      摘要:The subject of TCM research is a complex adaptive system(CAS) including the syndrome differentiation and treatment,drug application and formula composition,and entirety accommodation.The study and practice of the complexity science provides a new method for TCM research.The theoretic basis of the application of CAS is analyzed here on the base of the introduction to CAS theory and modeling.The success key is that CAS is applied in the modeling of TCM diagnostic and treatment instead of in the modeling of entity.The modeling way of CAS based on multi-agent provides the thinking train and method for TCM theoretic research based on CAS.

      摘要:Objective To observe the influence of hyocholic acid on the expression of intercellular adhesion molecule-1(ICAM-1) of brain microvascular endothelial cells(MVEC) in rats after ischemia and reperfusion injury and to investigate the mechanism of hyocholic acid for inhibiting the inflammatory reaction in cerebral ischemia.Methods The rat cerebral MVEC were cultured in vitro and the ischemia and reperfusion injury was simulated by using oxygen and glucose deprivation(OGD).The expressions of ICAM-1 and mRNA were detected semiquantitatively by immunocytochemical stain and RT-PCR.Results The expressions of ICAM-1 and mRNA were obviously increased in the model group than those in the normal group(P<0.01).Hyocholic acid reduced significantly the expression of ICAM-1 compared with the outcome of the model group(P<0.05).Conclusion Hyocholic acid may inhibit the inflammatory reaction in cerebral ischemia through intervening the expression of ICAM-1.

      摘要:Objective To observe the changes of renal function and histomorphology in rats treated with aristolochiae manshuriensis(AM),Longdanxiegan Decoction(LD) and Modified Longdanxiegan Decoction(MLD,removed angelica sinensis,rehmannia dride rhizome and glycyrrhizae uralensis).Method All SD rats were randomly divided into the normal control group(group 1),AM group(group 2),LD group(group 3) and MLD group(group 4).Group 1 was given distilled water orally for 8 weeks,and group 2,3 and 4 were given respectively AM(6 g/kg),LD(17.5 g/kg,containing AM 6 g/kg) and MLD(14.5 g/kg,containing AM 6 g/kg) all for 8 weeks.The biochemical indexes in serum and urine,and renal histomorphological changes in rats were detected at the end of 8th week.Result In group 2,3 and 4 the rats all had significant renal lesion,which was the most serious in group 2,and less in group 3,but in group 4 the renal lesion was serious too and some indexes was even higher than those in group 2.Conclusion AM,LD and MLD all can induce the renal lesion and MLD has stronger nephrotoxicity.It suggests that the toxicity of formulas containing AM is not as the same as that of AM.Angelica sinensis,rehmannia dride rhizome and glycyrrhizae uralensis contained in LD may inhibit the nephrotoxicity of AM.

      摘要:Objective To investigate the inhibitory effect of Shenluotong serum on the different compositions of extracellular matrix(ECM) and the influence on the connective tissue growth factor(CTGF) in mesangial cells(MCs).Methods The contents of fibronectin(FN) and collagen Ⅳ(Col Ⅳ) in ECM were detected by enzyme linked immunosorbent assay(ELAIS),and the expression of CTGF mRNA was measured by semi-quantitative reverse transcription-polymerase chain reaction(RT-PCR) based on cultured in vitro rat glomerular mesangial cells and the method of serum pharmacology.Results The contents of FN、Col Ⅳ in ECM and expression of CTGF mRNA were showed positive correlation and all inhibited by Shenluotong significantly.Conclusion Shenluotong can diminish the accumulation of ECM,and inhibit the secretion and expression of CTGF,which maybe one of the mechanisms of preventing the kidney from sclerosis.

      摘要:Objective To investigate the influence of microemulsion of Rhizoma Chuanxiong based on hemorheological indexes in rat model of blood-stasis syndrome.Method The rat model of blood-stasis syndrome was established by using adnephrin,which was then given different doses of the microemulsion of Rhizoma Chuanxiong and Chuanxiong Decoction.The indexes,such as the clotting time(CT),blood viscosity,hematocrit,erythrocyte aggregation and RBC deformation,were observed.Result The microemulsion of Rhizoma Chuanxiong had the effects of prolonging the clotting time,decreasing blood viscosity,relieving RBC deformation and reducing erythrocyte aggregation index in the rat model of blood-stasis syndrome.The group of the microemulsion of Rhizoma Chuanxiong had better outcomes than the group of Chuanxiong Decoction in some indexes.Conclusion The microemulsion of Rhizoma Chuanxiong has good effects of improving hemorheological indexes in the rat model of blood-stasis syndrome,and the effects are better then those of Chuanxiong Decoction.

      摘要:Objective To observe the antiviral effect of Dureping Injection in vitro and in vivo experiments.Methods The method of crystal violet stain was used to observe the antiviral effect in vitro of Dureping Injection(in different administrative methods) on FM1 strain of influenza virus A,time-effect relationship and the inhibitory effect on mouse viral pneumonia.Results Dureping Injection inhibited obviously the virus from adsorbing to cells and virus copy and proliferation.It also killed the virus directly at the concentration of ≥0.18 g/L.Its antiviral efficiency went to the peak on 48th hour and could be kept at the concentration of ≥0.27 g/L along with the time,and tended to decrease at a low concentration.Dureping Injection had an inhibitory effect on the pneumonia of influenza virus in mice.Conclusion Dureping Injection has a significant antiviral effect in vitro and in vivo.

      摘要:Objective To study the influence of ground cherry’s extraction on antioxidant ability in aging rats induced by D-galactose.Methods All 75 Wister male rats(6 weeks old) were randomly divided into five groups,including the young control group(group 1),model group(group 2),low-dose extraction group(10 mg/kg,group 3),middle-dose extraction group(15 mg/kg,group 4) and high-dose extraction group(20 mg/kg,group 5),besides other 15 Wister male rats(16 months old) were included in the old control group(group 6).The rats in group 2,3,4 and 5 were given injection of D-galactose for 5 months.After 5 months the contents of serum malonaldhydic acid(MDA) and liver MDA,activities of superoxide dismutase(SOD) and glutathion peroxidase(GSH-Px) in all groups were detected and compared.Results Compared with the young control group,the content of serum MDA,liver MDA were significantly increased(P<0.01) and the activities of SOD and GSH-Px were significantly decreased(P<0.01) in group 2,but those changes were as the same as those of group 6.The contents of serum MDA and liver MDA in group 3,4 and 5 were significantly decreased compared with those in group 2(P<0.01),while the activities of SOD and GSH-Px were significantly increased(P<0.01).Conclusion The extraction of ground cherry can increase the antioxidantive ability of rats in order to delay the consenescence induced by D-galactose.

      摘要:Objective To compare the influence of the therapy of clearing lung and dissolving phlegm,tonifying qi and activating blood,combined therapy,different stages of medicine administration,4 groups of Chinese herbal drugs and positive control drugs on vessel neogenesis of pulmonary fibrosis.Methods The male Wistar SPF rats(n=77) were randomly divided into 7 groups,including the sham-operauion group(group 1),model group(group 2),group treated with clearing lung and dissolving phlegm therapy(group 3),group with tonifying qi and activating blood therapy(group 4),group with combined therapy(group 5),group treated in different stages(group 6) and group with hydrocortisone(group 7).After 28 days the contents of vascular endothelial growth factor(VEGF) and angiopoietin-2(Ang-2) were detected by avidinbiotin-peroxidasecomplex method(ABC) and semiquantitative grade analysis.The integral optical density value(IODV) was calculated by the integral optical density method.The changes of microvessel density in fibrosis lung tissue were observed.Results In group 2 the contents of VEGF and Ang-2 were increased while decreased 28 days after the treatment with combined therapy.The results of grade and IODV after the treatment had a significant different compared with those of group 2(P<0.01).On 28th day the microvessel density in group 2 was much higher than that of group 1 which indicated that pathological vessel neogenesis of pulmonary fibrosis was activity(P<0.01).In different treatment groups there were significant different among group 4,5 and group 2(P<0.01).Conclusion The therapy combined clearing lung and dissolving phlegm,tonifying qi and activating blood can inhibit the secretion of VEGF and Ang-2 and microvessel neogenesis to delay the development of pulmonary fibrosis.

      摘要:Objective To investigate the relationship between the expression of transforming growth factor-β(TGF-β) mRNA and Bushentongluoshugan Formula in penile erection in senescent rats.Method All 32 mature male rats were randomly divided into: the senescent rat group(group 1),Bushentongluoshugan Formula group(group 2),testosterone propionate group(group 3) and control group(group 4,the rats were 2 months old).On the 24th day the penile tissue was taken from the rats and the expression of TGF-β mRNA was detected by PCR technique.Result In group 1 the expression of TGF-β mRNA was increased than those of other three groups.The groups with decreased the expression of TGF-β mRNA were ordered as follow ascent sequence:group4,group 1,group 3,group 2.The controlled amount of TGF-β mRNA in group 4,3 and 2 was lower and had a significant difference respectively compared with that of group 1(P<0.01).There was no significant difference between group 3 and 2(P>0.05).Conclusion Bushentongluoshugan Formula may play an important role in the penile erection through regulating the expression of TGF-β mRNA.

      摘要:Objective To establish a method for determinating the substitution degree of N-succinyl-chitosan by conductometric titration.Method The sample of N-succinyl-chitosan was dissolved in excess HCl,which then was titrated by standard NaOH.The conductivity of the solution was detected and the substitution degree of N-succinyl-chitosan was calculated.Result The RSD of the method was 1.45%.The substitution degree had no significant difference when the conditions of the experiment were changed.Conclusion The method is easy with a good reproducibility,and prompt to be performed.

      摘要:Objective To establish a method for the quantitative determination and thin layer chromatographical identification(TLCI) of hesperidin in Citrus medica L.in order to provide a scientific base for the quality standard of Citrus medica L.Method The determination was carried out by HPLC on a Kromasil-C18 column and the sample was extracted with methanol by means of distillation for 60 minutes.The mobile phase was methanol-water-acetic acid(30∶68∶3) and the detective wavelength was 284 nm.Result The result showed that hesperidin in sample solution was well separated and had a good linear relationship in the concentrations between 0.0950 to 1.4688 μg(r=0.999 9).The average recovery was 101.95 %,and RSD was 2.15%(n=6).Conclusion The method is accurate and reliable,which can be used in the qualitative determination of hesperidin.

      摘要:Objective To establish a high performance liquid chromatography-evaporative light-scattering detection(HPLC-ELSD) method for the determination of betaine in Cortex lycii.Methods The procedure was performed on a Luna C18 column(250 mm×4 mm,5μm) at 30℃.The mobile phase was water-acetic acid(99.9∶0.1).The flow rate was 0.15 mL/min.An evaporative light-scattering detector was used and the parameters were set as follows: drift tube temperature was 100 ℃ and gas flow rate was 2.50 L/min.Result The result showed that betaine had a good linear relationship in the concentrations between 0.486 μg to 2.430 μg(r=0.999 9) with an average recovery of 97.4%(RSD=1.3%,n=5).Conclusion The method is accurate with a good repeatability and can be used in the quality control of Cortex lycii.

      摘要:Objective To investigate the TCM syndrome characteristics of mild cognitive impairment(MCI) and the relationship between MCI and the Mini-Mental Status Examination(MMSE) in order to provide a scientific base for MCI treatment.Methods The patients with MCI were chosen and accepted the epidemical survey through the questionnaire of TCM common syndromes of MCI,and then the ground-plan of TCM syndrome differentiation was worked out and the patients were given TCM differentiation on the base of clinical experiences and "State Standard of the People’s Republic of China: Clinical Diagnostic and Therapeutic Terminology of Syndromes of Chinese Medicine".The characteristics of TCM syndromes in the patients with MCI and the relationship between these characteristics and MMSE were discussed on the base of combination of MMSE,epidemiology and mathematical analysis.Results In MCI patients the most common syndrome was the syndrome of deficiency of liver-yin and kidney-yin,followed by the syndrome of phlegm clouding the upper orifices,brain paralysis and marrow deficiency,constipation due to blood stasis,and deficiency of both qi and blood.After Spearman’s analysis a negative correlation was showed between TCM syndromes and MMSE.Conclusion In MCI patients the common syndromes include the syndrome of deficiency of liver-yin and kidneyyin,followed by the syndrome of phlegm clouding the upper orifices,brain paralysis and marrow deficiency,constipation due to blood stasis,and deficiency of both qi and blood,which are correlated to the cognitive impairment.It provides an objective base for the syndrome differentiation and treatment of MCI.

      摘要:Objective To study the revival effect and mechanism of electrostimulation in the Pericardium Channel of Hand Jueyin in coma patients induced by severe craniocerebral trauma.Methods The patients(n=18) were randomly divided into the control group and treatment group.The control group was treated with the routine therapy and the treatment group was given the electrostimulation in the Pericardium Channel of Hand Jueyin besides the routine therapy.Before the initial treatment all patients were given the examination by transcranial Doppler(TCD) to detect their systolic velocity(Vs),diastolic velocity(Vd),mean velocity(Vm) and pulsitility index(PI),and then the treatment group was given the electrostimulation in the Pericardium Channel of Hand Jueyin immediately for 30 minutes and detected by TCD for the second time in the same body position and condition.The instant changes of blood flow before and after the electrostimulation were compared.After that the treatment group was given the electrostimulation of 30 minutes every day for one month and the same indexes were detected and compared.The electrostimulation was continued for 3 months and Glasgow Coma Scale(GCS),recovery of nervous function and lingual function were compared.Results The instant effectiveness was observed on the disorder of blood flow rate of basilar artery in the treatment group.After one month the blood flow rate in the treatment group was significantly increased compared with that of the control group.After 3 months the results of GCS,scales of lingual function and living quality in the treatment group were much better than those in the control group.Conclusion The electrostimulation in the Pericardium Channel of Hand Jueyin can promote the blood flow rate of basilar artery in coma patients induced by severe craniocerebral trauma,and has a revival effect.
