摘要:Objective To set up a synthetic evaluation index, a generalized index, for symptoms and signs in diagnosis of Chinese medicine. Method To evaluate based on entropy the establishment method of generalized index of function. Result[WT] The outcomes from the principle component analysis (PCA) and nonlinear PCA were compared. The establishment method of generalized index based on entropy only reflected the total characteristics of symptoms and signs and met the clinical experiences. Conclusion It is workable to set up the quantitative synthetic evaluation index in Chinese medicine diagnostic system through the establishment of generalized index based on entropy. The method plays an important role in the quantitative evaluation of curative effect of Chinese medicine.

摘要:Objective To observe the influence of 3-step sequential therapy on serum interleukin-4 (IL-4), IFN-γ and lung mRNA expression in rat asthma model interfered with hormone. Methods To divide SD rats randomly into the normal control group (group 1), asthma model group (group 2), hormone interfered group (group 3), 3-step sequential therapy group (group 4), first-step group (group 5), second-step group (group 6), third-step group (group 7) and Pulmicort Respules group (group 8). After the establishment of rat asthma model interfered with hormone and on third week the hormone was withdrawn successively at the dose of 0.1 mg/kg per week, and until 7 th week the hormone was withdrawn completely. The contents of serum IL-4 and IFN-γ were detected with ELISA method, and the expressions of IL-4 mRNA and IFN-γ mRNA in the lung tissue were examined by in situ hybridization in different groups. Results The contents of serum IL-4 and its lung mRNA expression were decreased significantly in different phases (P<0.01), and the content of serum IFN-γ and its lung mRNA expression were increased significantly in different phases (P<0.01) in group 4 compared with that of group 2 and group 3. There was no statistical significance in the comparison between group 4 and group 8 (P>0.05) at the end of 2nd week, but a great significant difference at the end of 7 th and 9 th weeks (P<0.01). Conclusion The 3-step sequential therapy can decrease, in rat asthma model interfered with hormone, the content of serum IL-4 and its lung mRNA expression, increase the content of IFN-γ and its lung mRNA expression, regulate the imbalance of IL-4 and IFN-γ, and relieve the inflammation of respiratory tract.

摘要:Objective To observe the influence of Yaotongshu Capsules on the expressions of p53, bcl-2, PCNA, TGF-β1 in the model of osteoarthritis (OA). Method To observe the expressions of p53, bcl-2, PCNA, TGF-β1 by the light microscope after immunohistochemical staining. Result There were no expressions of p53 and bcl-2 observed in the most samples of the normal group. After the model was established the sporadic expressions of p53 and bcl-2 were observed in cartilage transitional layer. After the administration of Yaotongshu Capsules the bcl-2 expression was increased gradually and p53 expression was decreased gradually. There was the sporadic PCNA expression observed in the normal group, and it was significant in synovial membrane in the model group. The PCNA expression in synovial membrane were decreased in groups with medium and high dose of Yaotongshu Capsules compared with that of the model group, but in surface layer it was increased. There was the sporadic TGF-β1 expression observed in the normal group, after the model establishment it was increased significantly and decreased in synovial membrane in the groups with high and medium dose of Yaotongshu Capsules. Conclusion Yaotongshu Capsules can delay the apoptosis of cartilage cells, inhibit the proliferation of synovial membrane, and promote cartilage plerosis through regulating the p53 and bcl-2 expressions of cartilage and the PCNA and TGF-β1 expressions of cartilage surface and synovial membrane.
摘要:Objective To investigate the protective effect and mechanism of tripterygium po1yglycoside (TII) on rats with adjuvant arthritis (AA). Methods The rat AA model was established and 30 SD male rats were divided randomly into the normal group, model group and TII group. The swollen of rat claw were measured, the histological changes were observed by the light microscope, the activity of superoxide dismulase (SOD), MDA level and T-AOC were detected by the biochemical method, the contents of TNF-α and PGE2 in claw were detected by the radioimmunoassay, and the expressions of COX-2 and NF-κB in cartilage of knee joint were detected by immunohistochemistry. [WTHZ]Results Compared with the normal group in the model group the activities of SOD and T-AOC were decreased (P<0.01), MDA level was increased, the contents of TNF-α and PGE2 were increased, and the expressions of COX-2 and NF-κB were increased. In the TII group the activities of SOD and T-AOC were increased, the MDA level, the contents of TNF-α and PGE2, and the expressions of COX-2 and NF-κB were decreased. The result of the light microscope showed that in the TII group the hyperplasia of synovial membrane and inflammatory infiltration was relieved. Conclusion Tripterygium po1yglycoside has a protective effect on rat AA model. One of its mechanisms may be related to its antioxidation and inhibitory effects on inflammatory infiltration.
摘要:Objective To investigate the mechanism of acupoint dressing therapy for treating asthma. Methods To divide randomly 40 guinea pigs into the normal control group (group 1), asthma model group (group 2), acupoint dressing group (group 3) and dexamethasone group (group 4, each n=10). The apoptosis of bronchoalvealar eosinocytes (EOS) was detected by TUNEL, and the expressions of Fas, FasL and bcl-2 genes were detected by immunohistochemical assay. Result The difference was not significant (P>0.05) in EOS apoptosis index, expressions of Fas, FasL and bcl-2 genes between group 2 and group 1. The EOS apoptosis index, expressions of Fas and FasL in group 3 and group 4 were higher significantly than that in group 2 (P<0.01). There was a significant difference between group 4 and group 3. The index and expressions in group 4 was higher significantly than that of group 3 (P<0.01). The bcl-2 expression in group 3 was lower than that of group 2 (P<0.05). The difference was not significant between group 4 and group 2 (P>0.05). Conclusion The therapy of acupoint dressing can increase the expressions of Fas and FasL genes in bronchoalvealar tissue, decrease bcl-2 expression and promote the apoptosis of EOS, which may be its one of the mechanism for treating asthma.

摘要:The animal model of blood stasis syndrome is an important technological platform for the study on the biological foundation of the blood stasis syndrome and the therapy of activating blood to removing stasis. The model in the past was divided into two types. One was the type of disease cause, which was model of disease cause, simulated the cause of blood stasis, including wound, congealing cold, stagnating qi, qi- insufficiency, yin-deficiency, yang-insufficiency, bleeding and natural old. Another one was the type of pathology or pathophysiology, which was the model of integration of disease and symptoms, simulated the vascular or blood pathology or pathophysiology of western medicine, including vascular obstruction, microcirculation abnormal, thrombosis and the obstacle of hemorrheology and hemodynamics. These two types of model, however, were evaluated just on the basis of above-mentioned changes of biochemistry indexes.The biological superficial signs are ignored, which accord with the clinical manifestations of blood stasis syndrome, such as local swelling and pain, vascular dilatation and hyperaemia in conjuntiva, dark or dark red around the lip, in auricle, nail, tail and tongue and animal behaviors, and result in a phenomenon of “all syndromes with blood stasis and all herbal drugs for activating blood”, and give an impact to the research of Chinese medical fundamental theory. So it will be a key problem demanding prompt solution to establish the model of blood stasis syndrome with the biological superficial signs.

摘要:Objective To investigate the mechanism of cisplatin (CDDP) inducing premature ovarian failure (POF, deficiency syndrome of both liver and kidney yin). Method The changes of general state, weight, surface temperature, ovulation, genesial hormones and cAMP were observed in mice after the introperitoneal injection of CDDP. Result The result showed that the mouse weight was lost, water and food intake was decreased, and hypourocrinia, fervescence and ovulation inhibition were observed in mice after the administration of CDDP, while in the group with 3 mg/kg of CDDP, the levels of serum estradiol (E2) and progestin (P) were decreased, and the levels of serum follicle-stimulating hormone (FSH), luteinizing hormone (LH) and cAMP were increased. [WTHZ]Conclusion The model of POF ( deficiency syndrome of both liver and kidney yin) induced by the introperitoneal injection of CDDP has the same pathogenesis and biological behaviors with clinical pathology and symptoms. The incretion and changes of biochemical indexes are similar to those in clinic in mice injected 3 mg/kg of CDDP. This method of model establishment is suitable for systematic study on POF (deficiency syndrome of both liver and kidney yin).

摘要:Objective To observe the protective effect of Epimedium Flavonoids Injection (EFI) on cardiocyte lesion induced by H2O2. Methods[WT] The cardiocytes of neonatal rats were cultured in vitro. The proliferation of cardiocytes was detected by MTT assay, the apoptosis of cardiocytes detected by flow cytometer and the ultrastructure of cardiocytes was observed by the transmission electron microscope (TEM). Results H2O2 increased the inhibitory rate of cardiocyte proliferation in a dose-dependent way. The cardiocyte apoptotic rate was (23.94±3.52) % twelve hours after 100 μmol/L of H2O2 administration, which was higher than that of the control group [(1.98±1.22) %]. The cardiocyte ultrastructure showed the apoptotic changes. EFI in doses of 100, 200 and 400 mg/L decreased the inhibitory rate of cardiocyte proliferation in a dose-dependent way, decreased significantly the cardiocyte apoptotic rate respectively \[(15.12±3.01) %, (9.38±3.14) % and (4.49±0.52) %\], and EFI also relieved the change of cardiocyte ultrastructure. [WTHZ]Conclusion EFI has a protective effect on cardiocyte lesion induced by H2O2.
摘要:Objective To study the influence and mechanism of polysaccharides of Agaricus Blazei Muill (PAB) on the apoptosis of human liver cancer cell, Bel-7402. Methods The co-culture supernatant was prepared by using PAB in different concentrations and mouse splenic lymphocytes. The apoptosis, p53 expression and Ca 2+ level changes of Bel-7402 cells were analysed by flow cytometry in different groups. The cell ultrastructure was observed by the transmission electron microscope (TEM), and the p53 expression was observed by fluorescent microscope. Result The apoptosis rate and Ca 2+ level in Bel-7402 cells increased significantly detected by flow cytometer in the treatment group compared with that of the control group (P<0.01). The p53 expression was not different significantly detected by flow cytometer and fluorescent microscope (P>0.05). The typical morphological changes of apoptosis were observed by TEM. Conclusion PAB can induce the apoptosis of Bel-7402 cells.

摘要:Objective To establish a method for the quantitative determination of the contents of trifolirhizin in Sophora flavescens Ait. and the effective fraction of Kushen Decoction by HPLC. MethodsThe HPLC procedure was used in the determination as follows: the chromatographic column was Phenomenex Luna C 18 (250 mm × 4.6 mm, 5 μm), the mobile phase was acetonitrile: phosphoric acid water solution \[H3PO3-H2O (2:530)] (26:74) with a flow rate of 1 mL/min, and the detection wavelength was 310 nm. [WTHZ]Result The result showed that trifolirhizin had a good linear relationship in the range of 0.1616~0.8080 μg (r=0.9994). The average recoveries for trifolirhizin in three batches of samples were 98.97%, 99.93% and 97.94% respectively, RSDs were 1.80%, 1.84% and 1.67% respectively (n=3). Conclusion The procedure was easy, sensitive with a good result and repeatability. It can be used to control the qualities of Sophora flavescens Ait. and the effective fraction of Kushen Decoction.
摘要:Objective To establish a method for quantitative determination of puerarin, 3′- methoxyl- puerarin, mirificin, daidzin and paeoniflorin in Yishendaluo Formula by RP-HPLC. Method The RP-HPLC procedure was used in the determination as follows: the chromatographic column was ODS HYPERSIL (250 mm×4.6 mm,5 μm), the gradient elution program was acetonitrile: 4‰ H3PO4 water, the detection wavelength was 240 nm, and the column temperature was the room temperature. Results The results showed that puerarin, 3′-methoxyl-puerarin, mirificin, daidzin and paeoniflorin had a good linear relationship respectively when the concentration was between 0.152 μg~0.912 μg, 0.184 μg~1.104 μg, 0.240 μg~1.200 μg, 0.142 μg~0.852 μg, and 0.204 μg~1.224 μg respectively (r=0.999 5, 0.999 6, 0.999 3, 0.999 6 and 0.999 7 respectively, n=6). The average recoveries of the samples were 100.09%, 101.12%, 100.69%, 100.05% and 99.19% respectively, RSDs were 2.00%, 2.97%, 2.90%, 1.62% and 2.95% respectively. Conclusion The procedure, which is easy, accurate and reliable, can be used to control the quality of the effective fraction of Yishendaluo Formula. [JP4]
摘要:Objective To analyse retrospectively the influence of Chinese herbal drugs on the long-term prognosis in patients with first onset of acute myocardial infarction (AMI). Methods To select 162 AMI patients as the study objects, hospitalized from Jan. 1996 to Apr. 2002 and gotten through the acute phase (28 days) in the Cardiology Department of Dongzhimen Hospital and Dongfang Hospital of Beijing University of Chinese Medicine. The combined endpoint events (including death or severe cardio- cerebrovascular events) of the patients were followed up by regular visits. The Kaplan-meier method was used in mono-factor analysis to make the survival curve, the log-rank method was used in comparison test, and the Cox regression analysis was used in multi-factor analysis. Results The outcomes of mono-factor analysis showed that the patients taking Chinese herbal drugs had a higher survival rate without events (P<0.01) than the patients not taking Chinese herbal drugs. The outcomes of multi- factor analysis showed that there was a correlation between age, anterior myocardial infarction, cardiac insufficiency, arrhythmia, diabetes history and combined endpoint event risk, while after the administration of Chinese herbal drugs and ACEI there was a negative correlation. The relative risk (RR) of Chinese herbal decoctions was 0.093 (95%, CI =0.029~0.294, P<0.001). Conclusion The risk factors of AMI long-term prognosis included age, heart failure, anterior myocardial infarction, arrhythmia after infarction and diabetes history. The early administration of Chinese herbal decoctions and ACEI is a protective factor for AMI long-term prognosis. The study has offered the testimonies based on evidence based medicine for AMI treatment with Chinese medicine from the point of retrospective study.

摘要:Objective To survey the syndrome distribution of the patients infected HIV by intravenous drug. Method To draw up “Survey Table of Chinese Medicine Symptoms of Patients Infected HIV” according to the Chinese medical theory, relative documents and expert experiences, and then the table was used to survey the syndromes of 175 drug addicts infected HIV. The control group included 125 non-infected drug addicts was established. The result was counted and analysed. Result There were 24 symptoms with significant difference between the case group and control group. The difference was great significant in 8 symptoms, including non-desire to speak due to qi-insuficiency, headache, excessive phlegm and night sweating. The patients in the case group had a tend to emaciation and more serious symptoms than those in the control group. Conclusion The drug addicts infected HIV have more serious symptoms, including qi-insuficiency, blood deficiency, yin-deficiency, excessive phlegm and dampness, and negatively immunity.

摘要:Objective To observe the curative effect of a series of Ainaiji Granules on AIDS. Method According to AIDS diagnostic standard 106 patents with diagnosed AIDS or infected HIV were divided into the Shangjiao group, Zhongjiao group and Xiajiao group, and then Ainaiji Granules I, II and III were given to them respectively. The auto-control method was used to observe the curative effects before and after the treatment. Results According to the curative effect standard the observation results showed that the symptoms and signs were disappeared in 20, 22 and 17 cases in three groups respectively, significantly relieved in 16, 10 and 11 cases, relieved in 2, 1 and 4 cases, and not changed in 0, 1 and 2 cases. The results of absolute counts of CD4 in three groups showed respectively as follows: effective in 28, 27 and 28 cases, and ineffective in 10, 7 and 6 cases. The results of virus carrying capacity in three groups showed respectively as follows: effective in 19, 29 and 28 cases, and ineffective in 19, 5 and 6 cases. Conclusion Ainaiji Granules has curative effects on relieving symptoms and signs, improving immunity and decreasing virus carrying capacity of AIDS patients.

摘要:ObjectiveTo investigate the curative effects of differentiation and treatment of Chinese medicine on renal function indexes and symptoms of renal failure of diabetic nephropathy. Method To divide randomly 84 cases with diagnosed diabetic nephropathy into 3 groups, On the basis of routine treatment group A was given Chinese medical therapies after the syndrome differentiation, group B was given losartan, and group C was given Chinese and western therapies. The renal function indexes (including creatinine clearance, serum creatinine and ureanitrogen) were observed and the curative effect on a single symptom was evaluated after 3 months. Result The creatinine clearance was increased significantly in group A (P<0.05). In the symptom evaluation the curative effects on symptoms in group A and group C were better than that in group B. Conclusion Chinese medicine has some advantages in improving renal function and relieving specific symptoms, and also has a good prospect for relieving patient’s symptoms and promoting life quality.
