these works. The complexity and uncertainty of these textual materials are speculated to be the result of the differences in styles, principles and emphases during revision procedure of these works by scholars in the Song Dynasty, or complex spreading process of them for a very long time and repeatedly reprinting of them. On the core concepts of theory, principles, formulas and medicinal in Chinese medicine, especially on those with ambiguities and without consensus through the ages, the textual research should be conducted on a firm grasp of different texts instead of relying on only one text.

    Bencaojing Kaozhu (Annotation on Shennong’s Classic of Materia Medica) and Zhiyuan Congkao (A Series of Notes to Zhiyuan) were important works written by Mori Tachiyuki who was a well-known textual researcher of traditional Chinese medicine (TCM) during the Japanese Edo Period. Through carding the textual research on disease pattern terms in these two books, Mori Tachiyuki’s methodology and achievements in textology of disease pattern terms can be known. It has provided references for scholars to understand correctly disease pattern terms of TCM.

    Effects and mechanism of Luotuopeng’s aqueous extract on the high glucose induced-retinal vascular endothelial cells*

    Objective To investigate the effects of Luotuopeng (Herb of Common Peganum, Peganum hannal L, LTP) aqueous extract on the high glucose induced-retinal vascular endothelial cells and the molecular mechanism. Methods The retinal vascular endothelial cells, were divided into 5 groups: normal group (5.5 mmol/L glucose), model group (25 mmol/L glucose), high-dose LTP group (25 mmol/L glucose plus 250 mg/L LTP solution), mid-dose LTP group (25 mmol/L glucose plus 200 mg/L LTP solution), and low-dose LTP group (25 mmol/L glucose plus 150 mg/L LTP solution). After 3 days of incubation, cell viability was determined with CCK-8 assay. The transwell assay was performed to observe the amount of cell migration. Western Blot assay was used to measure the protein expression level of TEAD 1 and Lats 1. Results CCK-8 assay showed that compared with the normal group, the model group had promoted the cell viability of RF/6A (retinal vascular endothelial cells). In comparison with the model group, LTP’s aqueous extract at different doses had all caused a lower cell viability. Transwell Resultsproved that compared with the normal group, the model group had facilitated the ability of migration of RF/6A. In comparison to the model group, LTP’s aqueous extract at different doses had significantly inhibited the migration ability of RF/6A (P<0.05). Western Blot assay indicated that compared with the normal group, the model group had showed a lower expression level of TEAD1 and a higher expression level of Lats1. In comparison to the normal group, LTP’s aqueous extract at different doses had decreased the expression of TEAD1 and increased the expression level of Lats1 (P<0.05). Conclusion LTP’s aqueous extract can inhibit the cell viability and migration of retinal vascular endothelial cells by regulating the Hippo signal pathway, which accounts for its regulatory effects on retinal vascular endothelial cells.

    Resolving depression effects of artistic work on depressed college students: a analysis based on EEG α-band relative power*

    Objective To analyze the resolving depression effects of artistic work on depressed college students through detecting changes of EEG α-band relative power in depressed college students received or not received artistic work training. Methods The depressed college students (n=24) were divided, according to 1∶1 principle, into test group and control group, and the test group was given artistic work training and control group was not given any interventions. The changes of EEG α-band relative power were collected, compared and analyzed from 2 groups before the experiment and after the training. The collective period was divided into 7 phases: resting state 0, artistic work 1, resting state 1, artistic work 2, resting state2, artistic wark 3, resting state 3. Results The leads of FZ, F8, FT7, FCZ, FT8, C3, CZ, CPZ, TP8, P4, T5, P3, PZ, T6 and O1 showed significant difference between 2 groups at resting state 0 (P<0.05). The leads of FZ, FT7, FCZ, FT8, CZ, TP7, T5, PZ, CPZ, TP8, P3, P4, T6 and O1 showed significant difference between 2 groups in artistic work 1 (P<0.05). The leads of FP2, FC3, FT8, FT7, FCZ, FC4, TP7, P4, T6, T3, CP3, TP8, P3 and O2 showed significant difference between 2 groups in artistic work 2 (P<0.05). The leads of FP2, FT7, FCZ, FT8, F3, CZ, O1, TP7, TP8 and P3 showed significant difference between 2 groups in artistic work 3 (P<0.05). The leads of FCZ, FT8, TP7, TP8 and P3 showed significant difference between 2 groups in the whole operation of artistic work (P<0.05). Conclusion Artistic work can induce sexual responses of EEG α-band, shows a higher α-band relative power in metencephalon and parietal regions, and has some relieving effect on depression status.

    Regulating effect of Zuogui Jiangtang Jieyu Fang on hippocampal NVU: a study based on diabetes complicated by depression*

    Objective To discuss the influence of glutamic acid (Glu) imbalance on protein expressions of GluR2 and Beclin-1 in hippocampal neurovascular unit (NVU) in status of diabetes complicated by depression (DD), and interventional effect of Zuogui Jiangtang Jieyu Fang (Zuogui Blood Sugar-decreasing Depression-relieving Decoction, ZJJF). Methods The hippocampal neurons, astrocytes (Ast) and brain microvascular endothelial cells were primitively isolated, purified and cultured in SD rats to establish in vitro co-cultural system of hippocampal NVU. The immunocytochemistic staining method was used to identify cells, and high glucose (150 mmol/L) combined with corticosterone (CORT, 200 μmol/L) were used to intervene establishment of hippocampal NVU model. The non-modeled cells were randomly divided into normal group and blank group, and modeled cells were divided into model group, group of medicated serum of Zuogui Jiangtang Jieyu Fang (ZJJF group) and group of medicated serum of positive drugs [positive drug group, fluoxetine (0.18 g/kg)+metform (1.8 mg/kg)]. The normal group and model group were given the same amount of nutrient solution, and other groups were given medicated serum or blank serum in corresponding volume fraction of 10%. Glu content in supernatant of hippocampal NVU neurons was detected by using enzyme-linked immunosorbent assay (ELISA) in all groups. The protein expressions of GluR2 and Beclin-1 in hippocampal neurons were detected by using immunofluorescent staining and high content analysis (HCA) in all groups. The apoptosis of hippocampal neurons was detected by using Tunel staining in all groups. Results Compared with normal group and blank group, Glu content in supernatant of hippocampal NVU neurons increased significantly (P<0.01), GluR2 expression decreased significantly (P<0.01), Beclin-1 expression increased significantly (P<0.01), and apoptosis rate increased significantly in model group. Compared with model group, Glu content decreased significantly (P<0.01), GluR2 expression was up-regulated (P<0.01), Beclin-1 expression was down-regulated (P<0.01 or P<0.05), and apoptosis rate decreased significantly in positive drug group and ZJJF group. Conclusion ZJJF has significant regulation effect on the protein expressions of neuron GluR2 and Beclin-1 in hippocampal NVU model, which may be related to the relief of Glu imbalance. It may be an important mechanism of ZJJF inhibiting autophagy and antagonizing hippocampal neuron apoptosis of DD.

    Influence of water decoction of Huangqi on immune function in rats with deficiency cold pattern: a study based on transcriptome sequencing technique*

    Objective To investigate the molecular mechanism of occurrence of deficiency cold pattern and therapeutic effect of water decoction of Huangqi (Milkvetch Root, Radix Astragali seu Hedysari) on deficiency cold pattern. Methods The traditional Chinese medical compound [Sheng Shigao (raw Gypsum, Gypsum Fibrosum), Longdancao (Gentian Root, Radix Gentianae), Huangbai (Amur Cork-Tree, Cortex Phellodendri), Zhimu (Common Anemarrhena Rhizome, Rhizoma Anemarrhenae), in a ratio of 2∶1.2∶1∶1.5] was intragastrically given to rats [dosage=16 g/(kg·d), and dose=4 mL/(kg·d)] for 14 d for establishing rat model of deficiency cold pattern. The modeled rats were randomly divided into model group(deficiency model) and treatment group [treated with water decoction of Huangqi, dose=5.40 g/(kg·d), each n=12] by using SPSS software, and other rats (n=12) were chosen into blank group. The expressions of immune-related genes in liver were detected by using transcriptome sequencing technique. The differentially expressed genes (DEG) were screened, and immune-related genes were annotated according to GO functional standard. The Results of transcriptome sequencing were verified by using fluorescence quantitative polymerase chain reaction (PCR). Results Compared with blank group, there were 323 differential genes screened in model group. Compared with model group, there were 333 differential genes screened in treatment group, and they mainly involved the functions of stimulus response and defense response. The Results of GO functional analysis showed that model group had down-regulated expressions of multiple immune-related genes with stimulus response and defense response as representations, and immune response function was impacted and body immunity was impaired. After treatment with water decoction of Huangqi, the immune response of body to environment was improved and body immune status was recovered and enhanced through up-regulating the expressions of genes related to stimulus response, defense response and immune response. Conclusion The molecular mechanism of occurrence of deficiency cold pattern and therapeutic effect of water decoction of Huangqi may closely related to body immune function and expressions of relative genes.

    Influence of surrounding-hoop method of Modified Ruyi jinhuangsan Tincture Tincture, under guidance of huchang theory, on expressions of CD68 and myeloperoxidas in rats with skin abscess*

    Objective To observe the influence of surrounding-hoop method of Modified Ruyi jinhuangsan Tincture Tincture, under guidance of huchang theory, on expressions of macrophage markers (CD68) and myeloperoxidas (MPO) in rats with skin abscess. Methods All rats (n=50) were randomly divided into blank group, pretreatment group, treatment group, control group and model group. Except of blank group, other 4 groups were injected with Staphylococcus aureus to establish model of skin abscess, and treated externally with, respectively, Modified Ruyi jinhuangsan Tincture Tincture, Mupirocin Ointment and physiological saline. The samples of subcutaneous tissue around abscess for 1 cm,2 cm and 3 cm were collected for making into slices after treatment for 8 d, 13 d, 18 d, 22 d and 25 d, and were given immunohistochemistry staining . The expressions, changes and influence of CD68 and MPO were detected and analyzed by using microscopy and integrated optical density (IOD) detection. Results IOD values of CD68 and MPO showed a trend of firstly ascending and then descending during the treatment, and decreased with the increase of distance from abscess center and their positive expressions weakened also. The influence of Modified Ruyi jinhuangsan Tincture Tincture was two-sided on local abscess during abscess non-formation stage and late stage of abscess formation. The expressions of CD68 and MPO were lower in pretreatment group in varying degrees than those in other groups (P<0.05 or P<0.01) at different stages and distances during abscess non-formation stage. The expressions of CD68 and MPO were higher in treatment group in varying degrees than those in other groups (P<0.05 or P<0.01) after abscess formation. Conclusion The early administration of Modified Ruyi jinhuangsan Tincture Tincture for treatment skin abscess in rats can regulate the releases of CD68 and MPO, local proinflammatory cytokines, at different stages of abscess formation, and provide a favorable condition for huchang formation.

    Characteristics of Hamilton Depression Scale and Hamilton Anxiety Scale in patients with depressive disorder-liver depression and spleen deficiency pattern: a study based on association rules and entropy clustering algorithm*

    Objective To study the characteristics of Hamilton Depression Scale (HAMD) and Hamilton Anxiety Scale (HAMA) in patients with depressive disorder-liver depression and spleen deficiency pattern. Methods The general data, disease history information, Results of related psychological assessment and examination scales (HAMD and HAMA) and relevant information of TCM from 200 cases were input into TCMISS V2.5 software. All data was statistically analyzed by applying association rules, improved mutual information method and entropy clustering algorithm of complex system. Results HAMD item combinations, extracted based on association rules, included depressed mood, work and interest, mental anxiety, somatic anxiety, systemic symptoms, hypoesthesia, insomnia-early, insomnia-middle, early awakening and gastrointestinal symptoms. There were 7 core combinations evaluated based on entropy clustering algorithm of complex system. HAMA item combinations, extracted based on association rules, included depressed mood, anxious mood, tension, insomnia, symptoms of sensory system, respiratory system and vegetative nervous system, and dysmnesia or attention deficits. There were 3 core combinations evaluated based on entropy clustering algorithm of complex system. Conclusion Liver qi depression is the basic pathogenesis of depressive disorder. As the disease developing, liver depression and spleen deficiency will be the total trend of pathogenesis in depressive patients, and liver depression and spleen deficiency pattern is a most common pattern of depressive disorder. The clinical manifestations of depressive disorder are complex, and symptoms of multiple systems can be displayed. The symptom items from HAMD and HAMA are consistent in 200 patients, and assessment Results of HAMD and HAMA focus on vegetative nervous system, sensory system, respiratory system, gastrointestinal system, muscular system and cardiovascular system in patients with depressive disorder-liver depression and spleen deficiency pattern.

    Plasma metabolomic study on deficiency or excessiveness patterns of chronic recurrent ulcerative colitis based on H-nuclear magnetic resonance*

    Objective To discuss the plasma metabolomic characteristics in patients with ulcerative colitis (UC) and deficiency or excessiveness patterns. Methods UC patients (n=31, UC group) with spleen-stomach qi deficiency pattern or large intestinal dampness-heat pattern and health volunteers (n=40, control group) were chosen, and general conditions were observed in 2 groups. And UC group was divided into subgroup of large intestinal dampness-heat pattern (subgroup A) and subgroup of spleen-stomach qi deficiency pattern (subgroup B). The content of plasma metabolites was determined quantitatively by using H-nuclear magnetic resonance (1H-NMR). The multivariate statistical analysis was conducted by using principal component analysis (PCA), partial least squares-discriminant analysis (PLS-DA) and orthogonal partial least squares-discriminant analysis (OPLS-DA). Results There were 10 potential biomarkers screened. The levels of acetoacetate and acetone were higher, and levels of lactic acid, carnosine, alanine and isoleucine were significantly lower in UC group than those in control group. The levels of glucose, threonine and carnosine were higher, and levels of valine, alanine and glycine were lower in UC patients with large intestinal dampness-heat pattern than those in UC patients with spleen-stomach qi deficiency pattern. Conclusion There are obviously disorder of glycometabolism, amino acid metabolism and lipid metabolism in UC patients with deficiency or excessiveness patterns. The pattern differentiation and classification of UC has a metabolite basis.
