摘要:Objective To observe the reversibility of liver injury reduced by Fructus Xanthii and the influence of Fructus Xanthii on the synthesis of liver DNA.Methods The Fructus Xanthii decoction was given to mice orally for 4 weeks and after stopping for 2 weeks the influence of it on the body weight and liver function in mice was observed. The radioactivities of serum and tissue were studied by 3H-TdR tracing method.Results Fructus Xanthii had an inhibitory effect on the increase of mouse body weight but disappeared after stopping administration of Fructus Xanthii. After two weeks the blood biochemistry indexes (like AST) were higher obviously in the Fructus Xanthii group with medium or high dose than those in the blank group.Fructus Xanthii in low or medium dose injured liver DNA and in high dose influenced DNA synthesis. Conclusion The injury of liver function induced by Fructus Xanthii has a reversibility in a certain range of dose.Fructus Xanthii has an influence on DNA synthesis of liver tissue.
摘要:Objective To investigate the preventive and therapeutic effects of Jiaweibugan Decoction on peripheral neuropathy in experimental diabetic rats. Methods The diabetic rat model was established by streptozotocin (STZ). The rats were killed on 4th or 8th week from the beginning of treatment respectively, and serum content of malondialdehyde (MDA) was determined by chromatometry, the expressions of nerve growth factor (NGF) protein and mRNA of sciatic nerve was detected by immunohistochemical assay and reverse-transcriptase polymerase chain reaction (RT-PCR). Results ① The serum level of MDA of the model group on 4th or 8th week was higher than that of the normal control group (P<0.01) and increased gradually along with the disease course. Jiaweibugan Decoction could decrease the serum level of MDA. ②The results of immunohistochemical assay and RT-PCR showed that NGF expression of the model group on 4th or 8th week was lower than that of the normal control group (P<0.01), and enhanced after the intervening of Jiaweibugan Decoction and taurine (P<0.05). Jiaweibugan Decoction had a better effect than taurine with a significant difference (P<0.05). Conclusion Jiaweibugan Decoction has a preventive and therapeutic effect on peripheral neuropathy in experimental diabetic rats.
摘要:Objective To observe the influence of Qingkailing Injection on the contents of transforming growth factor-β (TGF-β) and heats shock protein 70 (HSP70) of brain in different cerebral ischemic periods, and to investigate, on the base of nerroprotective factor, the mechanism of effective components of Qingkailing Injection for antagonizing cerebral ischemic lesion.Methods The adult male SD rats were randomly divided into the sham-operation group, model group, different groups of effective components of Qingkailing Injection and combined formula groups. The rat model of focal cerebral ischemia was copied with the embolism of persistent middle cerebral artery for 12 or 24 hours by using thread-occluding method. The contents of TGF-β and HSP70 in brain homogenate were detected by enzyme-linked immunosorbent assay (ELISA).Results The contents of TGF-β and HSP70 in rat brain were obviously increased 12 or 24 hours after cerebral ischemia. The difference was significant statistically (P<0.05). The expression of TGF-β in brain was inhibited in the baicalin group, nacre group and combined formula groups 12 hours after cerebral ischemia (P<0.05), and was improved in the cholic acid group and nacre group 24 hours after cerebral ischemia (P<0.05). The HSP70 expression did not show any significant biologic effectiveness in different groups of effective components of Qingkailing Injection 12 hour after cerebral ischemia (P<0.05), and the expression was improved in the combined formula groups and the baicalin group (P<0.05). Conclusion The effective components of Qingkailing Injection has a preventive effect on focal cerebral ischemia of rat. One of the mechanisms may be that the effective components can improve the expressions of TGF-β and HSP70 with a chronological feature.

摘要:Objective To study the influence of 3, 4-oxo-isopropylidene-shikimic acid (ISA) on lipid peroxidation of human umbilical vein endothelial cells (HUVEC) induced by hydrogen peroxide (H2O2) in vitro. Methods The concentration of malonaldehyde (MDA) in HUVEC was detected by thiobarbituric acid (TBA) method, the activities of superoxide dismutase (SOD) was detected by xanthine oxidase system and catalase (CAT) was detected by visible spectrum assay. The clearance (C) of superoxide anion and OH radicals by ISA were tested by free radical prehensile experiment in vitro.Results The lipid peroxidation induced by H2O2 (200 μmol/L for 6 hours) was alleviated obviously in HUVEC cultured with ISA (10-5, 10-4 mol/L) for 6 hours, MDA was decreased and the activities of CAT and SOD were protected. The activities of SOD and CAT were improved after HUVEC incubation with ISA for 12 hours. ISA (10-510-3 mol/L) had an obvious clearing effect on superoxide anion, showed a relationship between quality and effectiveness. ISA (10-3 mol/L) could also clear OH radicals.Conclusion ISA has an antagonizing effect on lipid peroxidation of vein endothelial cells in vitro, but no direct influence on the activities of SOD and CAT.
摘要:Objective To observe the depressor effect of Zhenganxifeng Decoction in rats with essential hypertension and the influence on brain endothelin (ET) content and the expression of peroxisome proliferating activator receptor γ (PPARγ) mRNA. Methods All 24 male rats with essential hypertension (14 weeks old) were randomly divided into the essential hypertension control group, Zhenganxifeng Decoction group, enalapril group, and other 8 male WKY rats were taken as the WKY control group at the same time. The blood pressure was recorded by the sixteen leads physiologic record system, ET contents in serum and brain were detected by radioimmunoassay, and brain PPARγ mRNA expression was detected by reverse transcription-polymerase chain reaction (RT-PCR) after 8 weeks.Results The blood pressure and brain ET content were increased and PPARγ mRNA expression was decreased in the essential hypertension control group compared with those in the WKY control group. Zhenganxifeng Decoction reduced the ET content and improved PPARγ mRNA expression in rats with essential hypertension.Conclusion Zhenganxifeng Decoction protects brain by activating PPARγ mRNA expression and reducing brain ET content.

摘要:Objective To study the influence of berberine (Ber) on cardiovascular function in rats with hypertrophic cardiomyopathy (HCM) induced by L-thyroxin.Methods The model with HCM was established by hypodermic injection of L-thyroxin in SD rats.Ber was given to rats orally (5 or 10 mg/kg) for two weeks, and rat blood pressure was measured.Two weeks later the rat cardiac function was examined by catheterization in left ventricle of heart and nitrous oxide (NO) contents in serum and myocardium were determined.Result Ber (5 or 10 mg/kg)decreased the blood pressure of rat model remarkably and Ber (10 mg/kg) can reduce heart rate (HR), the maximum left ventricle systolic pressure (LVSP), increased left ventricle end-diastolic pressure (LVEDP), reduced +dp/dtmax, and increased NO content of left ventricle muscle. The NO content of left ventricle muscle was correlated to LVSP, LCEDP, +dp/dtmax, and HR.Conclusion Ber can prevent rats of HCM model from blood pressure increase and heart disorder induced by L-thyroxin. This effect is correlated to the increase of NO content.
摘要:Objective To study the effect of Gubenyiliu Formula II (GFII) on Lewis lung carcinoma and its combinative effect with cyclophosphamide (CTX). Method All C57BL/6 mice (n=60) were randomly divided into six groups as follows: the control group, CTX group (intraperitoneal injection of CTX in dose of 30 mg/kg), GFII group in medium dose (M group with GFII in dose of 35 g /(kg·d) orally), GFII group in high dose (H group with GFII in dose of 70 g /(kg·d) orally), group with medium-dose GFII and CTX (M+CTX group) and group with high-dose GFII and CTX (H+CTX group, each n=10). The effect of GFII on Lewis transplantation tumor and metastatic tumor, and combinative effect of GFII with CTX were observed.Result The inhibitory rate (IR) in M group and H group was 23.35% and 36.82% respectively, and the weight of tumor was (2.82±1.56) g and (2.32±1.06) g respectively, which was lower than that in the control group (3.68±1.10) g,P<0.05. The inhibitory rate in M+CTX group and H+CTX group was 71.49% and 67.13% respectively without significant difference compared with that in CTX group (63.35%) but the combinative effect was 0.85<q<1.15 which showed an additive effect. The cancerometastasis rate was 80% in M+CTX group and 70% in H+CTX group with an insignificant difference compared with that of CTX group (70%). The metastases were less in H+CTX group (2.8±1.7) than those in the control group (5.1±1.4),P<0.05.Conclusion GFII has an inhibitory effect on Lewis transplantation tumor in C57BL/6 mice. The combination of GFII and CTX shows an additive chemotherapeutic effect but no obvious inhibitory effect on metastatic tumor.

摘要:Objective To investigate the influence of Jiusuyu Suspension on hippocampus acetylcholinesterase (AchE) activity and striatum dopamine (DA) and 5-hydroxytryptamine (5-HT) contentsin acute alcoholism mice.Method The mouse model of acute alcoholism was established by giving Hongxing Erguotou Spirit (containing 56 per cent alcohol) to mice orally. The influence of Jiusuyu Suspension on drunken time, drunken rate, hippocampus AchE activity and striatum contents of DA and 5-HT were observed and compared with Gehuajiecheng Decoction.Result The rats of the model group showed an exciting at the first and repressive manifestations then. The activity of hippocampus AchE and striatum contents of DA and 5-HT in the model group were increased compared with those in the normal group (P<0.01). The drunken rate was obviously decreased (P<0.05) and drunken time was prolonged (P<0.05 or P<0.01) in the groups with high or low dose of Jiusuyu Suspension compared with those in the model group. The drunken time was longer in the group with high dose of Jiusuyu Suspension than that in Gehuajiecheng Decoction group (P<0.05). The contents of DA and 5-HT were obviously reduced in 3 treatment groups (P<0.05 or P<0.01). The activity of hippocampus AchE was obviously decreased in the group with high dose of Jiusuyu Suspension and Gehuajiecheng Decoction group (P<0.05 or P<0.01).Conclusion Jiusuyu Suspension can regulate the contents of brain monoaminergic neurotransmitter (DA, 5-HT) and AchE activity, and relieve the abnormal neurobehaviors of alcoholism.
摘要:Objective To study the relationship between the decocting quantity of saponin components from Radix Bupleuri and different component combinations of Sini Powder.Methods The reversed phased-high performance liquid chromatography (RP-HPLC) was used to determine the decocting quantity of saikosaponin-a and b2, and chromatoptometry was used to determine the decocting quantity of the total saikosaponin from the different component combinations of Sini Powder.Result The composition in Citrus aurantium L. increased the decocting quantity of saikosaponins.Conclusion The different component combinations of Sini Powder have a significant influence on the decocting quantity of saikosaponins.

摘要:Objective To establish the method for determining puerarin content from effective fraction of Naomaitong Formula.Method The chromatographic column was Hypersil ODS2 C18 (250 mm×4.6 mm,5 μm), the mobile phase was methanol to water (30∶70) with a flow velocity of 1.0 ml/min, and the detection wavelength was 250 nm in the phased-high performance liquid chromatography (RP-HPLC) procedure.Result The average recovery of puerarin was 99.72% (RSD=1.49%).Conclusion This procedure, which is easy and accurate, can be used as a quality control method for the effective fraction of Naomaitong Formula.
摘要:Objective To determine the method for extracting the liposoluble active principles from Quchiling Capsules and optimize the technique parameters.Method The supercriticle CO2 extraction was used and schisandrin content was taken as the index to test some action factors, such as temperature, pressure and CO2 flow rate. Result The influence of pressure on schisandrin content was significant, while the temperature and CO2 flow rate were not significant.Conclusion The best technique in supercriticle CO2 extraction for extracting liposoluble active principles from Quchiling Capsules is that the pressure is 15 MPa, temperature is 45℃ and flow rate is 25 L/h.

摘要:Objective To observe and test the effectiveness of TCM rehabilitation plan on apoplexia.Methods The patients with acute encephatorrhagia (n=180) and the patients with cerebral infarction (n=260) were chosen and then two kinds of patients were divided randomly into the control group and treatment group respectively. The treatment group was treated according to TCM rehabilitation plan and the control group was treated with basic therapies. The treatment course was 4 weeks. The kinetism, ability of daily living of the patient were assessed by FMA and Barthel index (MBI), and the level of nervous afunction was evaluated.Result After 4-week treatment the scores of FMA, MBI and nervous afunction of two kinds of apoplexia patients in the treatment group was better than those in the control group (P<0.05).Conclusion The plan can reduce handicap rate in patients with apoplexia, which can be a complement in TCM rehabilitation of apoplexia.
摘要:Objective To study the clinical manifestation and distribution characteristics of symptoms and TCM basic syndromes of subhealthy people group.Methods The questionnaires of subhealth with TCM feature were used, through the epidemiological investigation in subhealthy people group in Beijing area, combining the judgements from TCM clinical experts, and applying mathematical statistics, like cluster analysis and factor analysis, to discuss the main symptoms, syndrome key elements and basic syndrome characteristics.Results The analysis on 1 828 questionnaires, in Beijing subhealthy people group, showed that the main symptoms were fatigue, insomnia, emotional disorder and pain. The syndrome key elements were mainly deficiency, dampness, heat, depression, blood stasis, qi-stagnation, fluid consumption and phlegm. The dysfunction of zang-fu organs were mainly observed in heart, liver, spleen, kidney, gallbladder and stomach. The most common basic syndromes were mainly deficiency of both heart and spleen, liver stagnation and spleen insufficiency, dampness encumbering due to spleen-qi insufficiency,deficiency of liver-yin and kidney-yin, deficiency of both qi and yin, and qi insufficiency,blood stasis,accumulation of damp-heat,liver depression transforming into fire,deficiency of both spleen and kidney,and mixture of phlegm and qi. Conclusion The clinical manifestation characteristics in subhealthy people group showed as a symptom group with one main symptom and some other symptoms.The syndrome factors and basic syndrome distribution are mainly consumption in heart, liver and spleen with a regularity.

摘要:Purpose To study the change characteristics of interleukin 2 (IL-2) level in rats with immunity disorder and reveal the mechanism of immunomodulation by acupuncture through the observation on the influence and effective law of acupuncture, bondage and pathogenic factors on IL-2 level changes. Methods The electroacupuncture was used in six acupoints of Conception Meridian and Guanyuan (CV4) and the serum IL-2 level was determined by radioimmunoassay in the adjuvant arthritis (AA) model and bondage stress model.Results The IL-2 level had an increasing tend in AA model group and bondage model group compared with that in the normal group, but the difference was not significant (P>0.05). The IL-2 levels were all increased in AA model group with six acupoints, AA model group with CV4, bondage model group with six acupoints and bondage model group with CV4 compared with those in AA model group and bondage model group. The difference was significant in AA CV4 group (P<0.01) and too in bondage CV4 group (P<0.05).Conclusions Acupuncture can up-regulate IL-2 level in rats of AA model and bondage model, and regulate the body immunity. The acupuncture in CV4 has more effective result.
摘要:Objective To establish a model of hyperlipidemia combined with cerebral ischemia and to investigate the therapeutic effect of eletroacupuncture.Methods The rats were divided into the normal control group, model group and treatment group Ⅰ and Ⅱ.The model group and treatment group Ⅰ and Ⅱ were fed with high fat fodder to establish the hyperlipidemia model. The treatment group Ⅰ was given electroacupuncture in Sanyinjiao (SP6) and Fenglong (ST40) for 10 days, and then two groups were fed with FeCl3 to induce thromboangiitis obliterans of middle cerebral artery for establishing the model of hyperlipidemia combined with cerebral ischemia. These two groups were treated then with electroacupuncuture in SP6 and ST40, and acupuncture in Baihui (GV20) and Renzhong (GV26) for 7 days. The blood lipid indexes, neuron specific enolase (NSE) content in the rats were detected.Results The serum levels of cholesterol (CHO), low-density lipoprotein-C (LDL-C) and triglyceride (TG) of rats in different groups fed with high fat fodder 6 weeks after model establishment were increased with a significant difference compared with those in the normal control group (P<0.01). After acupuncture CHO level was decreased in the treatment group I and II with a great significant difference (P<0.01). The levels of TG and LDL-C in the treatment group I were decreased with a great significant difference (P<0.01). After HE stain the ischemia focus was constant with a lot of neuron degeneration and necrosis, and the manifestations of cerebral edema were observed too. The NSE detection result showed that there was a great significant difference between the model group and treatment group (P<0.01). The decrease of NSE in the treatment group I and II had a significant difference compared with that in the model group (P<0.01,P<0.05), and the increase in the treatment group II had a significant difference compared with that in the treatment group I (P<0.05). Conclusion The model of hyperlipidemia combined with cerebral ischemia was established successively. Electroacupuncture can decrease the levels of CHO, LDL-C and TG, relieve cerebral ischemia and reduce NSE concentration,and alleviate cerebral lesion in rats with hyperlipidemia combined with cerebral ischemia.
