      摘要:The authors hold that the history of the traditional Chinese medicine (TCM) in the past one hundred years may be thought of as a history of argument about TCM between the giving-up party and keeping-down party. The two parties may be considered as the renovation party and the traditional development party according to the results of the analysis of what they held. The opposites and conflicts between the two parties are the inevitability in the course of the amalgamation of the eastern and western cultures and thoughts, and at the same time, they are the reflection of some special phenomena during the dissemination of scientific spirit and scientific outlook. The authors have tried to analyze these special phenomena and to understand their cultural and ideaistic contents from the angles of cognitive psycology, scientific spirit, etc., in order to show the complexity, difficulty and hardship in the development of TCM, hoping to help the adoption of appropriate conception in the research of TCM.

      摘要:Objective To observe the effects of the effective fractions of Qingkailing (EFQs) on the levels of NGF, bFGF and BDNF at different intervals in the rat brain undergoing ischemia, in order to find the mechanism of EFQs against cerebral ischemic injury on the basis of neurotrophic factors. Methods The Longa’s procedure was used to induce cerebral ischemia in the rat, and EFQs were given to the model for intervention, and ELISA was used to detect the levels of NGF, bFGF and BDNF in the brain of the model rats. Results The levels of the mentioned neurotrophic factors were reactively increased at the 12 th hour and 24 th hour, respectively during the cerebral ischemia in the model group; the level of NGF was markedly decreased at the 12 th hour and 24 th hour, respectively in both the geniposide group and the Concha Margaritifera Usta group; the level of NGF was markedly increased at the 12 th hour in the cholic acid group; the level of bFGF was markedly decreased at the 24 th hour again in the cholic acid group while it was increased in all other groups; and the level of BDNF was decreased at the 12 th hour and 24 th hour in all the EFQs-treating groups.Conclusion Promotion of the production of bFGF by the astrocytes is the main mechanism of EFQs in protecting neurons from being injured by cerebral ischemia.

      摘要:Objective To find new angiogenic inhibitors from TCM drugs and investigate their mechanisms of inhibiting angiogenesis.Methods Hydroxysafflor yellow A was separated from the dried flowers of Carthamus tinctorius; the chick embryo chorioallantoic membrane (CAM) test was carried out to observe the effect of hydroxysafflor yellow A on angiogenesis; RT-PCR was used to observe the effects of hydroxysafflor yellow A on the mRNA expressions of bFGF, VEGF and VEGF-F(flt-1) in the CAM.Results Hydroxysafflor yellow A extracted from the dried flowers of Carthamus tinctorius was of purity of 98.%; hydroxysafflor yellow A could markedly reduce the number of CAM vessels when it was at the concentration of 0.59 g/L (P<0.01), and remarkably inhibit the mRNA expressions of bFGF, VEGF and VEGF-F(flt-1) in the CAM (P<0.05). Conclusion It is the first time when hydroxysafflor yellow A has been discovered to have the effects of markedly inhibiting angiogenesis in CAM, one of the mechanisms of which may be its inhibiting the mRNA expressions of bFGF, VEGF and VEGF-F(flt-1). The results of the experiment suggest that hydroxysafflor yellow A may become a potential angiogenic inhibitor.

      摘要:Objective To observe the effects of different doses of the serum medicated with Tongmaining Capsules (TCs) on LPC-induced activity of proliferation of vascular smooth muscle cells (VSMCs) and the cell cycle. Methods The explant technique was used to cultivate VSMCs. With LPC at 10 -8 mol/L as the stimulant, the serum pharmacology procedure was used to observe the effects of TCs, which were divided into large, medium and small doses. MTT method was used to detect the activity of VSMCs proliferation, and the flow cytometry was used to measure the cell cycle. Results All the three doses of TCs could markedly decrease the OD value of VSMCs and inhibit the LPC-induced VSMCs proliferation in a dose-dependent manner. The three doses of TCs also could markedly decrease the percentage of the number of VSMCs at phase S, and increase the percentage of the number of VSMCs at phase G 0G 1 to halt VSMCs in phase G 0G 1 and stop them into phase S. Conclusion TCs may exert their proliferation-inhibiting effect on VSMCs mainly through affecting the DNA synthesis, decreasing mitochysis, increasing the percentage of the number of VSMCs at phase G 0G 1, and decreasing the percentage of the number of VSMCs at phase S.

      摘要:Objective To establish the animal model of post-stroke depression. Method Adult Wistar rats were adopted in the experiment. First, the model of cerebral ischemia was set up by removing the blood vessels in the left cortex of the rat, and then the model rats were separately raised and put into chronic unpredictable mild stress, in order to induce the model of cerebral ischemia into the model of post-stroke depression. Results After undergoing the model-inducing procedures, the rats were examined by behavioral tests. It was found that the amount of sugar-water consumption by the rats in the model group was much less than that by the rats in the normal group (P<0.01), implying the absence of interest and anhedonia of the rats in the model group. In the open-field test, both the crossing time and rearing time were found to be much less in the model group than those in the normal group (P<0.01), implying the underactivity and absence of interest of the rats in the model group. In the water maze test, it was found that the rats in the model group needed more time to reach the end-point and had more errors than the rats in the normal group did (P<0.05 or 0.01), implying the decreased ability to learn and memorize of the rats in the model group. Conclusion With good repeatability, the anhedonia and underactivity found in the rats with post-stroke depression may represent the core symptoms in the patients of post-stroke depression, and the rat model of post-stroke depression is an ideal model for post-stroke depression research.

      摘要:Objective To observe the changes in the blood rhenology and the fibrinolytic activity in the condition of hyperuricemia plus hypertriglyceridemia, and to investigate the effects of Chicory Extract N2 (CE-N2) on the blood rhenology and the fibrinolytic activity in the model of hyperuricemia plus hypertriglyceridemia. Method Yeast was used to establish the partridge model of hyperuricemia plus hypertriglyceridemia, in which the changes in the blood rheno-logy and the fibrinolytic activity, and the effects of CE-N2 were observed. Results It was found that the criteria of blood rheology and fibrinolytic activity became obviously abnormal in the model, and CE-N2 could markedly improve the state of blood rheology and the activity of fibrinolytic system.Conclusion The criteria of blood rheology may become obviously abnormal in the condition of hyperuricemia plus hypertriglyceridemia, and CE-N2 can reverse the abnormality.

      摘要:Objective To investigate the effects of fatigue and purgation, the two hemorrhagic factors, on certain links in the hemorrhagic and coagulative processes in the rat, and to observe the corrective effects of the formula for benefiting qi and checking hemorrhage on those links. Methods Swimming, senna leaf and leech were used together to the rat to create an original animal model of splenic failure in controlling blood, and leech alone was used to the rat of splenic deficiency to create a new animal model of splenic failure in controlling the blood. PT, APTT and the blood routine criteria were examined in both models. Result Although the rats in both model groups manifested tendency to bleed, the rats in the new model of splenic failure in controlling the blood showed a stronger hemorragic tendency.Conclusion The condition of splenic deficiency is more sensitive to the hemorragic factor (leech powder), and the formula for benefiting qi and checking hemorrhage has a good corrective effect on the new animal model of splenic failure in controlling the blood.

      摘要:Objective To investigate at the molecular level the mechanisms of Fuganjiedu Formula (FF) in treating alcoholic liver disease.Methods 40 Wister rats were randomly divided into the normal control group (10 rats) and the model group (30 rats);all the rats in the normal control group and 10 rats in the model group (1 died of pulmonary choke during the experiment) were decollated at the end of the 16th week,and the samples of the serum and liver tissue were collected;the rest 20 rats in the model group were randomly divided into the TCM drug-treating group and treating-control group (10 rats in each group),and the rats in both groups were executed after 24 weeks.HE,Sudan IV and Masson’s trichrome staining were respectively used to observe the microscopic pathologic changes and the changes in the electronical microscopic ultrastructures of the hepatic tissue,and the immunohistochemical staining and RT-PCR were used to observe the effects of FF on the expressions of peroxisome proliferator-activated receptor-γ(PPAR-γ) protein and gene.Results In the model group,the levels of ALT and TNFαin the serum and the level of MDA in the liver homogenate were increased;obvious adipose denaturalization,inflammation,necrosis and fibrosis were observed in the hepatic tissue;and the expressions of PPAR-γ protein and mRNA were decreased.In the TCM drug-treating group,the severity of adipose denaturalization,inflammation,necrosis and fibrosis of the hepatic tissue was markedly alleviated (which was close to that in the normal control group),and the expressions of PPAR-γ protein and mRNA were increased (P<0.05 as compared with those in the treating-control group).In the treating-control group,the level of ALT and the severity of hepatocellular adipose denaturalization were reduced as compared with those in the model group,but there was no marked change in the fibrosis and the expression of PPAR-γ.Conclusion FF can effectively reverse the alcoholic hepatic lesion,the mechanism of which is related with FF’s activating PPAR-γ,and inhibiting the inflammatory reaction in alcoholic hepatic lesion.

      摘要:Objective To investigate the possible mechanisms of the effects of Shengjiangxiexin Decoction (SD) in preventing and treating reflux esophagitis. Methods 36 Rats were randomly divided into the control group, model group, and treating group; the operation for inducing reflux esophagitis was performed on the rats in the model group, and the sham-operation was performed on the rats in the control group. 1 Week after the operations, the treating group was given SD intragastrically, and the other two groups were given normal saline intragastrically. The rats in all groups were decollated 4 weeks after the operations, and their esophagi were collected for gross scoring and histopathologic observation and at the same time, the esophageal homogenate was prepared for measuring the levels of VIP in the esophageal tissue and the blood, the level of MDA in the esophageal tissue, and the activities of SOD and GSH-PX in the esophageal tissue. Results SD could markedly decrease the levels of VIP and MAD, and increase the activities of SOD and GSH-PX, and improve the results of gross scoring and with the histopathologic scale. Conclusion SD can decrease the level of VIP and increase the ability of antioxidation in the rat model of reflux esophagitis, and possesses the effects of preventing and treating reflux esophagitis.

      摘要:Objective To observe the protective effects of the active fraction of Huanglianjiedu Decoction (AFHD) against glutacid-induced cytotoxicity to nerve cells in culture.Method New-born rat cortical neurons were cultivated in vitro, and glutacid and AFHD were added into the culture medium during the cultivation, in order to observe glutacid-induced cytotoxicity to cortical neurons and the protective effects of AFHD against the cytotoxicity.Results It was found that AFHD could markedly increase the activity of the model cells at the dose of 305 mg·kg -1 ; markedly decrease the leakage rate of LDH and production of MDA at the doses of 610 mg·kg -1 , 305 mg·kg -1 and 153 mg·kg -1 ; reduce the production of NO at the doses of 305 mg·kg -1 and 153 mg·kg -1 ; and increase the activity of SO, at the dose of 610 mg·kg -1 . Conclusion AFHD has a remarkable protective effect against glutacid-induced cytotoxicity to cortical neurons, the mechanism of which may be related with its decreasing the level of NO and resisting oxidation.

      摘要:Objective To observe the changes in [Ca 2+ ]i, apoptotic rate, cell activity and mitochondrial membrane potential when Danshensu (3,4-dihydroxyphenyllactic acid) was administered to the SH-SY5Y cells injured by hypoxia/hypoglycemia, and to investigate the possible mechanism of the protective effects of Danshensu on the mitochondria of the nerve cells.Methods The cell cultivation techniques and MTT colorimetric test were used to examine cell activity; and the flow cytometry was used to detect the [Ca 2+ ]i, apoptotic percentage and mitochondrial membrane potential.Results The [Ca 2+ ]i increased markedly, reaching to 8.46 nmol/L (P<0.01); and the apoptotic rate increased remarkably, being 18.59% (P<0.01), in the SH-SY5Y cells after being with hypoxia/hypoglycemia for 2 hours.After 4 hours of hypoxia/hypoglycemia, the [Ca 2+ ]i reached the peak, 9.89 nmol/L (P<0.01), and after that, it tended to decrease. The apoptotic rate increased markedly along with the prolonging of the duration in which the cells were with hypoxia/hypoglycemia, and the rate was 45.91% when hypoxia/hypoglycemia lasted for 12 hours. The mitochondrial membrane potential and cell activity decreased to 29.17% (P<0.01) and 38.80% (P<0.01), respectively after 2 hours of hypoxia/hypoglycemia; they further decreased along with the prolonging of the time of hypoxia/hypoglycemia, being 56.72% (P< 0.01) and 36.58% (P<0.01), respectively when hypoxia/hypoglycemia lasted for 12 hours. Danshensu was found to markedly decrease [Ca 2+ ]i, inhibited the onset of apoptosis and increase mitochondrial membrane potential and cell activity (P<0.05 to 0.01).Conclusion Danshensu exerts a stabilizing effect on mitochondrial membrane potential by inhibiting the decrease in mitochondrial membrane potential due to hypoxia/hypoglycemia. It also inhibits the onset of apoptosis, which may be related with its effect for inhibiting the calcium overload in nerve cells.

      摘要:Objective To investigate the effects of Naolutong Formula (NF) on the expression of cerebral bFGF in the rats with photochemically-induced cerebral ischemia. Method Immunohistochemical procedures were used to observe the expression of bFGF in the cerebral ischemic foci, surrounding area of the cerebral ischemic foci, cortex, hippocampus and cerebellum in each group in the experiment, 5 days after the cerebral ischemia was induced in the rats by photochemical procedures.Results It was found that the expression of bFGF mainly existed in the surrounding area of the cerebral ischemic foci and hippocampus, and the positive cells were primarily gliacytes and neurons in the rats undergone 5 days of cerebral ischemia; the expression of bFGF was markedly increased in the treating group compared with that in the model group.Conclusion NF can promote the survival of neurons undergone ischemia and take part in the repair of cerebral injury and nerve regeneration through increasing the expression of bFGF in the surrounding area of the cerebral ischemic foci and hippocampus.

      摘要:Objective To examine the in vitro dissolution of chemical components of the coptis root powder of different granule diameters.Method HPLC techniques were used to detect the in vitro dissolution amount and the in vitro dissolution velocity of berberine in regular coptis root powder, ultramicroscopic coptis root powder and nano coptis root powder.Results The in vitro dissolution amount and the in vitro dissolution velocity of berberine in ultramicroscopic coptis root powder and nano coptis root powder (especially in nano coptis root powder) were found to be increased, compared with those of berberine in regular coptis root powder.Conclusion The in vitro dissolution amount and the in vitro dissolution velocity of berberine in the coptis root are markedly increased when coptis root is processed into ultramicroscopic powder and nano powder.

      摘要:Objective To establish a method for the quantitative determination of crocetin in the effective fraction of Qingnaoxuanqiao Formula (QF) by RP-HPLC. Method The RP-HPLC technique was used in the experiment. YWG-C 18 column (250 mm×4.6 mm, 10 μm) was used, the mobile phase was methanol-water-glacial acetic acid (83:17:34) at a flow speed of 1 mL/min, and the detection wavelength was 423 nm.Result Crocetin showed, between 0.022~0.110μg, a fine linear relationship with the peak areas (r=0.9991). The average recovery of this procedure was 99.4%, with RSD equal to 1.90% (n=5).Conclusion This procedure is easy and accurate, and can be used as one of the methods for controlling the quality of the effective fraction of QF.

      摘要:Objective To determine the contents of aloe-emodin, rheic acid, emodin, chrysophanol, and emodin monomethyl ether in the active fraction of Yiqihuoxue Formula (YF). Method HPLC techniques were used in this experiment; the Diamonsil-C 18 column (250 mm×4.6 mm, 5 μm) was adopted, the mobile phase was methanol-0.1% phosporic acid (87∶13) at a flow speed of 1.0 mL/min, the detection wavelength was 436 nm, and the column was at the room temperature. Result The values of the average recovery of aloe-emodin, rheic acid, emodin, chrysophanol, and emodin monomethyl ether were 98.30%, 97.12%, 98.18%, 97.66% and 97.16%, respectively; and the values of RSD were 1.64%, 1.15%, 0.95%, 1.51% and 1.64%, (n=5.), respectively. Conclusion With a good repetition, this procedure is easy and accurate, and can be used in the analysis of the quality of the active fraction of YF.

      摘要:Objective To establish the methods for detecting the chemical components and identifying the pharmacognostic characteristics of the seed of Artemisia halodendron Turcz (SAHT).Methods GC-MS techniques were used to detect the chemical components of SAHT extract by CO 2 supercritical extraction, and TLC was used to identify the pharmacognostic characteristics of SAHT. Results 72 Chemical components were found from the CO 2 supercritical extract of SAHT collected from Zhelimu League of Inner Mongolia, and 60 of them were identified. In addition, the percentages of these compounds in the volatile oil containing them were detected. Conclusion In the CO 2 supercritical extract of SAHT, it is found that the content of 9,12-octadecadienoic acid is the highest, reaching 28.52%, and the content of n-hexadecanoic acid is the next. In addition, the TLC identification procedure for identifying the pharmacognostic characteristics of SAHT is established.

      摘要:To establish a method for quantitatively determining astragloside IV in Qinggan Granules (QG). Method HPLC-ELSD procedure was adopted in the experiment. Result A good linear relationship was showed between 1.01 μg~10.1μg of astragloside IV (r=0.9998,n=5), and the average recovery was 98.27%.Conclusion The result provides a new method for controlling the content of astragloside IV in medical preparations.

      摘要:Objective To provide the evidence of identification for correctly using the Chinese herb Melastoma candidum D. Don. in the clinical practice by carrying out a pharmacognostic study of the whole plant. Method Anatomical techniques were used to study the root, stem, leaf, and the powder of root of Melastoma candidum D. Don. Results Clusters of crystal of calcium oxalate were found in the phloem cells of the root, stem and leaf of Melastoma candidum D. Don. The structure of the transverse section of the stem was found specific, and there were anomalous vascular bundles in the cortex and medulla.Conclusion The data found in the experiment can be taken as the evidence for identifying the plant Melastoma candidum D.       摘要:Objective To find new slow-release adjuvant materials for preparing Slow-Release Total Notoginsenoside Tablets (SRTNT) by the solid-dispersing technology. Method Ginsenosides Rg 1 and Rb 1 were taken as criteria, the one-factor method was used to examine the influence of adjuvant materials on slow-release action, and the recipe of SRTNT was optimized at the same time.Result EC was chosen as a slow-release adjuvant material and PEG4000 as a rapid-release adjuvant material.Conclusion The slow-release effect of SRTNT is basically achieved.

      摘要:Objective To observe the clinical efficacy of Naoreqing Oral Solution (NOS) in the treatment of postoperative fever in craniocerebral operations. Methods Patients from the neurosurgical diseases section of the hospital were randomly divided into the NOS group (86 cases), the NOS plus western-style drug group (the NOS-WD group, 276 cases), and the western-style drug group (the WD group, 89 cases). The NOS group was given NOS at a dose of 50 ml/2 or 3 times/day; the WD group, the routine western-style drugs; and the NOS-WD group, routinewestern-style drugs and NOS; all groups were administered for the same doses for 7 days successively. The degrees of temperature lowered, cure rate, cure plus effectual rate and the total effective rate were compared among the three groups; the degrees of temperature lowered and the effective rates for both infective fever and noninfective fever were compared between the NOS group and NOS-WD group; and the degrees of temperature lowered and the effective rates for meningoma and glioma were also compared between the NOS group and NOS-WD group. Results The extent of the temperature lowered in the NOS group and the NOS-WD group was much greater than that in the WD group; the cure rate, cure plus effectual rate and the total effective rate in the NOS group and the NOS-WD group were much higher than those in the WD group; and as far as these criteria were concerned, there was no difference between the NOS group and NOS-WD group. NOS manifested a very good antipyretic effect on both intracranial infective fever and noninfective fever, without obvious difference between the treatments of the two types of fever concerning the degrees of temperature lowered, cure rate, cure plus effectual rate and the total effective rate; and NOS-WD, except for a larger extent of temperature decrease for intracranial infective fever, showed no obvious difference between the treatments of the two types of fever concerning the above-mentioned criteria. Both NOS and NOS-WD showed a good antipyretic effect on the postoperative fever due to the operations of meningoma and glioma, without obvious difference between the treatments of the two types of tumor concerning the degrees of temperature lowered, cure rate, cure plus effectual rate and the total effective rate.Conclusion NOS possesses a marked antipyretic effect on postoperative fever, being effective for both infective and noninfective fever, and its antipyretic effect is not related with the properties of the cerebral tumors.

      摘要:Objective To observe the influence of Sanqidan Granules (SG) on the function of islet β cells and insulin resistance in senile diabetic patients. Method 60 Senile patients of diabetes were randomly divided into the treating group (30 cases) and the control group (30 cases). The control group was given oral blood-sugar decreasing drugs only, and the treating group was given oral blood-sugar decreasing drugs plus SG. The function of islet β cells, insulin sensitivity index, and changes in blood lipid were observed prior to and after the treatment in both groups. Results It was found that the index of islet β cell function, insulin sensitivity index, and the level of HDL-C were increased, and the levels of TG and fasting blood-glucose were decreased in the treating group after the treatment, and there was a significant difference was found, as compared with the same criteria before the treatment in the treating group or with the same criteria after the treatment in the control group.Conclusion The mechanism of SG for decreasing the level of blood-sugar may be through regulating the disorder of fat metabolism, and improving β cell function and insulin resistance.

      摘要:Objective To evaluate the quality of life of senile patients of cerebral infarction in convalescence and analyze the correlative factors. Methods GQOLI-74 was used to measure and evaluate the quality of life of 135 senile patients of cerebral infarction in convalescence, and the statistical method of one-factor and multiple-element stepwise regression was used to analyze the correlative factors. Results It was found that the total score of the quality of life was very low (46.78±8.95 scores) in senile patients of cerebral infarction in convalescence. The influence of the factors of sex, age, marital status and body weight index on the quality of life of senile patients of cerebral infarction in convalescence showed no statistical significance. The major medical and social factors influencing the quality of life of the patients were found to be the duration of illness, concomitant diseases and anamnesis, nervous functional status, Barthel index, fasting blood-glucose, TCM syndromes, self-evaluation of health status, education status, life style, economic income, occupation, and living environment. Conclusion Senile patients of cerebral infarction in convalescence has a rather low quality of life, and the functions of their body is seriously damaged; social factors, medical factors and TCM syndromes have obvious influence on their quality of life. The quality of life of senile patients of cerebral infarction in convalescence can be improved by strictly controlling blood sugar and blood pressure, strengthening psychological nursing, and providing family warmth and social support.
