    In the social process of aging getting worse, a positive view of aging should be established under the guide of concept of traditional Chinese medicine (TCM). It will enable the elderly to face elderly life, keep their physical and mental health and integrate into social as an important resource, which is an important part of maintaining brain health. Based on 3 aspects, “process observation of TCM cognitive emphasis on brain and brain aging”, “brain health examination and warning prevention and treatment of mild cognitive impairment” and “observation and treatment of mild and moderate cognitive impairment according to evidence-based medicine and narrative medicine (controlling diseases based on patterns) in elder population”, it is believed that positive aging should be taken as one of important fields of TCM academic research. The studies on positive aging are hoped to be recommended into China brain science research program

    Ye Tianshi’s treatment methods of warming spleen yang and regulating stomach yang: resolving a doubt*

    In visceral manifestation theory of traditional Chinese medicine (TCM), spleen and stomach are coupled viscera and usually, they are also mentioned together. However, their physiological and pathological characteristics, and therapeutic Methods and suitable formulas and herbs for spleen and stomach are all different. The differences in pathogenesis, pattern manifestations, therapeutic Methods, formulas and medicinals between spleen and stomach are analyzed and summarized based on Ye Tianshi’s clinical records. The therapeutic Methods for different levels of spleen yang deficiency and stomach yang deficiency are elucidated in detail with specific medical cases, for examples, Li Dongyuan’s therapeutic method of replenishing qi and upraising yang, and therapeutic Methods of Linggui Zhugan Tang, Lizhong Tang and Fuzi Lizhong Tang for warming spleen yang, and therapeutic Methods of Dabanxia Tang, Wuzhuyu Tang and Fuzi Jingmi Tang for regulating stomach yang. In some circumstance, spleen yang deficiency and stomach yang deficiency may appear simultaneously, and they should be treated together or in proper order according to pattern manifestations. It is hoped to provide reference for clinical diagnosis and treatment of spleen-stomach diseases.

    Formula combination thought of Fangfeng Tongsheng San understood from angle of thinking of classification according to manifestation*

    Fangfeng Tongsheng San (Miraculous Powder of Ledebouriella) is a representative classic formula used widely in treatment of various types of diseases. However, there are disputes on its classification, and even in the determination of the sovereign medicinal there are many differences. As the only Liu Hejian’s formula, who was a master of cold school, Fangfeng Tongsheng San is included in textbook-Science of Herbal Formula. It may embody Liu Hejian’s thought of clearing heat and is a pure heat-clearing agent. The thinking of classification according to manifestation is an important thinking pattern of the theory of traditional Chinese medicine (TCM). Therefore, combining with TCM thinking characteristic, the formula design thought of Fangfeng Tongsheng San is analyzed by applying TCM formulation. It is believed that Fangfeng Tongsheng San uses the therapies of treating heat with cold medicinals, removing fire stagnation, extracting the firewood from under cauldron, inducing diuresis and clearing heat, and nourishing blood and clearing heat. The understanding of Fangfeng Tongsheng San from the angle of thinking of classification according to manifestation will open a new door for us to use it flexibly.

    Discussion on “One” in Huangdi Neijing*

    One” contains the meaning of “Dao” and “origin” from the perspective of ancient philosophy, which is the manifestation of the law of “Dao”. Also, it is both the trigger power and silent selfless one, and at the same time, it is the agility and keeping awareness. “One” in Huangdi Neijing (Yellow Emperor’s Internal Classic) contains “Shen” and “Dao”, and is related to “essence of yin and yang”, also, it is used to describe the state of ordinary people. In the aspect of treatment, Huangdi Neijing thinks that doctors should strive towards the state of “One”, and puts forward the thought of “keeping One”. The shallow level of “One” can be understood as integrating medical theory and practice, grasping of core theory, and maintaining of all diseases and Methods being inseparable from this core by doctors in clinical practice. The deep level of “One” requires the doctor to be very quiet and aware of the changes around him without being disturbed by the influence, which means in the state of “Dao”. Therefore, if we do not understand “One” and “Dao”, the more knowledge we have while studying medicine, the more doubts we may have. That is, we should aim at “One” as required by Huangdi Neijing. We should not be complacent about having medical knowledge. Since if there is no “One” in the base of these knowledge, it will only increase confusions. And let people confused in the vast sea of medicine, then, it will affect the clinical efficacy

    Intervention of therapy of replenishing qi, activating blood and resolving aggregation on apoptosis of prostate tissue in patients with benign prostatic hyperplasia*

    Objective To observe the influence of therapy of replenishing qi, activating blood and resolving aggregation on apoptosis of prostate tissue in patients with benign prostatic hyperplasia (BPH). Methods The samples of prostate tissue were collected from BPH patients for cell cultivation, and then divided randomly into blank group, tamoxifen group (1.00 mg/L), high-dose (6.25 mg/L), mid-dose (3.13 mg/L) and low-dose groups (1.56 mg/L) of medicated solution with medicinal used in therapy of replenishing qi, activating blood and resolving aggregation. After medicated for 24 h, the apoptosis index was detected by using TUNEL method, cell cycle was detected by using flow cytometer, and expressions of bcl-2, bax and caspase-3 were detected by using RT-PCR. Results The apoptosis index was promoted in high-dose, mid-dose and low-dose groups, and the difference had no statistical significance among high-dose and mid-dose groups and tamoxifen group. The quantity of G0/G1 phase cells increased and G2/M phase cells decreased in high-dose, mid-dose and low-dose groups compared with blank group, and the difference had no statistical significance among high-dose, mid-dose and low-dose groups and tamoxifen group. The expression of bcl-2 gene was inhibited and meanwhile bax gene expression was promoted in high-dose and mid-dose groups and tamoxifen group. The expression of caspase-3 gene was promoted in high-dose, mid-dose and low-dose groups. Conclusion The therapy of replenishing qi, activating blood and resolving aggregation can improve apoptosis through down-regulating bcl-2 gene expression, up-regulating bax gene expression, promoting caspase-3 gene expression, and blocking proliferated prostate tissue cells at G0/G1 phase.

    Influence of Erbu Zhuyu Tang on expressions of endometrial autophagy genes-Beclin-1 and LC3B in mice with embryo implantation dysfunction*

    Objective To investigate the influence of Erbu Zhuyu Tang (Decoction of Promoting Fertility with Two Tonifying Medicinals) on embryo implantation rate and mRNA and protein expressions of endometrial autophagy genes-Beclin-1 and microtubule-associated protein light chain 3 (LC3B) in mice with embryo implantation dysfunction. Methods Female ICR mice (n=50) were randomly divided into blank group, model group, low-dose and high-dose Erbu Zhuyu Tang groups and aspirin group (each n=10). The model of embryo implantation dysfunction was established by using mifepristone. The mice in all groups were orally given corresponding medicinals each day and executed in the afternoon of the 5th d of pregnancy. The morphology of uterus was observed, pregnancy rate and average number of embryo implantation sites were calculated, and mRNA and protein expressions of Beclin-1 and LC3B were detected. Results The color of uterus was ruddy and embryo implantation sites were intumescent and distributed equally in blank group. The color of uterus was pale and slender in model group. The uterus was slender and its color was pink in medicated groups. The number of embryo implantation sites decreased in model group compared with blank group (P<0.001). Compared with model group, the number of embryo implantation sites all increased in high-dose Erbu Zhuyu Tang group and aspirin group (P<0.05). The mRNA and protein expressions of Beclin-1 and LC3B were lower in model group than those in blank group (P<0.05). Compared with model group, the mRNA and protein expressions of Beclin-1 and LC3B increased in low-dose and high-dose Erbu Zhuyu Tang groups and aspirin group (P<0.05). Conclusion Erbu Zhuyu Tang can increase the mRNA and protein expressions of Beclin-1 and LC3B, promote autophagy level of endometrial cells and improve endometrial receptivity and average number of embryo implantation sites.

    Mechanism of Huotan Jiedu Tongluo Fang in intervening endoplasmic reticulum stress-autophagy signaling pathway in rabbits with carotid atherosclerosis*

    Objective To study the effect of Huotan Jiedu Tongluo Fang (HTJDTLF, Sweeping Phlegm-Detoxicating-Freeing Collateral Vessels Formula) on intervening endoplasmic reticulum stress (ERS)-autophagy signaling pathway in rabbits with carotid atherosclerosis (CAS). Methods Male Japanese rabbits aged 10 weeks old were randomly divided into sham-operation group, model group, HTJDTLF group and atorvastatin group. CAS model was established through feeding methionine fodder and balloon injury of common carotid artery intima. The samples of injured common carotid artery were collected and given HE staining for observing histopathological structure. The content of serum homocysteine (Hcy) was detected by using ELISA in all groups. The mRNA levels of protein kinase-like endoplasmic reticulum kinase (PERK, an ERS marker molecule), eukaryotic initiation factor 2ɑ (eIF-2ɑ) and autophagy marker molecule-Atg5 in common carotid artery tissue were detected by using real-time fluorescence quantitative polymerase chain reaction (RT-PCR). The protein expressions of eIF-2ɑ and Atg5 in common carotid artery tissue were detected by using Western blotting assay. Results Compared with sham-operation group, damaged common carotid artery had significant intimal hyperplasia and vascular lumen stanosis, and the content of serum Hcy increased (P<0.01) in model group. Compared with model group, the intimal hyperplasia was significantly relieved and content of serum Hcy decreased in HTJDTLF group (P<0.01). The mRNA levels of PERK, eIF-2ɑ and Atg5 and protein expressions of eIF-2ɑ and Atg5 increased in model group compared with sham-operation group (P<0.01). The mRNA levels of PERK, eIF-2ɑ and Atg5 decreased (P<0.01) and protein expressions of eIF-2ɑ and Atg5 decreased (P<0.01) in HTJDTLF group and atorvastatin group. Conclusion HTJDTLF can significantly relieve CAS and reduce serum Hcy content in rabbits with CAS, and the mechanism maybe related to the inhibition of excessive activation of ERS-autophagy signaling pathway

    Influence of Siwu Tang on proliferation of MC3T3-E1 cells based on GPER-mediated EGFR/PI3K pathway*

    Objective To investigate the influence of Siwu Tang (Four Ingredients Decoction) on proliferation efficacy of mouse embryonic osteoblasts (MC3T3-E1) based on G-protein-coupled estrogen receptor (GPER)-mediated pathway of epidermal growth factor receptor (EGFR)/ phosphatidylinositol 3-kinase (PI3K), and study the molecular mechanism. Methods Female SD rats (n=40) were randomly divided into normal group, estradiol group, and high-dose, mid-dose and low-dose Siwu Tang groups (each n=8). At the same time, the high-dose Siwu Tang group was divided into high-dose Siwu Tang+G1 group, high-dose Siwu Tang+G15 group, high-dose Siwu Tang+AG1478 group and high-dose Siwu Tang+LY294002 group. Taken E2 medicated serum as positive control, MC3T3-E1 was intervened with high-dose, mid-dose and low-dose Siwu Tang medicated serum. The influence of 24-h intervention with medicated serum on MC3T3-E1 proliferation was detected by using methyl thiazolyl tetrazolium test (MTT) in all groups. The influence of high-dose Siwu Tang in high dose on protein expressions of EGFR, PI3K, cyclin B1 and cyclin D1 in MC3T3-E1 were detected by suing Western blot assay and immunofluorescence technique(IF). The effective pathway and molecular mechanism of Siwu Tang in high dose influencing MC3T3-E1 proliferation were detected by using GPER agonist (G1), GPER antagonist (G15), EGFR inhibitor (AG1478) and PI3K inhibitor (LY294002). Results The proliferation of MC3T3-E1 was improved in high-dose, mid-dose and low-dose Siwu Tang groups compared with normal group (P<0.01), and G1 increased significantly the improving effect of Siwu Tang in high dose on proliferation of MC3T3-E1 (P<0.01). G15, AG1478 and LY294002 decreased the improving effect of Siwu Tang in high dose on proliferation of MC3T3-E1 (P<0.01). The Results of Western blotting assay and FIA showed that the protein expressions of EGFR, PI3K, cyclin B1 and cyclin D1 in MC3T3-E1 increased significantly in all Siwu Tang groups compared with normal group (P<0.01). G1 promoted the above-mentioned expressions (P<0.01), and G15, AG1478 and LY294002 reduced the above-mentioned expressions (P<0.01, P<0.05) compared with Siwu Tang in high dose. Conclusion Siwu Tang has significant improving effect on the proliferation of MC3T3-E1, and the effect mechanism may be related to the conduction of GPER-mediated EGFR/PI3K signaling pathway.

    Influence of medicated serum of Houshi Hei San on expressions of PLCγ2/ERK and TGF-β/Smad2/3 in vascular endothelial cells after injury of oxygen-glucose deprivation*

    Objective To investigate the influence of medicated serum of Houshi Hei San (Hou’s Black Powder, HHS-MS) on expressions of phospholipase Cγ2 (PLCγ2), extracellular regulated kinase (ERK), transforming growth factor-β (TGF-β) and Smad2/3 in human umbilical vein endothelial cells (HUVEC) after injury of oxygen-glucose deprivation (OGD). Methods HUVEC were randomly divided into control group, model group, 2.5% HHS-MS group, 5.0% HHS-MS group, 10.0% HHS-MS group and 20.0% HHS-MS group. The OGD model was established through putting HUVEC in a hypoxic incubator for continuous cultivation for 6 h after changed to sugar-free nutrient solution in model group and all HHS-MS groups. HUVEC were cultured normally in the same duration. After modeling, the nutrient solution was changed to complete nutrient solution with 10% blank serum in control group and model group, and MS with corresponding mass fraction of HHS-MS was used in all HHS-MS groups. The HUVEC total protein was extracted after normal cultivation for 24 h. The protein expressions of PLCγ2, ERK, p-ERK, TGF-β and Smad2/3 in HUVEC were detected by using Western blotting assay. Results Compared with control group, the expression of PLCγ2 and Smad2/3 were up-regulated (P<0.01) and expression of TGF-β was down-regulated (P<0.01) in model group. Compared with model group, the expression of TGF-β was up-regulated in 5.0% HHS-MS group and expressions of PLCγ2 and Smad2/3 were down-regulated in 2.5% HHS-MS group, 5.0% HHS-MS group and 10.0% HHS-MS group (P<0.01 or P<0.05). Conclusion HHS-MS can regulate PLCγ2/ERK signaling pathway and TGF-β/Smad2/3 signaling pathway, and improve the proliferation of HUVEC after OGD injury.

    Effect of Liyan Tongqiao Fang on tongue muscle contraction in rats after hypoglossal nerve injury*

    Objective To discuss the therapeutic effect of Liyan Tongqiao Fang (Soothing-throat Freeing-orifice Formula) on tongue muscle contraction in rats after hypoglossal nerve injury by applying nerve-muscle electrophysiology method. Methods Male adult Wistar rats (n=60) were randomly divided into sham-operation group, model group and Liyan Tongqiao Fang group (each n=20). The model of hypoglossal nerve injury was established by clamping in model group and Liyan Tongqiao Fang group, while in sham-operation group hypoglossal nerve was only separated without champing. After modeling for 2 d, Liyan Tongqiao Fang group was given Liyan Tongqiao Fang (0.3 mg/d), and sham-operation group and model group were given the same quantity of saline solution. The threshold strength, single contraction amplitude and tonic contraction amplitude were detected in all groups respectively on the 7th d and the 14th d. Results The threshold intensity of injured tongue muscle increased in model group compared with sham-operation group (P<0.05) at the same time point, and decreased in Liyan Tongqiao Fang group compared with model group (P<0.05) at the same time point. The threshold intensity of injured tongue muscle increased in model group and Liyan Tongqiao Fang group compared with non-injured tongue muscle within the same group (P<0.05). The threshold intensity of injured tongue muscle decreased in Liyan Tongqiao Fang group and model group on the 14th d compared with that on the 7th d (P<0.05). The single contraction amplitude and tonic contraction amplitude of injured tongue muscle decreased in model group compared with sham-operation group (P<0.01) at the same time point, and increased in Liyan Tongqiao Fang group compared with model group (P<0.05) at the same time point. The single contraction amplitude and tonic contraction amplitude of injured tongue muscle decreased in model group and Liyan Tongqiao Fang group compared with non-injured tongue muscle within the same group (P<0.05). The single contraction amplitude and tonic contraction amplitude of injured tongue muscle increased in Liyan Tongqiao Fang group and model group on the 14th d compared with those on the 7th d (P<0.05). Conclusion Liyan Tongqiao Fang can reduce the contraction threshold intensity, promote excitability of hypoglossal nerve-tongue muscle conduction system, improve the contraction ability of tongue muscle, and relieve dysphagia in rats after hypoglossal nerve injury.

    Effects of Wumei Wan on interleukin-1β, phosphorylase nuclear factor-κB and forkhead transcription factor 3 in rats with immune colitis*

    Objective To observe the influence of Wumei Wan (Mume Pill) on the expressions of interleukin-1β (IL-1β), phosphorylase nuclear factor-κB (pNF-κB) and forkhead transcription factor 3 (Foxp3) in serum and colon tissue in rats with immune colitis, and discuss the effective mechanism. Methods SD rats (n=40) were randomly divided into normal group, model group, methylprednisolone group (MPSL group, 2.0 mg/kg), low-dose Wumei Wan group (13.4 g/kg) and high-dose Wumei Wan group (26.8 g/kg, each n=8). The model of immune colitis was established through intracolonic instillation of 2,4,6-trinitrobenzenesulfonic acid (TNBS) and ethyl alcohol. The rats in all groups were orally given corresponding medicinals once a day for 14 d after modeling for 3 d and then all indexes were detected. The colon mucosa damage index (CMDI) was observed after anatomy. The level of IL-1β was detected by using ELISA, protein expression of pNF-κB was detected by using Western blotting assay, and gene expression of Foxp3 was detected by using qPCR in all groups. Results Compared with normal group, the level of serum and colon IL-1β increased, and protein expression of colon pNF-κB increased and expression of colon Foxp3 decreased in model group (P<0.05). Compared with model group, level of serum and colon IL-1β increased and expression of Foxp3 increased in MPSL group (P<0.05). The level of colon IL-1β and protein expression of pNF-κB decreased, and level of serum IL-1β and expression of colon Foxp3 increased in low-dose Wumei Wan group and high-dose Wumei Wan group (P<0.05). Compared with MPSL group, the level of serum IL-1β increased (P<0.05) and level of colon IL-1β decreased (P<0.05) low-dose Wumei Wan group and high-dose Wumei Wan group. Conclusion Wumei Wan can down-regulate the expressions of pNF-κB and IL-1β in damaged colon tissue through increasing Foxp3 expression, and thereby relieve colon pathological damage and prevent the progression of immune colitis.

    Influence of acupuncture on ethology and expressions of brain-derived neurotrophic factor and glial fibrillary acidic protein in hippocampus, cortex and amygdala in rats with post-stroke depression*

    Objective To observe the influence of acupuncture on behavioral changes and expressions of brain-derived neurotrophic factor (BDNF) and glial fibrillary acidic protein (GFAP) in hippocampus, cortex and amygdala in rats with post-stroke depression (PSD), and discuss the mechanism of acupuncture treating PSD. Methods Male SD rats (n=60) were randomly divided into normal group (n=10) and modeling group (n=50). The rat model was established by using thread approach of middle cerebral artery occlusion (MCAO) combined with chronic unpredictable mild stimulation (CUMS). The successful modeling rats were randomly divided into model group, acupuncture group and fluoxetine group. The acupuncture group was given needling in Baihui (GV20), Fengfu (GV16), Shenmen (HT7) and Taichong (LR3) once a day for 3 weeks, and fluoxetine group was orally given fluoxetine (2 mg/kg) once a day for 3 weeks. The behavioral changes were closely observed, and protein expressions of BDNF and GFAP in hippocampus, cortex and amygdale were detected by suing Western blotting assay in all groups. The mRNA expressions of BDNF and GFAP in hippocampus, cortex and amygdale were detected by using RT-qPCR, and ultrastructural changes of astrocytes (Ast) in brain tissue were observed by using transmission electron microscope (TEM). Results Compared with normal group, the protein and mRNA expressions of BDNF and GFAP in hippocampus, cortex and amygdale all increased in model group (P<0.01 or P<0.05). Compared with model group, the protein and mRNA expressions of BDNF and GFAP in hippocampus, cortex and amygdale all increased in acupuncture group and fluoxetine group (P<0.01). Conclusion Acupuncture can relieve the symptoms in PSD rats, and the mechanism may related to that it can promote the expressions of BDNF and GFAP in hippocampus, cortex and amygdale, improve ATP release in Ast, ameliorate neuron injury and improve neurogenesis.

    A comparative study on tongue diagnostic parameters between phlegm-stasis binding pattern and non-phlegm-stasis binding pattern of coronary heart disease at different time points*

    Objective To measure repeatedly the tongue manifestations at different time points in patients with coronary heart disease (CHD)-phlegm-stasis binding pattern (CHD-PSB pattern), and provide Objective evidence for differentiating and diagnosing CHD-PSB pattern based on traditional Chinese medicine (TCM).Methods The eligible subjects were chosen, and the information of 10 indexes, including tongue fur color, tongue fur area, teeth marks, ecchymosis, fissures and thorn-like protrusions, were collected and analyzed by using tongue diagnostic instrument. Firstly, the indexes of tongue manifestations were compared among CHD-PSB patients, non-CHD-PSB patients and healthy persons in the day of hospital admission (the 0th d), and then those indexes were compared between CHD-PSB patients and non-CHD-PSB patients on the 0th d, 5th d and 10th d after they were hospitalized and received treatment.Results There were totally 149 cases been chosen, among them 8 was not accordant with inclusion criteria and 141 were final eligible cases, who were divided into CHD-PSB pattern group (n=83), non-CHD-PSB pattern group (n=58), and health group (n=22) was established in addition. There were 39 cases (24 in CHD-PSB pattern group and 15 in non-CHD-PSB pattern group) received repeated measures at 3 time points. The differences in the area of tongue fur, G value of tongue fur, amount and area of teeth marks, amount and length of fissures, amount and area of ecchymosis, and amount and area of thorn-like protrusions had statistical significance between CHD-PSB pattern group and health group. The differences in amount and area of teeth marks, amount and area of ecchymosis and amount and area of thorn-like protrusions had statistical significance between CHD-PSB pattern group and non-CHD-PSB pattern group. The comparison in the measurement indexes of tongue manifestations of the same pattern types at 3 time points showed that, except of amount and area of thorn-like protrusions in CHD-PSB pattern group, the difference in other indexes had no statistical significance.Conclusion Tongue diagnostic instrument can realize the possibility of objectification and quantification of TCM diagnosis, and the measurement Results can be used as a reference for diagnosis.
