摘要:Objective To study the Fatigue Self-assessment Scale for evaluating reasonably the characteristics and degree of fatigue.Methods The Fatigue Self-assessment Scale was developed through the following way: insurance of the object,aim and usage of assessment,study on fatigue connotation and category,selection of scale type and item form,confirmation of scale structure,determination of scale items,analysis and selection on the items,and analysis on the reliability and validity of the scale by the analytic method of reliability and searching factor,and frame equation in LISREL8.54.Results The results showed that the lowest coefficient of Cronbach’s alpha of different scale dimensions was 0.772 and highest 0.908.RMSEA=0.065,NNFI=0.95,and CFI=0.96 in the first part of the scale.RMSEA=0.10,NNFI=0.93,and CFI=0.96 in the second part of the scale.Conclusion The Fatigue Self-assessment Scale is good in reliability with a better structural validity.
摘要:Objective To investigate the influence of panaxnotoginsenosides on the expressions of renal endothelin-1(ET-1) protein and mRNA in rats with adriamycin nephropathy.Methods All 45 rats were randomly divided into the normal group,sham-operation group,model group,Benazepril group and panaxnotoginsenosides group.The model was established by removing the right kidney of rat and injecting adriamycin intraperitoneally.In the end of 10th week the rats were killed and then their renal tissue were taken out aseptically.After the renal tissues were stained by HE and Masson methods the pathomorphological changes of renal tissue were observed.The expressions of ET-1 protein and mRNA in renal tissue were analyzed by immunohistochemistry and in situ hybridization.Results In the model group the expressions of EF-1 and mRNA in renal tubular epithelial cells(TEC),glomerular mesangial cells(GMC) and glomerular epithelium cells(GEC) were strongly positive.The expressions of ET-1 and mRNA in the panaxnotoginsenosides group were decreased obviously with a statistical significant(P<0.05) compared with those in the model group.Conclusion Panaxnotoginsenosides can inhibit the expressions of ET-1 and mRNA in TEC,GMC and GEC in rats with adriamycin nephropathy,protect the kidney and postpone the development of chronic renal failure.
摘要:Objective To study the protective effect of Wenmaitong(WMT) on vascular endothelial cells(VEC) from injury induced by hyperlipidemic serum to investigate the therapeutic mechanism of WMT in treating arteriosclerosis obliterans(ASO).Methods Hyperlipidemic serum was used to establish injured model of human umbilical vascular endothelial cell(HUVEC).Sera with WMT or its decomposed formulas,warming yang and boosting qi part formula and promoting blood circulation part formula,were applied with hyperlipidemic serum together.The morphologic changes of HUVEC were observed under the light microscope and HUVEC activity was determined by MTT method.The contents of lactate dehydrogenase(LDH),nitrogen monoxidum(NO) and endothelin(ET) in the supernatant of HUVEC were detected by spectrophotometer,nitrate reductase method and radioimmunoassay respectively.Results WMT and its decomposed formulas made morphologic changes of HUVEC induced by hyperlipidemic serum trend to normal,increased the activity of HUVEC and lessened the injury of cell membrane.NO content in the supernatant was increased by WMT and its decomposed formulas,while ET content in the supernatant was decreased.The effects of either of two decomposed formulas were inferior to WMT.Conclusion WMT has a protective effect on VEC from injury due to hyperlipidemic serum,which may be one of its mechanisms in treating ASO.The protective effect of WMT on VEC from injury is stronger than that of the decomposed formula,which demonstrates the rationality of combination of WMT to some extent.
摘要:Objective To observe the regulation effect of Jiangzhitongmai Formula on the expression of heparan sulafae proteoglycan(HSPG) of liver basal membrane in mice with ApoE-knock-out arthrosclerosis.Methods The normal control group(group 1) included 12 C57/BL mice and other 36 mice of ApoE knock-out were randomly divided into the model group(group 2),Jiangzhitongmai Formula group(group 3) and Simvastitin control group(group 4).Group 1 and 2 were given orally normal saline and group 3 and 4 were given respectively Jiangzhitongmai Formula and Simvastitin orally in the dose higher than normal(/kg) by 10 times.After 90 days the serum content of blood lipid was determined by fully automatic biochemical analyzer,the expression of HSPG was tested by immunoblot method and the photodensity was analyzed by the software of Quantity One 6.0 in different groups.Results In group 3 and 4 the levels of total cholesterol(TC) and low-density lipoprotein-C(LDL-C) were obviously decreased with a great significant difference(P<0.001) compared with those in group 2.There was no significant difference between group 3 and 4.The expression of HSPG was much higher in group 3 and 4 than that in group 2 but much lower than that in group 1.Conclusion Jiangzhitongmai Formula has an effect of decreasing TC and LDL-C levels and regulating the expression of HSPG of liver basal membrane in mice with ApoE-knock-out arthrosclerosis induced by feeding high fat food.The up-regulative effect of Jiangzhitongmai Formula on the expression of HSPG is one of its mechanisms for preventing and treating arthrosclerosis.

摘要:Objective To observe the effects of Naoxintong Capsules on cerebral edema induced by focal lesion of cerebral ischemia/reperfusion,myeloperoxidase(MPO) activity and the expressions of intercellular adhesion molecule1(ICAM-1),vascular cell adhesion molecule-1(VCAM-1) and E-selectin in rats.Method The model of brain middle cerebral artery ischemia/reperfusion was established by the thread approach.The influence of Naoxintong Capsules on brain water content,MPO activity and the expressions of ICAM-1,VCAM-1 and E-selectin were observed.Result Naoxintong Capsules in the dose of 0.24 g/kg or 0.48 g/kg relieved cerebral edema,decreased MPO activity,reduced the expressions of ICAM-1,VCAM-1 and E-selectin.Conclusion Naoxintong Capsules has a protective effect on rat brain from cerebral edema induced by the injury of focal cerebral ischemia/reperfusion.The mechanism is related to that Naoxintong Capsules can decrease MPO activity and reduce the expressions of ICAM-1,VCAM-1 and E-selectin in rat brain.
摘要:Objective To understand the relationship between renal lesion and nuclear transcription factor-κB/IκBα(NF-κB/IκBα) expression in residual kidney of rat after nephrectomy by 5/6 and observe the influence of Wenpi Decoction on NF-κB/IκBα expression.Method The rat model of renal fibrosis was established by nephrectomy by 5/6.The NF-κB p65/IκBα expression in residual kidney was detected by using two-step stain of immunohistochemistry and their actions in renal lesion were analyzed according to some indexes,such as levels of Scr,GUN and urine protein of 24 hours and renal pathological lesion degree.The relationship between the renal protective effect of Wenpi Decoction and NF-κB/IκB was explained on the base of above analysis.Result In the model group the NF-κB p65 expression in renal tissue was higher than that in normal renal tissue and correlated to the increase of urine protein and Scr,while IκBα expression was lower than that in normal renal tissue.By comparing with the model group Wenpi Decoction can up-regulate IκBα expression and inhibit the over activation of NF-κB p65.Conclusion Wenpi Decoction can relieve the renal lesion induced by nephrectomy by 5/6,which may be related to it can inhibit the abnormal increase of NF-κB expression in renal tissue.
摘要:Objective To observe the effects of isopsoralen and ecdysterone on the proliferation of T47D cells and detect their influence on the expressions of T47D subtypes-estrogen receptor α(ERα) and estrogen receptor β(ERβ) cells in human breast cancer.Methods The influence of isopsoralen and ecdysterone(the active principles of Fructus Psoraleae,Radix Cyathulae) on the proliferation of ERα and ERβ cells and on the expression levels of ERα and ERβ mRNA by MTT assay and RT-PCR respectively.The changes of cell proliferation cycle were detected by flow cytometry.Results Isopsoralen and ecdysterone(in dose of 10-6 mol/L and 10-7 mol/L respectively) promoted obviously the proliferation of cultured T47D cells,increased the proliferation indexes with an estrogenic effect.The result of RT-PCR showed that isopsoralen(10-6 mol/L) and ecdysterone(10-7 mol/L) improved the expression of ERα mRNA and isopsoralen improved the expression of ERβ mRNA at the same time.Conclusion Isopsoralen and ecdysterone have the phytoestrogenic effects and can influence the mRNA expression level of ER subtype.

摘要:Objective To investigate the formation and actions of endogenetic toxins from the dynamic changes of inflammatory factors of local cerebral ischemia reperfusion.Methods All 63 rats were randomly divided into the sham-operation group and model group(ischemia reperfusion group).The thread approach was used to establish the model of cerebral ischemia reperfusion in middle cerebral artery(ischemia for 1.5 hours and reperfusion for 1,3,6,12,24 and 48 hours respectively).The immunohistochemistry was used to determine the expressions of tumor necrosis factor-α(TNF-α) and interleukin-1β(IL-1β) at above different time points and then the histopathological changes were observed.Results The result of immunohistochemistry showed that in the model group IL-1β expression began to increase 6 hours after the ischemia reperfusion and reached the peak on 12th hour,while TNF-α expression began to increase 3 hours after the ischemia reperfusion and reached the peak on 24th hour.The result of HE stain showed that in the model group the disorganization of structure layer of cerebral cortex was aggravated along with reperfusion time and reached the peak on 24th hour.Conclusion The expressions of IL1β and TNF-α are the important points of cascade reaction of cerebral ischemia.They are involved in the pathological lesion of cerebral ischemia and their dynamic changes and toxic effects reflect the formation and actions of endogenetic toxins.
摘要:Objective To detect the gene expressions of procollagen-lysine 1,2-oxoglutarate 5-dioxygenase 1(PLOD1) in different types of mucous membrane of tongue coating by using real time fluorescent quantitative PCR and analyze the relationship between the expression levels and tongue coating.Methods The chief cDNA were collected from natural fetus and five types of pathologic mucous membrane of tongue coating by RT-PCR.The gene expression levels and tends in different types of mucous membrane of tongue coating were detected and observed by the real time fluorescent quantitative PCR.Result The result of gene expression levels of PLOD1 in pathologic mucous membrane of tongue coating showed a descending tend as follows.White thin coating group>yellow-thin coating group>yellow-thick coating group>white thick coating group>no coating group>normal coating group.The multiple proportion of copy ratio of PLOD1 showed by 2.97,2.68,2.21 and 1.50 times respectively compared with the no coating group.Conclusion The changes of expression levels of PLOD1 in pathologic mucous membrane of tongue coating can influence the quality of tongue coating.

摘要:Objective To study the influence of tetramethylpyrazine on fibroblasts and conduction of TGF-β signal and its mechanism of anti-fibrosis.Methods The influence of tetramethylpyrazine on the growth and proliferation of fibroblasts was detected through the incorporation H3-thymidine into DNA taking NIH3T3 as the model and on the expression of collagen I was observed by immunoluciferase stain test,and on conduction of TGF-β/Smads signal was studied by using the cell line of Mv1Lu-(CAGA)12-Luc.Results Tetramethylpyrazine inhibited the expression of collagen I of fibroblast.Without pathological factor stimulation tetramethylpyrazine had no obvious influence on NIH3T3 proliferation and TGF-β conduction in the concentration range from 0.2 to 20 mg/L.Under the stimulation of TGF-β tetramethylpyrazine had an obvious inhibitory effect on NIH3T3 proliferation in the concentration of 20 mg/L(P<0.05).The influence of tetramethylpyrazine was not obvious in the concentration from 0.2 to 20 mg/L on the luciferase activity of normal cell responding to TGF-β,but under the stimulation of TGF-β(2 μg/L) the luciferase activity responding to TGF-β was inhibited obviously in different groups(P<0.05 or P<0.001).Conclusion Tetramethylpyrazine can inhibit the expression of collagen I of fibroblast,the proliferation of fibroblast.The inhibitory effect on conduction of TGF-β/Smads signal may be one of its mechanisms of anti-fibrosis effect.

摘要:Objective To study the characteristics of TCM syndromes of the model of ventricular restructure and the characteristics of ventricular restructure induced by partial coarctation of abdominal aorta in rats.Methods The rats were randomly divided into the normal group,sham-operation group and model group.The ventricular restructure model was established by partial coarctation of abdominal aorta.With the standard of syndrome differentiation for deficiency syndrome and blood stasis syndrome of China Society of Integration of Chinese and Western Medicine,the general condition,heart rate,time of exhausted swimming,electrocardiogram,ultrasonograph of rat in every week were observed after the operation,and the TCM syndromes were evaluated.Results The general condition of the model group manifested as deficiency syndrome and blood stasis syndrome.In the model group,compared with the sham-operation group after 4 weeks,the heart rate(P<0.05) and number of variation ST-section were increased(P<0.01),but time of exhausted swimming(P<0.01)is shortened markedly.Inter ventricular septum thickness(IVSTd,P<0.01) and left ventricular mass index(LVMI,P<0.01) increased significantly.After 8 weeks in the model group,compared with the sham-operation group,the number of variation ST-section,heart rate,time of exhausted swimming,IVSTd,LVPWTd and LVMI were the same as those after 4 weeks,but LVDd(P<0.05) increased markedly and EF(P<0.05) decreased markedly.Conclusion The rat with left ventricular restructure induced by partial coarctation abdominal aorta had the TCM syndrome characteristics of heart-qi deficiency and blood stasis.The syndrome of blood stasis existed in the whole pathological cause,while the syndrome of heart-qi deficiency emerged and aggravated along with the disease development.The model had typical pathological changes of left ventricular concentric hypertrophy in early stage.After 8 weeks the model had left ventricular synthetic hypertrophy with not only concentric hypertrophy but also centrifugal hypertrophy.

摘要:Objective To observe the protective effect of Biejiajian Pills on experimental rat kidney from renal interstitial fibrosis and discuss the mechanism.Method The model of renal obstruction was copied in rats by unilateral ureteral obstruction(UUO) and then the rats were divided into UUOV group(treated with Valsartan),UUOB group(treated with Biejiajian Pills),model group and sham-operation group.The protein and gene expressions and content of adrenomedullin(ADM) in obstructed kidney were observed and determined.Result In the model group and UUOV,UUOB groups the content of protein and expressions of mRNA and ADM protein decreased obviously than those in the sham-operation group,and those in UUOB and UUOV groups were higher than those in the model group(P<0.05) but there was no significant difference between UUOB and UUOV groups.Conclusion Biejiajian Pills can up-regulate ADM protein and mRNA expressions in rats with renal interstitial fibrosis to protect the kidney.
摘要:Objective To study the chemical principles in the flavonoid fraction of alcohol extract from Kushen Decoction.Method The chemical principles were isolated by different column chromatographies and their structures were elucidated with spectral methods.Result There were seventeen compounds isolated and identified as follows.Luteolin(Ⅰ),chrysin(Ⅱ),oroxylin A(Ⅲ),7-methoxyflavone(Ⅳ),baicalein(Ⅴ),baicalin(Ⅵ),wogonin(Ⅶ),wogonoside(Ⅷ),skullcapflavone Ⅱ(Ⅸ),5,8-dihydroxy6,7-dimethoxyflavone(Ⅹ),5,7-dihydroxy-6-methoxyflavanone(Ⅺ),xanthohumol(Ⅻ),kuraridin(ⅩⅢ),4,4’-dimethoxychalcone(ⅩⅣ),trifolirhizin(ⅩⅤ),formononetin(ⅩⅥ)and 2’,4-dihydroxy-4’,6’-dimethoxychalcone(ⅩⅦ).Among them the compounds Ⅰ~Ⅺ were identified from Scutellaria baicalensis and the compounds Ⅻ~ⅩⅦ identified from Sophora flavescens by HPLC.Conclusion All these compounds were isolated from Kushen Decoction for the first time.2’,4-dihydroxy-4’,6’-dimethoxychalcone was found firstly in Sophora flavescens.
摘要:Objective To discuss the survival quality of patient with cerebral infarction in sub-acute seizure phase,who were in different TCM syndrome types.Method The patients with cerebral infarction in sub-acute seizure phase targeted to the inclusion criteria were divided into the phlegm-heat group and non-phlegm-heat group through the syndrome differentiation of TCM.After routine treatment of Western medicine the survival quality of patients were evaluated with the Health Questionnaire(SF-36),a general adaptable scale developed by American Medical Outcomes Study(MOS).Result In different dimensions of SF-36 the score of body health dimension in the phlegm-heat group was obviously higher than that in the non-phlegm-heat group(P<0.05),while the score of mental health dimension in the phlegm-heat group was obviously lower than that in the non-phlegm-heat group(P<0.05).Conclusion The patients with cerebral infarction in sub-acute seizure phase in different TCM syndrome types have different contents of survival quality,which may be closely related to the characteristics of syndrome manifestations.
摘要:Objective To investigate the change law of non-linear dynamic characteristics of electroencephalogram(EEG) in the patients with the syndrome of stagnation of liver-qi.Methods The patients with the syndrome of stagnation of liver-qi(group 1,n=15) and normal persons(group 2,n=15) were paired according to the sex and age.The data of EEG were analyzed under three conditions(eye closed,eye opened and mental arithmetic with eye opened) by the parameter of correlation dimension(D2).Results The data of D2 in parts of brain zone in group 1 increased significantly than those in group 2.The data of D2 in the cortex in two groups all increased along with the enhancement of stimulation,but the difference in D2 between two groups was reduced gradually.Group 1 had some changes in cognitive function.Conclusion The non-linear dynamic characteristics of EEG reflects objectively and accurately the cortex changes in the patients with the syndrome of stagnation of liver-qi,and it can be used for the study on the cerebral mechanism of the syndrome of stagnation of liver-qi.

摘要:Objective To investigate the correlation between TCM syndrome manifestations and the degree of neurological parafunction of ischemia stroke.Method The patients with acute ischemia stroke(n=414) were selected and their scores of six syndrome factors(wind,fire,phlegm,stasis,qi-deficiency and hyperactivity of yang due to yin-deficiency) were recorded respectively according to the Diagnostic Standard of Syndrome Differentiation of Stroke and the National Institutes of Health Stroke Scale(NIHSS) on 1st,2nd,3rd,7th and 14th day after their hospitalization.The correlation between syndrome factors and neurological parafunction, between syndrome factors at different time points were studied by using the methods of linear correlation and multiple linear regressions.Result The scores of NIHSS were correlated to several syndrome factors at different time points.The syndrome factors explained 60% of changes in NIHSS.Conclusion There is a correlation between TCM syndrome manifestations and neurological parafunction or between syndrome factors in acute ischemia stroke.The weight of correlation degrees and syndrome factors are dynamic in the prediction of scores of NIHSS.
摘要:Objective To discuss the law of colour changes of sublingual collaterals in patients with lung cancer to provide an objective base for clinical diagnosis,and to verify the important meaning of TCM tongue inspection in disease diagnosis.Method The images,values of R,G,B and H,S,I of colour of sublingual collaterals were collected and analyzed from 423 patients with lung cancer.Result In the lung cancer group the values of R, G,B were obviously lower than those of other groups(P<0.05 or P<0.01),H value was obviously higher than that of other groups(P<0.05 or P<0.01),S value was obviously lower than that of the tuberculosis group and healthy control group(P<0.01),and I value was obviously lower than that of other groups(P<0.01).Conclusion The values of R,G,B,S and I of sublingual collaterals colour are lower and H value is higher in the patients with lung cancer.The observation and analysis on the colour changes of sublingual collaterals is significant for the diagnosis of lung cancer in clinic.
摘要:Objective To find out the distributive characteristics and main trend of TCM syndromes of epilepsy,and provide a theoretical basis for the clinical diagnosis and treatment of epilepsy.Method Through choosing clinical records of 202 epilepsy patients the common symptoms and main syndromes of epilepsy were summed up,and the diagnostic standard of TCM syndromes of epilepsy was set up.Result After the syndrome differentiation 202 patients were classified into 5 syndrome types.There were 65 patients with the syndrome of wind-phlegm stagnation(32.2%),59 with the syndrome of phlegm-fire up-disturbing(29.2%),31 with the syndrome of superabundance of phlegm due to spleen-deficiency(15.3%),29 with the syndrome of deficiency of both heart and spleen(14.4%) and 18 with the syndrome of deficiency of liver-yin and kidney-yin(8.9%).The most common TCM symptoms and manifestations of tongue and pulse were included in the syndrome of wind-phlegm stagnation or phlegm-fire up-disturbing.Conclusion The common TCM syndromes of epilepsy are the syndrome of wind-phlegm stagnation and phlegm-fire up-disturbing in clinic.
