      摘要:The constitution is an important manifestation of the human life.Hippocrates,an ancient Greek physician,and Huangdi Neijing in the Qin and Han Dynasty of ancient China had paid attention to the study of the human constitution,but the TCM constitutional study has not formed a special academic system for more than 2000 years.Here three key scientific problems are raised on the base of researches and practices for many years including "theory on constitution being separable","theory on relationship between constitution and disease" and "theory on constitution being recuperable",and they form a general framework of constitutional studies.In human life process there are individual differences among people groups influenced by many factors,including time,space,inheritance,diet structure and psychology,which are the basis of the "theory on constitution being separable".There are internal relationships between different constitutional types and diseases and they affect the types and changes of TCM syndrome manifestations,which are the basis of the "theory on relationship between constitution and disease".To regulate the whole organism of the human body based on the biased status and dynamic changeability of the constitution for health recovering,which is the basis of the "theory on constitution being recuperable".

      摘要:There are four basic problems that are under concerned nowadays:the essence of substances,the origin of the universe,the hypostasis of lives and the emergence of intelligence.The cognitive science,thinking science and artificial intelligence science are all related intimately to the "emergency of intelligence",one of problems mentioned above.Being under the circumstance of development of discipline cross,the cognitive science focuses on the researches of cognitive psychology,cerebral science,neurophysiology,artificial intelligence and pattern recognition,which shows a merging trend of multi-discipline and multi-field.The "cognitive complexity" itself becomes the characteristics of contemporary science.We introduce here the cognitive science and system science,and discuss the possibility to explain the complex giant system of the human body with TCM theory focusing on the crossing of sciences mentioned above and TCM theory,and to lay a foundation for describing the rationality and scientific nature of TCM theory.

      摘要:Objective To study the changes of brain auditory working memory of Chinese character phrases in patients with liver-qi stagnation syndrome by applying functional magnetic resonance imaging(fMRI),find the brain functional region of the liver-qi stagnation syndrome and the changes of cognitive function,and to reveal the cerebral mechanism of the syndrome.Methods The patients with liver-qi stagnation syndrome(n=16,male 5 and female 11)from 25 to 50 year-old and healthy postulants(n=16)according to the age and sex matching were chosen and scanned by magnetic resonance imaging.The task of brain auditory working memory function of Chinese character phrases was carried out in 1-bck of block-design.The scan was started from resting and the sequence was resting-baseline-resting-stimulating circularly by using magnetic resonance machine of Plilips1.5T.The data was processed with SPM software based on MATLAB platform.Results The number and area of activated brain regions of working memory in the healthy group were much more than those in the patient group.Besides the left inferior frontal gyrus(BA44/47,Broca motor speech region)of activated brain regions in the healthy group other brain regions and the right parietal lobe were inhibited in the patient group when the working memory reduced.Conclusion The brain regions were positioned related to the changes of working memory in the patients with liver-qi stagnation syndrome.The liver-qi stagnation syndrome can induce cognitive function changing,which provides the visual objectified evidence for TCM syndrome types.

      摘要:Objective To observe the expression changes of neurocytes and neurotrophic factors in rats with cerebral ischemia after of bone mesenchymal stem cells(BMSC)transplantation,and investigate the mechanisms of combination of rhubarb aglycone and BMSC transplantation based on the influence of rhubarb aglycone on these changes.Methods BMSC were purified and increased by using the methods of adherence and screening in vitro.The model of middle cerebral artery occlusion(MCAO)was established with suture method.After 24 hours BMSC were transplanted from internal carotid artery to the brain,and then rhubarb aglycone was used by intragastric administration.The expressions of the specific signs of neurocytes(NSE and GFAP),as well as neurotrophic factors(NGF and GDNF),were determined by using immunohistochemical method on 7th(earlier period),14th(metaphase)and 28th(anaphase)day respectively after BMSC transplantation.Results The expressions of NSE and GFAP were significant and expressions of NGF and GDNF showed weak positive in the sham-operation group.The expressions of GFAP,NGF and GDNF all increased on 7th day,while those of GFAP and NGF decreased on 14th and 28th day,and GDNF expression reduced on 14th day and increased on 28th day in the model group.In rhubarb aglycone group,compared with the model group,NSE expression increased and GFAP expression decreased on 7th day,and NGF expression increased on 28th day,while GDNF expression decreased on 7th and 14th day.In he transplantation group,compared with the model group,the expressions of GFAP,NGF and GDNF increased all on 28th day.In the combined group NSE expression increased at all time points,GFAP expression decreased on 7th day and increased on 14th day,and NGF expression,compared with the rhubarb aglylcone group and transplantation group,increased on 14th and 28th day.Conclusions Neuron showed a decreased response progressively following the prolongation of the lesion of cerebral ischemia reperfusion.The neurogliocytes showed the expression increased at first and then decreased.The responsiveness of neurotrophic factors showed increasing in earlier period,and then NGF expression decreased progressively.GDNF showed decrease at first and then increase.The transplantation BMSC in the brain showed significant effects in metaphase and anaphase on increasing neurotrophic factors and neurocytes responses.Rhubarb aglycone can advance the time of protecting neurocytes after BMSC transplantation.The mechanism may be related to that it can up-regulate the expressions of NGF and GDNF in earlier phase and increase NGF expression in metaphase and anaphase.

      摘要:Objective To investigate the influence of Biejiajian Pills(BP)on matrix metalloproteinase system of human mesangial cells(HMC).Methods HMC were cultured by applying cell cultural technique.The serum pharmacological method was used in the preparation of BP serum.The expressions of matrix metalloproteinase-9(MMP-9)and tissue inhibitor of metalloproteinase-1(TIMP-1)were detected by enzyme-linked immunosorbent assay(ELISA)taken lipopolysaccharide(LPS)as stimulating factor.Results The contents of MMP-9 in every group were not different(P>0.05).The content of TIMP-1 in LPS group were obviously higher than that in the blank group(P<0.01),that in BP group was obviously lower than that in the blank group(P<0.05).The expression of TIMP-1 in the LPS combined BP group or in the valsartan combined BP group was obviously lower than that in the LPS group(P<0.01).Conclusion LPS has an improving effect on the expression of TIMP-1,but less effect on the expression of MMP-9.The serum of BP or serum of valsartan can inhibit the expression of TIMP-1 in the condition of normal or LPS stimulation,which may be the mechanism of BP in the delay of glomerular sclerosis.

      摘要:Objective To review the phytoestrogenic activity of five kinds of Chinese herbal medicines(CHM)including Fructus Psoraleae and possible mechanism.Methods All 96 normal Wistar female rats(just weaning)were randomly divided into 12 groups(solvent control group,diethylstilbestrol group,5 groups of CHM and 5 groups of CHM combined with diethylstilbestrol).After 4 days all rats were killed for taking blood to extract the serum samples.The rat uterus coefficients and the gain in weight were calculated.The influence of the medical serum on cell proliferation was detected by using MTT assay in estrogen receptor(ER)positive MCF7 cells,and the effect of it on cell cycle was tested by using flow cytometry.Results ①Fructus Psoraleae and Radix Cyathulae administrated intragastrically increased significantly the rat uterus coefficient(P<0.05),while Fructus Psoraleae,Radix Cyathulae and Flos Carthami administrated intragastrically inhibited significantly the increase of rat uterus coefficient induced by diethylstilbestrol(P<0.05).Radix Cyathulae administrated intragastrically raised rat weight significantly(P<0.05).②In the groups of CHM the medical serum containing Fructus Psoraleae,Radix Cyathulae,Flos Carthami or Radix Salviae Miltiorrhizae promoted the cell proliferation of MCF7 cells(P<0.01),but promotion effect could be reduced by ERβ sagonist DPN.In the antagonism test the promotion effect of medical serum in the groups of CHM decreased significantly than that in the diethylstilbestrol group,and DPN could improve this decreasing effect furthermore(P<0.05 or P<0.01).③The medical serum containing Fructus Psoraleae,Radix Cyathulae,Radix Salviae Miltiorrhizae or Semen Cuscutae increased the indexes of cell proliferation in the groups of CHM.The medical serum had no obvious antagonism effect on the increase of cell proliferation indexes induced by diethylstilbestrol in the groups of CHM combined with diethylstilbestrol.Conclusion Fructus Psoraleae,Radix Cyathulae,Flos Carthami,Radix Salviae Miltiorrhizae or Semen Cuscutae have phytoestrogen-like effects,but they are different in intensity and action pathway.

      摘要:Objective To observe the influence of curcumin on proliferation of human renal epithelia(HK-2)induced by transforming growth factors-β1(TGF-β1),and discuss the mechanism of preventing and treating renal fibrosis by curcumin.Methods HK-2 cells were cultured with culture medium containing 10% of fetal bovine serum FBS,DMEM-F12(1∶1).The samples were divided into 5 groups,including the blank control group,TGF-β1 group(10 μg/L of TGF-β1),intervention group 1(10 μg/L of TGF-β1+1 μmoL/L of curcumin),intervention group 2(10 μg/L of TGF-β1+5 μmoL/L of curcumin),and intervention group 3(10 μg/L of TGF-β1+10 μmoL/L of curcumin).The morphological changes of cells in every group were observed with inverted microscope and the influence of curcumin on cell proliferation by using MTT assay.Results The proliferation of HK-2 cells was induced significantly by 10 μg/L of TGF-β1 with a significant difference compared with the blank control group(P<0.05).This effect increased with treatment time passing,but when TGF-β1 and curcumin were combined and effected on the cells this effect was inhibited obviously(P<0.05).Conclusion Curcumin can maintain the morphological stability of HK-2 cells,prevent cells from fibrosis,and inhibit cell proliferation induced by TGF-β1 to some extent.It may be the mechanism of preventing and treating renal fibrosis by curcumin.

      摘要:Objective To study quantitatively the indexes of microscopic tomography of tongue manifestations and tongue coating and texture in the rats with spleen-stomach damp-heat syndrome in vivo by applying optical coherence tomography(OCT),and to investigate the change laws of the tongue manifestations of spleen-stomach damp-heat syndrome in microscopic detection and value of the application of optical coherence tomography in tongue diagnosis.Methods The rat model of spleen-stomach damp-heat syndrome was established by using combined factors,and rat tongue surface was detected in vivo by microscopic tomography and the relevant parameters of tongue coating were processed and analyzed quantitatively on Lab VIEW software platform by applying self-made OCT equipment.Results The tongue coating layer and connection layer between tongue coating and body were observed clearly by microscopic tomography,which was similar to the result of microstructure of rat tongue by histological detection.The tongue coating was thicker significantly in the rats with spleen-stomach damp-heat syndrome than that in the normal rats(P<0.01),while the fluid of tongue body was less significantly(P<0.01).Conclusion The tongue manifestations of the model of spleen-stomach damp-heat syndrome have characteristic changes in microscopic detection.OCT technology can be used in the study of rat tongue manifestations quantitatively.

      摘要:Objective To observe the influences of Radix Astragali on the contractile and electric activities of small intestine smooth muscle in rabbits and discuss the mechanism.Methods The influences of Radix Astragali on the spontaneous contraction and electric activities of small intestine in healthy rabbits(male or female)after the isothermal perfusion of small intestine in vitro.The perfusate was added respectively propranolol,glibenclamide and nitro-L-arginine methyl ester(L-NAME)before adding Radix Astragali,the action mechanism of Radix Astragali was studied and its influences on two kinds of contractile response induced by acetylcholine were observed.Results Radix Astragali reduced the amplitude,frequency and area under the curve of contractile and electric activities of small intestine smooth muscle in rabbits in a does-depended manner.IC50 of the contractive amplitude was 15.24 g/L by Logit analysis.Propranolol and L-NAME could partly block the inhibitive effect of Radix Astragali on the contraction of small intestine smooth muscle in rabbits.Comparing with the control group the influences of propranolol and L-NAME had significant difference in the contractile amplitude and area under the curve after variance analysis(P<0.05),while glibenclamide had no obvious blocking effect on the inhibition of contractile activity of small intestine smooth muscle in rabbits.Radix Astragali promoted significantly the intracellular calcium-depended contractile amplitude induced by acetylcholine,but inhibited significantly the extracellular calcium-depended contractile amplitude.Conclusion Radix Astragali inhibits significantly the contractile and electric activities of small intestine smooth muscle in rabbits.The mechanism may be related to that it can promote β-receptor,increase NO concentration and inhibit extracellular Ca2+ inflowing into cells via cell membrane.

      摘要:Objective To observe the influence of Jiaweigualou Powder(JP)on gastrointestinal hormones in rats with portal hypertension.Method The radioimmunity method was used to detect the serum contents of gastrointestinal hormones(motilin,gastrin and glucagon)in rats of the normal group,model group,propranolol group and JP group.Result The serum contents of motilin,gastrin and glucagon increased significantly(P<0.01)in the model group than those in the normal group.The content of motilin did not change(P>0.05),but the contents of gastrin and glucagon decreased significantly(P<0.05)in the propranolol group than those in the model group.In JP groups with different doses the serum contents of motilin,gastrin and glucagon decreased significantly than those in the model group(P<0.01 or P<0.05),and the contents of motilin and glucagon decreased significantly than those in the propranolol group(P<0.05).There was no statistical difference among JP groups with different doses.Conclusion JP can decrease effectively the serum contents of motilin,gastrin and glucagon,and has a better curative effect than propranolol.The mechanism may be related to that it can antagonize liver fibrosis,prevent hepatocirrhosis,reduce portal pressure and improve liver function.

      摘要:Objective To discuss the anti-inflammatory and convergent effects of Shuangfujin on the model of acute eczematous dermatitis.Methods The group of normal saline,group of boric acid solution(3%),group of Pifukang Lotion(1∶5)and group of Shuangfujin were established respectively.Kunming female mice(n=40)were chosen and randomly divided into 4 groups.The anti-inflammatory effects in different groups were analyzed according to the swell degree and thickness difference of mouse ear induced by xylene.Wista male rats(n=40)were chosen and randomly divided into 4 groups.The convergent effects in different groups were analyzed according to the splash area,skin thickness on rat back and optical density when skin dissolving in acetone solution induced by histamine phosphate after Evans blue modeling.Results The group of Shuangfujin had the highest anti-inflammatory effect(P<0.01).The group of Pifukang Lotion had higher anti-inflammatory effect than the group of boric acid solution(P<0.05).The group of normal saline had the lowest anti-inflammatory effect(P<0.01).The group of Shuangfujin and group of Pifukang Lotion had similar convergent effect(P>0.05),which was higher than that of the group of boric acid solution(P<0.05).The group of normal saline had the lowest convergent effect(P<0.01).Conclusion Shuangfujin has good effects of anti-inflammation and convergence in the model of acute eczematous dermatitis.

      摘要:Objective To establish a HPLC method for the identification of Semen Sinapis and the varying decoction pieces of Semen Sinapis Albae,and provide a scientific base for revealing the processing principle of Semen Sinapis Albae.Method The HPLC method was performed on a column of kromasil C18 with the temperature of 35℃,using acetonitrile-0.1 % of phosphoric acid solution as the mobile phase in gradient elution.The detection wavelength was 254 nm and the flow rate was 1.0 mL/min.Result The HPLC maps were different significantly among different varieties of Semen Sinapis and varying decoction pieces of Semen Sinapis Albae.Semen Sinapis Albae had two bigger chromatogram peaks at about 6 and 9.3 minute but Semen Sinapis Albae.P.E.had not.There was obvious different in HPLC maps between raw Semen Sinapis Albae and stir-baking Semen Sinapis Albae when water was used as extraction solvent.The chromatogram peak disappeared at 6 minute and a quite big peak was detected at 10 minute in raw Semen Sinapis Albae,while the peak was not detected in stir-baking Semen Sinapis Albae.Conclusion The method established here can be used for the HPLC identification of Semen Sinapis and varying decoction pieces of Semen Sinapis Albae,which supplies a reliable clue for the quality evaluation.

      摘要:Objective To study on the synthesis and transformation of sinomenine microbial hydroxylation for the first time,and obtain the best technique on the basis of the optimization of transformation conditions.Methods Ten microbe strains with hydroxylation abilities were used in the transformation of sinomenine.The methods of TLC and HPLC-MSn were applied to identify the original sinomenine and its metabolites,and to optimize the microbe strains with high activities and the best transformation condition for the hydroxylation of sinomenine.Results After the screening there were three microbe strains showed the hydroxylation abilities among ten,and Cunninghamella M2 was the best one after secondary screening.When the rotation speed was 180 r/min,optimum culture time was 6 days,initial pH value was 6.5,culture temperature was 28℃,and highest concentration of feeding substrate was below 200 mg/L,the hydroxylation rate of sinomenine would be over 95%.Conclusion Cunninghamella M2 can transform and modify sinomenine into hydroxy-sinomenine,which has strong specificity and high transformation efficiency.

      摘要:Objective To establish a method for quantitative determination of baicalin in Qingnaojiangya Capsules by HPLC.Methods The ultrasonic extracting method was applied in sample pre-treatment.The HPLC procedure was performed on the chromatographic column of Agilent zorbax Eclipse XDB-C18(4.6 mm×250 mm,5 μm),the mobile phase was methanol-0.2% of phosphoric acid water solution(42∶58).The detection wavelength was 278 nm and flow velocity was 0.8 mL/min.Results Baicalin showed a good linear relationship at range of 0.366~1.832 μg with correlation coefficient of 0.999 1.The average recovery of baicalin samples was 99.84%(RSD=0.969%).Conclusion The method was sensitive,simple and reliable,which was can be used for the quantitative determination of baicalin in Qingnaojiangya Capsules.

      摘要:Objective To prepare the colon-positioning-timing tablets of berberine from Chinese compound formula by using film coating technique.Method The colon-positioning-timing tablets were prepared with enteric-coated acrylic resin Eudragit L100 and Eudragit S100 as pH-sensitive layer material,and aqueous ethylcellulose dispersion(Surelease)as time-lag layer material by using film coating technique.The in vitro release percent was taken as the test indexes so the characteristics of the colon-positioning-timing tablets would be in the agreement with design requirement basically.Result The test of in vitro release percent showed that berberine released 3 hours after reaching the small intestines when the pH-sensitive layer was weighted by 4% and that of time-lag layer by 6%.The release was not influenced by gastric emptying.Conclusion The colon-positioning-timing tablets can achieve expected effect of positioning release in the colon through regulating the coating thickness of the time-lag layer and enteric layer.

      摘要:Objective To study the effect of Ginkgo biloba extract(GBE)on coronary flow and endothelial function in aged patients with coronary heart disease(CHD).Method The patients with CHD(n=50)were randomly divided into the treatment group treated with GBE injection and control group.Before and after the intravenous injection,the blood flow of left anterior descending coronary artery(LAD)and changes of flow-mediated endothelium-dependent vasodilation(FMD)of brachial artery in two groups were detected by using vascular ultrasound.Result In the GBE group diastolic peak velocity(DPV),systolic peak velocity(SPV)and diastolic time velocity integral(DTVI)of LAD were improved significantly by GBE(P<0.01)and FMD of brachial artery was increased(P<0.01).In the control group LAD flow and FMD of brachial artery did not change(P>0.05).The Pearson analysis showed that the improvement of DPV,SPV and DTVI of LAD was correlated to the promotion of FMD of brachial artery(P<0.01).Conclusion GBE injection can improve LAD flow in aged patients with CHD,which may be related to the promotion of FMD of brachial artery.

      摘要:Objective To observe the effect of Jianweiyuyang Pills on the expression changes of platelet activating factors(PAF)of gastric mucosa in the patients with chronic helicobacter-pylori-related gastritis,and study the potential mechanism.Methods The patients with chronic helicobacter-pylori-related gastritis(n=80,finally diagnosed by gastroscope)were chosen,and then randomly divided into two groups,including the treatment group(n=42)treated with Jianweiyuyang Pills and the control group(n=38)treated with three combined Western medicates.The pykno-urokinase in the gastric mucosa was detected by using gastroscopic biopsy,the helicobacter pylori was tested by Warthin-Starry stain method,and the expression changes of PAF and content of aninohexose and phosphatide were determined at the same time.On the forth week the patients were given reexamination with gastroscopy.Results The total effective rate of the treatment group was similar to that of the control group.The contents of aninohexose and phosphatide were significantly higher after the treatment than before(P<0.01).In the treatment group the contents of aninohexose and phosphatide increased with a significant difference compared with those in the control group(P<0.01).The expression of PAF was down-regulated by Jianweiyuyang Pills in the treatment group but had no changes in the control group.Conclusion The mechanism of Jianweiyuyang Pills for anti-gastritis may be related to killing helicobacter pylori,down-regulating PAF expression,influencing the contents of aninohexose and phosphatide,effecting on the hydrophobicity of gastric mucosa,strengthening the stability of gastric mucus gel layer,and preventing gastritis from recurrence.

      摘要:Objective To observe the curative effect of the manipulation of fixing bones and relaxing tendons used in the early rehabilitation of patellar closed fracture.Method All 60 cases of patellar closed fracture for half month were divided into the control group(n=30)with the contraction-relaxing activities of quadriceps femoris under the splint fixation,and the treatment group(n=30)treated with the manipulation of fixing bones and relaxing tendons to fix effectively the fracture ends and control the active and passive activities of the knee.Result In the treatment group the average healing time was 4.33 weeks,which was shorter significantly than that in the control group(P<0.01).The range of knee motion was improved significantly(P<0.01 after one month,P<0.05 after 2 months),and after 3 months the excellent rate of knee functional evaluation score was higher than that in the control group(P<0.05).Conclusion The manipulation of fixing bones and relaxing tendons has a good curative effect in the early rehabilitation of patellar fracture and recovery of the knee joint.
